目的:构建重组人白细胞介素7(rhIL-7)的原核表达载体,在大肠杆菌中表达并鉴定.方法:以RT-PCR方法扩增编码IL-7的cDNA片段,插入至pET-3d载体中,构建rhIL-7的原核表达载体,阳性克隆经测序鉴定,转化至大肠杆菌BL21(DE3),优化rhIL-7表达条件,利用免疫印迹鉴定重组蛋白.结果:重组载体经DNA测序显示包含正确的rhIL-7编码序列,重组载体转入大肠杆菌后,经IPTG诱导后外源蛋白表达,相对分子质量为17 400,与IL-7理论分子质量一致;免疫印迹显示,外源蛋白可以被人IL-7特异性抗体识别,表明在原核表达的外源蛋白是rhIL-7.rhIL-7主要表达于包涵体中,包涵体表达优化条件为:温度为37 ℃、IPTG浓度为0.4 mmol/L、诱导时间为4 h.结论:成功的构建了rhIL-7的原核表达载体.  相似文献

构建IL-18和IL-2成熟区的融合基因IL18-IL2,并实现其在大肠埃氏菌中的大量表达.抽提经PHA刺激增殖的人淋巴细胞总RNA,RT-PCR法获得IL-18和IL-2基因的成熟区序列,并通过一段Linker相连得到融合基因IL18-IL2;将IL18-IL2克隆至原核表达载体pBV220,并转化至大肠埃氏菌BL21(DE3);阳性克隆经42℃温度诱导表达后用SDS-PAGE和Western blot分析重组蛋白的表达情况及正确性.经SDS-PAGE分析,融合基因IL18-IL2在宿主菌中能够表达,其相对表观分子质量约为34.5 kD;Western blot进一步证明重组蛋白正确.实验结果表明,已成功构建了表达人IL18-IL2的菌株,为进一步研究融合蛋白IL18-IL2体内外抗肿瘤活性提供了基础.  相似文献

脱水蛋白DHN1表达载体pBV221-dhn1的构建及基因表达   总被引:1,自引:1,他引:0  
以克隆载体 pTZ 19R dhn1(ZM )为基础 ,构建脱水蛋白DHN1表达载体 pBV2 2 1 dhn1.采用PCR技术从克隆载体 pTZ 19R dhn1(ZM )上扩增dhn1片段 ,并引入NcoⅠ /BamHⅠ酶切位点 ,然后与 pBV2 2 1原核表达载体连接 ,得到 pBV2 2 1 dhn1表达载体 .阳性克隆经PCR和NcoⅠ /BamHⅠ酶切检测都得到 516bp的dhn1片段 ,且序列正确 .表达载体pBV2 2 1 dhn1转化宿主菌后能够表达产生相对分子质量为 2 2× 10 3的DHN1,该蛋白具有高温可溶性 .以上结果表明 ,本实验得到了高效表达的脱水蛋白DHN1表达载体 pBV2 2 1 dhn1.  相似文献

为研究重组人截短型白细胞介素6(rhIL-6)工程菌高效表达的影响因素及纯化方法的优化.观察不同培养温度对重组人截短型IL-6工程菌生长密度和IL-6表达的影响;复性蛋白终浓度对复性效率的影响;细菌诱导培养后,经破菌-洗包-溶包初步纯化后,再经柱层析纯化IL-6.其结果为30℃培养后42℃诱导培养5h的IL-6表达效率最高,达32.8%;复性蛋白终浓度在0.5 g/L以下时,蛋白复性效率最佳,比活性为4.42×108 μmol/min*mg-1;进一步柱层析后,IL-6的纯度达96.5%,得率可达46.5%.最后得出培养和纯化工艺简便易行,可获得高纯度、高比活性的截短型rhIL-6的结论.  相似文献

研究目的:克隆表达人肠激酶轻链编码基因,以期应用于融合蛋白的切割与纯化.方法:从人的全基因组中扩增出编码人肠激酶轻链的5个外显子基因片段,经过酶切、连接后得到正确编码的人肠激酶轻链完整基因序列;随后,将目的基因片段插入原核表达载体pBV220中,构建成融合型表达载体pBV—EKL,转化大肠杆菌BL21(DE3),调节温度至42℃诱导重组蛋白表达;通过镍亲和层析纯化得到重组蛋白.结果:所表达的重组蛋白经SDS—PAGE分析,相对表观分子质量约为31kDa,表达量占菌体总可溶蛋白量的46%;通过镍亲和层析纯化得到的重组蛋白经复性后显示出具有催化切割活性.结论:成功构建了能大量表达重组人肠激酶轻链的菌株,为进一步进行重组人肠激酶活性的研究及其在生产和医学方面的应用研究奠定了基础.  相似文献

人神经珠蛋白(hNGB)是新发现的人类脑特异的携氧蛋白.为对其结构和功能进行深入的研究,首先,利用分子生物学实验辅助设计软件BioSun1.0分析了hNGB基因与pBV221载体连接处的二级结构和hNGB基因的局部密码子偏爱性,并依据原核系统外源基因高效表达数学模型对hNGB基因进行了高效表达设计;其次,构建了hNGB基因编码区原核表达载体pBV221-hNGB;最后通过转化大肠杆菌JM109,热激诱导,获得了hNGB基因的高效表达.表达水平约为12%,从而为进一步研究hNGB基因的结构和功能奠定了重要基础.  相似文献

为构建HCV截短型ns5bΔ21基因的原核表达质粒;在大肠杆菌中表达NS5B蛋白并纯化,制备其抗体。运用软件设计引物,以BB7复制子为模板PCR扩增ns5bΔ21基因;克隆入原核表达载体pRSETA。将重组表达质粒pRSETA-ns5bΔ21转化BL21大肠杆菌,并诱导表达、可溶性分析及纯化;用纯化的NS5B蛋白免疫小鼠制备多克隆抗体。结果酶切鉴定和序列测定表明成功的构建了重组表达质粒pRSETA-ns5bΔ21;其经IPTG诱导后行SDS-PAGE可见新生表达条带,Western-blot证实了其特异性和抗原性良好;利用镍离子亲和层析和电洗脱方法获得了纯化NS5B蛋白;用纯化蛋白免疫小鼠制备出了多价抗血清。说明表达和纯化的截短型HCV NS5B RNA聚合酶可用于下一步筛选单链抗体。  相似文献

将编码木薯醇腈酶(HNL)cDNA克隆到原核表达载体pBV220,构建了大肠杆菌(Escherichia coli)中表达质粒pBV220-HNL.含重组质粒pBV220-HNL的大肠杆菌D145α菌株经温度诱导后,其表达产物以包涵体形式存在.SDS-PAGE分析及酶活测定的结果表明木薯HNL cDNA在大肠杆菌中成功表达.  相似文献

 将甲肝病毒vp1编码基因(aa 24~171)和戊型肝炎病毒ORF2基因(aa 431~615)通过一段编码柔性甘氨酸链(Gly4 Ser Gly4)的基因片断连接,获得编码HAV/HEV融合蛋白基因(AEAg342).将该融合基因插入质粒pBV220,构建表达质粒pBV220-AEAg342.将pBV220-AEAg342转化入大肠杆菌BL21中进行表达,表达量约占菌体总量的20%,经离子交换层析纯化,目的蛋白纯度达90%以上.Western blot证实该蛋白能与甲肝及戊肝患者阳性血清发生特异性反映,为进一步开发双价疫苗和联合诊断试剂奠定了一定基础.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
