<正> T细胞抗原受体(TCR)是由可变的α和β链组成的异二聚体,α和β链与稳定的单链复合物CD3非共价结合,TCRα和β链的基因译成的密码是经过再排列的,并在个体发育时表达。在胸腺微环境内,TCR遗传正常,最近证实在先天性无胸腺裸鼠,也发现TCR表达。 TCR的功能是胸腺内可变的α和β链选择定型的。在无胸腺鼠只有少数V_β链被利用。这种限制的异原性是DNAβ链重排列的限制型和对不同V_β亚区的单克隆抗体可变染色型所证明。此外,无胸腺鼠的T细胞对各种抗原的反应功能分析表明,与缺乏无性  相似文献

目的:制备负载SIV抗原肽的中国恒河猴Mamu-B*1703可溶性单体及其四聚体.方法:以含Mama-B*1703重链cDNA序列的pMD19-T克隆为模板,通过PCR的方法克隆Mama-B*1703重链基因,进而构建羧基端融合生物索化酶BirA底物肽(BSP)的Mamu-B*1703重链胞外域融合蛋白的表达载体,并在大肠杆菌中获得表达.β微球蛋白、SIV抗原肽共存时,通过稀释法复性可溶性Mamu-B*1703单体,经生物素化并纯化后与荧光素标记的链亲和素按4:1的比例混合形成四聚体.结果:ELISA检测显示获得具有正确构象的负载SIV抗原肽的Mamu-B-1703四聚体.结论:印度恒河猴Mamu.B}1701特异性抗原肽IW9,与中国恒河猴的Mamu-B*1703相结合形成可溶性Mamu-B*1703/IW9单体和四聚体.  相似文献

通过酸溶、胃蛋白酶酶解和氯化钠盐析等方法,从鲤鱼肌肉中纯化得到了V型胶原蛋白.SDS-PAGE电泳图谱分析发现,其至少由2条α链(α_1和α_2)组成,其中α1分子质量约为135 ku,α_2分子质量约为120 ku.通过圆二色光谱仪分析其二级结构,发现其具有胶原蛋白三股螺旋结构的典型特征,其热变性温度为30.5℃.将V型胶原蛋白作为抗原免疫新西兰大白兔,成功制备了多克隆抗体.采用免疫印迹法对该多克隆抗体的特异性进行分析,证明其仅与鱼类V型胶原蛋白产生反应,特异性良好.  相似文献

目的分析比格犬雌激素受体ERβ以及剪切异构体ERβ_(419)、ERβ_(431)的转录活性。方法将细胞分为6组,空白对照组(正常含有培养液的293T细胞),LV5-ERβ组(稳定表达ERβ的293T细胞),LV5-NC组(感染慢病毒LV5空载体的293T细胞),Lenti-OE-Flag-ERβ_(431)组(稳定表达ERβ_(431)的293T细胞),Lenti-OE-Flag-ERβ_(419)组(稳定表达ERβ_(419)的293T细胞),Lenti-OE-Flag-ERβ_(419)-NC组(感染慢病毒Lenti-OE-Flag空载体的293T细胞)。其中每组组内又分为2组,每组细胞瞬时转染构建的重组质粒ERE-LUC和RLUC,每组中的其中一组加入终浓度为10μmol/L的E_2,而另一组内加入等量无水乙醇。分别检测每组加入刺激(E_2和无水乙醇)前后的荧光素酶活性。结果成功地将雌激素反应元件ERE克隆到荧光素酶报告载体pGL3-Vector启动子上;设计引物扩增出的hRluc-neo基因成功克隆到pGL3-Basic载体;加入雌激素后,比格犬ERβ转录活性明显升高(P0.01),而ERβ_(419)和ERβ_(431)转录活性在雌激素加入前后变化不大(P0.05)。结论成功建立了比格犬ERβ的转录激活系统,而ERβ_(419)和ERβ_(431)的LBD区域都有部分的缺失,使得其不能与雌激素结合,继而转录活性无法被激活。  相似文献

人绒毛膜促性腺激素(βHCG)由α、β两条链组成,β链决定其生理功能。编码βHCG的基因及假基因已完成定位。张锡元等报导,在低等动物基因组中存在βHCG基团同类物,并报道了该同类物在鳙鱼染色体上的定位。张锡元、嵇庆等报告了牛蛙的βHCG基因同类物染色体定位结果。研究低等脊椎动物基因组中βHCG基因同类物染色体定位,对于探索βHCG基因从鱼类到人类的进化历程以及分析低等脊椎动物基因进化和染色体演化均有明显的意义。  相似文献

目的:分析五类免疫球蛋白重链可变区基因的特征.方法:收集IMGT/LIGM-DB数据库中人类五类Ig重链可变区的基因序列,利用IMGT/HighV-QUEST分析其基因取用、V区AA变化、CDR3氨基酸长度和构成以及连接多样性.结果:IgM、IgG和IgE均较多地取用IGHJ4、IGHV1-69和IGHV5-51,且在IgM、IgG、IgA与IgE中可观察到几个相似的优势V-J配对.IgD具有较高的V区突变,主要体现在FR3区的高突变(AA变化值为7.2±2.4),而IgM的每个结构域的突变均明显低于其他类Ig.此外,IgD具有较高P插入发生率和插入核苷酸数明显多于其他四类Ig.结论:本研究从基因特征上描绘了人类五类Ig相互间的相似性与差异性,这些结果可为抗体工程领域提供重要参考数据.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
