家猪BMP-2、BMP-3基因克隆及其组织表达探究

本研究以长白猪(Landrace)肺cDNA为模板,扩增获得猪骨形态发生蛋白BMP-2和BMP-3基因的cDNA全长.BMP-2基因cDNA全长为1742bp,编码395个氨基酸的前体蛋白,与人、牛、绵羊、大鼠、小鼠等的BMP-2基因的氨基酸序列一致性分别94.18%,95.4%,95.2%,90.3%,90.60%;BMP-3基因cDNA全长为1639bp,编码475个氨基酸的前体蛋白,猪与人、牛、绵羊、大鼠、小鼠等的BMP-3氨基酸序列的一致性分别是84.7%、87.7%、82.3%、80.4%、79.5%.采用RT-PCR方法,对BMP-2、3基因在家猪主要组织的表达进行探究.结果显示:BMP-2在所有组织中均有表达;而BMP-3在脑、肺、小肠中表达量较高,其他组织中表达量弱.     

一个麻疯树C2H2型锌指蛋白基因JcZFP1的克隆与表达分析

从麻疯树胚乳cDNA文库中克隆得到一个C2H2型锌指蛋白基因,JcZFP1,全长931bp,开放阅读框全长735bp,编码244个氨基酸残基,等电点pI=7.0,分子量PA=27.38kD.C2H2型锌指结构位于第81至103个氨基酸残基中,包括植物锌指蛋白特有的保守氨基酸序列QALGGH.在该基因的顺式作用元件上发现了与生长发育和逆境胁迫相关的启动元件.荧光定量PCR(qPCR)检测发现,JcZFP1基因在麻疯树各器官中的表达量依次为:绿果皮花叶根茎种仁,且幼嫩器官表达量高于成熟器官;对麻疯树幼苗进行低温、干旱胁迫时,JcZFP1基因的表达量明显增加,表明该基因可能在麻疯树的生长发育及逆境胁迫应答过程中起重要作用.     

猪SOSC1、SOCS3和SOCS4基因克隆及组织表达研究

本文以长白猪(Landrace)大脑cDNA为模板,克隆得到长白猪细胞因子抑制因子SOCS-3基因,并首次克隆得到长白猪SOCS-1和SOCS-4基因.此外,还利用长白猪基因组DNA为模板克隆得到SOCS-3假基因(pseudo-SOCS-3).序列分析显示:长白猪SOCS-1基因cDNA编码220个氨基酸的前体蛋白,与人、小鼠、大鼠和牛的氨基酸序列一致性分别为94％、91％、91％和94％;SOCS-3基因cDNA全长690 bp,编码229个氨基酸的前体蛋白,与人、大鼠和小鼠的氨基酸序列一致性高达94％以上;SOCS-4基因cDNA全长1404 bp,整个编码区没有内含子插入,编码441个氨基酸的前体蛋白,与人、大鼠和小鼠的氨基酸序列一致性分别为97％,89％和86％.氨基酸结构分析显示,长白猪SOCS-1,SOCS-3和SOCS-4基因都具有典型的中央SH2结构域和C末端有40个氨基酸的SOCS Box结构域;SOCS-4有长约285个氨基酸的N末端,且没有特殊的结合基序,其长末端存在的生理意义有待进一步研究.采用RT-PCR方法,对长白猪SOCS-1,SOCS-3和SOCS-4基因进行组织表达分析.结果显示它们在所有组织中都有表达.     

枸杞抗坏血酸氧化酶基因的克隆与表达分析

以枸杞为材料,利用同源克隆技术,克隆了枸杞抗坏血酸氧化酶(AO)的全长cDNA序列,命名为LcAO基因(GenBank:KP712033),该基因开放阅读框(ORF)大小为1737bp,编码578个氨基酸,与西红柿AO蛋白的同源性达到90%.LcAO编码的氨基酸序列包含3个多铜氧化酶结构域和跨膜信号序列.采用实时荧光定量PCR分析显示,LcAO基因在枸杞的花和果实中表达量最高,成熟叶中表达量最少.     

波纹唇鱼神经肽Y基因的克隆及原核表达

以波纹唇鱼为实验对象,采用同源基因克隆技术和RACE技术得到神经肽Y基因的cDNA全长序列.波纹唇鱼npy的cDNA全长序列为645 bp,包含59 bp的5'UTR、300 bp的ORF和286 bp的3'UTR.分子进化的比对分析表明,波纹唇鱼NPY前体与斜带石斑鱼和锯隆头鱼的亲缘关系最近.波纹唇鱼ORF编码99个氨基酸,NPY前体多肽包括:信号肽、成熟肽、Gly-Lys-Arg翻译后加工位点和羧基末端侧翼肽段(CPON).该NPY前体的预测分子质量为11.27 kD,理论等电点为5.51.采用pET-32a(+)原核表达系统构建波纹唇鱼npy基因的原核表达重组子pET-32a(+)-NPY,并获得重组菌株.对重组蛋白表达菌株进行培养条件优化,结果表明:该菌株在30℃,0.6 mmol/L IPTG,培养时间8 h的条件下,重组蛋白的表达效率最高.     

元江普通野生稻金属硫蛋白基因的获得和序列分析

对SMARTTM技术构建的元江普通野生稻叶片cDNA文库进行随机测序,获得了元江普通野生稻的金属硫蛋白基因cDNA序列.该序列全长525 bp,开放阅读框长186 bp,编码62个氨基酸,10个半胱氨酸集中分布在肽链的N端和C端,该蛋白的分子量为6.42 kD,理论等电点为5.21.氨基酸序列(Blastp)同源性分析表明该基因属于金属硫蛋白家族.     

青蛤丝裂原活化蛋白激酶(MAPKs)基因的克隆及其在鳗弧菌胁迫下的表达分析

采用Illumina MiSeq二代测序方法筛选得到青蛤丝裂原活化蛋白激酶(MAPK/p38)基因相似序列,进一步克隆得到全长cDNA.利用荧光定量PCR技术检测该基因在青蛤各组织中以及在鳗弧菌胁迫下的组织表达特征.结果显示,该基因全长共1781bp,开放阅读框长度为1104bp,编码367个氨基酸.在正常条件下该基因在青蛤体各组织中都表达,其中血淋巴中表达量最高,肝脏次之,鳃中最少.经菌液侵染后12h青蛤血淋巴中表达量即达到峰值,随后表达量逐渐降为正常水平.     

山羊KLF17基因克隆与组织表达

克隆山羊Krüppel样因子17基因(KLF17)的c DNA序列,并检测其组织表达水平,为KLF17基因的功能研究奠定基础.随机选取2~3周岁的健康简州大耳羊和藏山羊羯羊各4只,屠宰采集山羊不同组织样品(心、肝、脾、肺、肾、肌肉和皮下脂肪),提取组织总RNA,利用RT-PCR法克隆山羊KLF17基因序列,进行生物信息学分析,并利用实时荧光定量PCR(q PCR)法检测KLF17基因mRNA的表达水平.结果表明,克隆得到山羊KLF17基因c DNA序列1 450 bp,包含CDS区全长858 bp,5'UTR序列12 bp,5'UTR序列580 bp,编码285个氨基酸残基.其CDS区与牛、绵羊相比在5'端少99 bp,而在531位多出361 bp,这导致其编码区在858位提前终止.山羊蛋白序列较牛、绵羊少113个氨基酸残基.组织表达结果显示,KLF17基因在藏山羊肺中具有最高的表达水平,而在简州大耳羊皮下脂肪中具有最高的表达水平.两种山羊均在背最长肌中具有最低的KLF17表达量.这些结果表明KLF17可能在山羊脂质代谢过程中具有重要调控作用.     

珍珠鸟和家鸡甲状旁腺激素3型受体(PTH3R)的结构与表达图谱分析

本研究采用RT-PCR方法,首先克隆珍珠鸟和家鸡的PTH3R基因全长cDNA序列.结果显示,家鸡PTH3R(cPTH3R)cDNA全长1632bp,编码543个氨基酸,珍珠鸟PTH3R(zPTH3R-w)cDNA序列全长1563bp,编码520个氨基酸,其蛋白均含有信号肽序列、七次跨膜区等特征性结构.此外,在珍珠鸟中还发现一个新剪接变体zPTH3R-v1,其cDNA序列全长1468bp,编码488个氨基酸,其缺失第3外显子进而导致第1跨膜结构域缺失.利用生物信息学方法,我们还对珍珠鸟和家鸡PTH3R蛋白序列进行三维建模.采用RT-PCR方法,本研究也对珍珠鸟PTH3R基因进行组织表达分析.结果显示,zPTH3R及其剪切变体zPTH3R-v1在珍珠鸟脑及外周组织中广泛表达.     

青蛤(Cyclina sinensis)亲环素A基因的克隆与鳗弧菌刺激后的表达分析

采用SMART-cDNA文库和高通量测序方法,筛选出青蛤亲环素A基因(CypA)的全长cDNA.采用荧光定量PCR法分析该基因在鳗弧菌刺激后的表达情况,并对青蛤Cyp A基因进行原核表达与蛋白纯化.结果表明,青蛤CypA基因全长共962 bp,其中5′UTR为87 bp,3′UTR为346 bp,开放阅读框长度为495 bp,可以编码164个氨基酸,理论相对分子质量约为19.39 kDa,等电点约为5.25.在菌液侵染刺激条件下,刺激3 h后表达量即达到峰值,随后表达量逐渐降为正常水平.本研究成功表达了重组CypA蛋白,并运用镍离子交换柱法分离纯化出该蛋白,其相对分子质量约为19 kDa.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
