不同型口蹄疫病毒1D蛋白的βG和βH链之间都含有一个高度保守的RGD基序,其结合细胞受体,起始病毒感染.但本文通过对Asia1型FMDV的1D序列分析证实,田间毒株含有RGD和RDD基序(RGD中的G突变为D)的两种类型毒株.通过反向遗传技术制备了分别含有RGD和RDD基序的两种病毒,测定其生物学特性,并通过细胞感染力和乳鼠致病力比较这两种病毒感染力的差别.结果表明,含有RGD和RDD的毒株都具有感染性,含有RDD毒株的细胞感染力和乳鼠致病力比含RGD的高10倍左右,为进一步阐明RDD基序Asia1毒株的感染机制的分子基础奠定必要的基础.  相似文献

2009年4月, 北美地区出现甲型H1N1流感疫情, 并且疫情快速扩散至欧亚非三大洲, 风险等级上升至5级. 截止至5月13日, 甲型H1N1流感病毒已扩散至33个国家和地区, 实验室确认病例5728例, 死亡61人. 从NCBI的IRV和GISAID的EpiFluDB数据库下载甲型H1N1流感病毒序列425条, 进行序列比对与进化分析. 分析结果显示: (ⅰ) 当前流行的甲型H1N1流感病毒是一个三重排A型流感病毒: HA, NA, MP, NP和NS来源于猪流感病毒; PB2和PA来源于禽流感病毒; PB1来源于人流感病毒. (ⅱ) 猪流感病毒的来源可细分为: HA, NP和NS来源于H1N1亚型经典猪流感毒株; NA和MP来源于H1N1亚型禽源猪流感毒株. (ⅲ) 甲型H1N1流感病毒在流行扩散过程中没有显著变异, 同时病毒基因组没有发生重排. 本文除了分析美国代表毒株A/California/04/2009(H1N1)外, 还以墨西哥代表毒株A/Mexico/4486/2009(H1N1)为主进行了同源性和进化分析, 地理范围相对较大, 分析结果更具科学性.  相似文献

以10个甲型流感病毒北美毒株的基因PB2, PB1, PA, HA, NP, NA, NS和M进行遗传进化分析, 并用不同的软件对PB2, HA, NS1和M2蛋白氨基酸序列、糖基化位点及抗药性位点分析. 结果表明, 甲型流感病毒北美毒株的基因片段是一个来源于不同宿主、不同地域的重组株. HA蛋白氨基酸序列分析表明, 北美毒株HA蛋白的裂解位点氨基酸序列均为IPSIQSR↓G, 不具有高致病性流感病毒的特性, 同时NS1蛋白第92位氨基酸残基由天冬氨酸突变为谷氨酸(Asp→Glu), PB2蛋白氨基酸序列的第627位均为谷氨酸(E), 这些特性表明对人具有明显的亲和性, 但推测对人具有较低的致病力. M2蛋白的同源建模表明了北美毒株M2蛋白的药物敏感位点突变为抗药型位点, 推测北美毒株具有抗金刚烷胺类药物的结构特点, NA蛋白序列分析表明了北美毒株仍然对神经氨酸酶抑制剂类药物敏感. 这些研究结果对于我国预防和控制甲型流感北美毒株具有重要的参考价值.  相似文献
