To further realize the action of superoxide radicals (O2− .) in photoinhibition of photosystem Ⅱ (PSⅡ), we employed 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) as a spin trap, associated with EPR spectroscopy, to study the effect of illumination time on O2− . formation during high light photoinhibition in PSⅡ membranes and Mn-depleted PSⅡ membranes. Results indicated that the removal of Mn cluster from PS membranes has a strong influence on the dynamics of superoxide formation. The relative mechanism was also discussed. These novel findings may further promote the studies of the structure and function of PSⅡ and the mechanism of photoinhibition. 相似文献

Light and heat induced denaturation of CP47, the core antenna complex of photosystem Ⅱ purified from spinach, were investigated using absorption and circular dichroism spectra.Light caused the destruction of chlorophyll a and excitonic interaction of chlorophyll a in CP47, while the protein secondary structure was not apparently changed.Heat induced the destruction of protein secondary structure and excitonic interaction of chlorophyll a, but the chlorophyll a molecule was not damaged.The results suggest that both the chlorophyll a molecular structure and the protein native conformation are necessary for excitonic interaction of chlorophyll a and the energy transfer function of the chlorophyll a binding protein. 相似文献

Calcium ions play an important role in the oxygen_evolving process of photosystem Ⅱ as demonstrated in many experiments. The changes of the secondary structures of PS Ⅱ induced by the depletion of Ca 2+ were reported. The results indicated that the removal of Ca 2+ led to the transition of α helix to turns and sheet structures. While Ca 2+ was re_added to the media, only the structures changed to turns could be recovered. The protein conformational changes of PS Ⅱ during the donor side photoinhibition induced by the depletion of Ca 2+ were also studied. This showed that the protein conformational changes differed between the control and Ca 2+ _depleted samples in a short period of illumination (within 10 min). The influence of digalactosyldiacylglycerol (DGDG), one of the photosynthetic membrane lipids, on heat inactivation of the process of oxygen evolution has been studied in vitro in photosystem Ⅱ (PSⅡ) core complex. It was found that the temperature of semi-inactivation of oxygen evolution in the complex increased from 40.0 to about 43.0℃ in the presence of DGDG with 5-min heat treatment in the dark. Furthermore, when PSⅡ core complex was incubated for 5 min at 45.0℃, the oxygen evolution in the complex was completely lost, whilst the DGDG-complexed PSⅡ core complex still retained a 16% of activity (100% for 25.0℃). In addition, a 1-h incubation at 38.0℃ inactivated absolutely the oxygen evolution for the PSⅡ core complex. By contrast, there remained about 20% of activity (zero time for 100%) for the complex in the presence of DGDG under the same condition. These results indicate a new role of DGDG in the protection of PSⅡ core complex against the deleterious effects of temperature. It was most likely that DGDG-mediated stability toward thermal denaturation of oxygen evolution in PSⅡ core complex is due to the protective effect of DGDG on the release of the 33 kD protein from PSⅡ core complex. 相似文献

Hippocampal neurons were treated by thrombin and thrombin receptor activating peptides (TRAP). Cell survival rate was decreased in a dose-dependent manner by MTT assay. The numbers of apoptotic cell and apoptotic rate of hippocampal neurons treated by different concentrations of thrombin were increased in a dose-dependent manner by terminal deoxynucleotidyl transferase (TdT) mediated dUTP-biotin nick end-labeling (TUNEL) method and Flow Cytometry. When the concentration of thrombin is 40 U/mL, TUNEL positive cells and apoptotic rate of hippocampal neurons reached peak value, were 27. 3 4.0 and (29.333 4.633 ) %, respectively. Immunocytochemistry assay show that Bcl-2 protein expression was down- regulated and Bax protein expression was up-regulated with the concentration of thrombin increased. TRAP can mimic the effect of thrombin to induce apoptosis on hippocampal neurons. These data demonstrated that thrombin induced hippocampal neuron apoptosis in a dose-dependent manner through activating protease-activated protein-1 (PAR-1). The change in expression of Bcl-2 and Bax was related with the effect of high concentration thrombin induced apoptosis on hippocampal neurons. 相似文献

The effect of mutation of strongly conserved porelining residues in the chloride channel EcClC on the electrostatic potential and binding free energy of the chloride ion was studied using explicit protein-membrane structures. Electrostatic potential distribution and binding free energy of the chloride ion at different binding sites in the wild-type and mutated EcClC were calculated with APBS. The potential data reveal that the electrostatic potential around the selectivity filter, especially around the site Sext and Scen becomes more negative as the residue R147 was mutated to C147. The electrostatic binding free energy shows that the binding free energy of the chloride ion at all binding sites becomes more positive as R147 was mutated. It follows that mutation of R147 decreases ion stabilization at binding sites and affects channel's gating. 相似文献

This paper presents a study of the variation of the elevation of Tongguan, which is located in the backwater zone of the Sanmenxia Reservoir, in response to changes in flow runoff. The analysis indicated that the rise of the elevation of Tongguan, which is defined as the stage corresponding to a discharge of I000 m^3/s at Tongguan station, is controlled by the stream energy. A close relationship existed between the elevation of Tongguan and the superimposed stream energy that integrates the current and the preceding years‘ flow and dam operation conditions. When the flow runoff remains relatively constant and the pool level of the dam has a relatively large range of variations, then the elevation of Tongguan is primarily controlled by the dam operation conditions. On the other hand, if the flow runoff has a relatively large range of variations and the pool level of the dam remains relatively constant, then the elevation of Tongguan is primarily controlled by the flow conditions. These findings are of importance for optimizing the dam operation in order to lower and control the elevation of Tongguan, and therefore to minimize the backwater effect of the dam operation. 相似文献

This study intends to examine the effects of different concentrations of four kinds of degradations of chitin: glucosamine (GLC), N-acetyl-D-glucosamine (NAG), chitooligosaccharide(COS), CM-chitooligosaccharide (CM-COS)—on the proliferation of MC3T3-E1 cell line cultured in vitro. Results suggest that all of the glucoses mentioned above promoted the proliferation of osteoblast, and various concentrations have different effects: the proliferation was remarkable when the concentration of GLC, NAG, COS, CM-COS was 100 , 100, 500, 500 μg/mL ,respectively. Furthermore we choose the glucosamine as the material and study the effect on the bone strength in ovariectomized rats. The results showed that the middle does of glucosamine can significantly increase the hone strength of femur in ovariectomized rats. 相似文献

The detailed structure of catalytic center of water oxidation, Mn4Ca-cluster, in photosystem ⅡI (PSII) has been reported recently. However, due to the radiation damage induced by X-ray and the complexity of the Mn4Ca-cluster, the assignment of the μ4-O5 atom coordinated by three Mn and one Ca2+ ions is still lack of essential evidences. In this article, we synthesized one Mn complex containing two μ4-O atoms. It is found that the lengths of all μ4-O-Mn bonds in this Mn complex are in the range of 1.89-2.10 , which are significantly shorter than 2.40-2.61 distance of μ4-O5-Mn bonds in Mn4Ca-cluster observed in the crystal structure of PSII. In addition, DFT calculations have been carried out on the Mn4Ca-cluster. It is found that the O atom of μ4-O or μ4-OH always trends to deviate from the center position of four metal ions, resulting in unequal bond lengths of four μ4-4-M (M=Mn or Ca), which is obviously different with larger and nearly equal distances between μ4-O and four metal ions observed in the crystal structure. Based on these results, we suggest that the μ4-atom in Mn4Ca-cluster of PSII is unlikely to be a μ4-O, μ4-OH or μ4-OH2 , and its assignment is still an open question. 相似文献

The techniques of oxygen electrode polarogra-phy and Fourier transform infrared (FT-IR) spectroscopy were employed to explore the roles of polar head-group of phosphatidylglycerol (PG) molecules in the functional and structural aspects of photosystem Ⅱ (PS Ⅱ) through enzymatic approach. It was shown that the depletion of PG by treatment of phospholipase C (PLC) on PS Ⅱ particles caused the inhibition of oxygen evolving activity in PS Ⅱ. This effect also gave rise to changes in the protein secondary structures of PS Ⅱ, that is, an increase in a-helical conformation which is compensated by the loss of p-strand structures. It revealed that the head-group of PG molecules plays an important structural role in the maintenance of normal structure of PS Ⅱ proteins, which is required to maintain the appropriate physiological activity of the PS Ⅱ complex such as the oxygen evolving activity. It is suggested that there most probably exist hydrogen-bonding interactions between PG molecules and PS Ⅱ proteins. 相似文献

Understanding bacterial adsorption and the evolution of biofilms on arsenopyrite with different surface structures is of great significance to clarifying the mechanism of microbe–mineral interfacial interactions and the production of acidic mine drainage impacting the environment. In this study, the attachment of Sulfobacillus thermosulfidooxidans cells and subsequent biofilm formation on arsenopyrite with different surface structures in the presence of dissolved As(Ⅲ) was studied. Arsenopyrite slices with a specific surface were obtained by electrochemical corrosion at 0.26 V. The scanning electronic microscopy-energy dispersion spectra analyses indicated that the arsenopyrite surface deficient in sulfur and iron obtained by electrochemical treatment was not favorable for the initial adsorption of bacteria, and the addition of As(Ⅲ) inhibited the adsorption of microbial cells. Epifluorescence microscopy showed that the number of cells attaching to the arsenopyrite surface increased with time; however, biofilm formation was delayed significantly when As(Ⅲ) was added. 相似文献

The electromagnetic field energy distribution inside the left-handed metamaterials (LHMs) with or without blank and line defects is firstly investigated. The LHMs consist of an array of periodic unit cells of copper hexagon split ring resonators (SRRs) and wires. The internal energy distribution of the LHMs is measured in a microwave measuring line device at X-band frequencies. It is found that the LHMs only respond to the electromagnetic field around the resonance frequency. The electromagnetic field amplitude inside the LHMs generally decreases with the introduction of defects. It is suggested that the electromagnetic field energy reduction may arise from the symmetry breaking in the LHMs. 相似文献
