An improved electrophoretic method for a screening program for haemoglobinopathies

Summary A method for a screening program for haemoglobinopathies in a starch agar gel mixed with saponin is presented. Normal and abnormal blood containing haemoglobins S, C, I, M Boston, D Punjab, beta thalassaemia major and beta thalassaemia minor, were applied, in a tray with the capacity for 100 samples. The electrophoresis was performed in 45 min using 300 V. This method offers special advantages for the examination of a large number of samples, using a small amount of whole blood and without the previous preparation of haemoglobin solution.Acknowledgments. Effect of seasonal variations and cold acclimation on serum transaminase activity of common Indian frogRana tigrina

Summary A definite seasonal change is observed in 2 transaminases, SGOT and SGPT ofR. tigrina. Cold acclimation significantly depresses transaminase activity of serum.Acknowledgments. Freeze tolerance in the frog,Rana sylvatica

Summary Wood frogs survive extracellular freezing at moderate subzero temperatures (–4°C) for at least 11 days. Freezing survival is aided by the accumulation of high concentrations of glucose as a cryoprotectant in blood and tissues. Glucose production was accompanied by a rapid decline in liver, but not muscle, glycogen levels suggesting that liver is the organ controlling cryoprotectant synthesis.Acknowledgments. A component of polyphloretin phosphate (PPP) may be an alternative substrate for ATP-glycerol transferase

Summary A low molecular weight fraction of a polyphloretin phosphate (PPP) preparation simulated glycerol in a milieu designed for enzymatic assay of glycerol. It is suggested that a component of this PPP mixture is able to act as an alternative substrate for the ATP-glycerol transfer enzyme; it becomes phosphorylated and thus initiates the series of reactions resulting in a increased conversion of NADH to NAD and the observed changes in absorbance.The authors are pleased to acknowledge the support of Dr D.C. Preservation ofBordetella pertussis

Evolutionary implications of ascorbic acid production in the Australian lungfish

Summary It is shown that the Australian lungfish (Neoceratodus forsteri) synthesizes ascorbic acid in its kidney, suggesting that ascorbic acid synthesis started before the origin of the tetrapods.Acknowledgments. The local anesthetic potency of norcocaine,a metabolite of cocaine

Summary The local anesthetic effects of cocaine and one of its main metabolites norcocaine, were investigated comparatively on isolated ganglion cells of the marine gastropod,Aplysia californica. During a 1-hour-period, different action potential parameters such as amplitude, duration, maximum rate of rise were observed, which demonstrated that norcocaine exhibits a higher local anesthetic potency than cocaine.Acknowledgments. Selective and reversible modification of essential thiol groups of D-glyceraldehyde-3-phosphate dehydrogenase by isothiocyanates

Summary Aralkyl and alkyl isothiocyanates, like aryl isothiocyanates, undergo a selective and reversible reaction with essential thiol groups of D-glyceraldehyde-3-phosphate dehydrogenase; the former 2 substances require for a reversible reaction course a more alkaline medium and presence of a thiol.The authors are indebted to Dr J. Effective dose present in cockroach larvae exposed continuously to a juvenile hormone active insect growth regulator

Summary Larvae of the German cockroach exposed to filter paper impregnated with juvenile hormone (JH) active substance contain, at the middle of the last instar, about one-hundredth of the dose applied to 1 cm². Anti-endotoxin (anti-lipid-A) antibodies

Summary Anti-lipid-A, anti-endotoxin, antibodies have been measured by a passive haemolysis test using antigen from anE. coli Re mutant. Titres in the normal population are low but do rise in situations in which there has been gram-negative sepsis. Absence of raised titres in other situations has profound implications.We are most grateful to Dr Günter Schmidt of Freiburg for his gift of Lipid-A antigen. We also thank Dr L. α-Thioglycerol sensitive excitatory oxytocin receptors in fowl rectum

Summary The selective antagonism of the excitatory responses of oxytocin by -thioglycerol and its potentiation by Mg⁺⁺ indicate the presence of specific oxytocin receptors in fowl rectum. In addition, the sensitivity of the rectum to very small doses of oxytocin suggests a possible facilitatory role of neurohypophyseal hormones in oviposition.Acknowledgment. Metabolically regulated cyclical contractures in microinjectedSpirostomum: A pharmacological study

Summary Spirostomum was treated extracellularly and intracellularly with a range of metabolites to investigate the intracellular regulation of cyclic calcium movements. The results indicate, close links between calcium movements and mitochondrial metabolism.We are grateful to Dr.A. Chromsome aberrations in cattle raised on bracken fern pasture

Summary Thirteen cows maintained on natural bracken fern (Pteridium aquilinum) were analyzed cytogenetically. The frequency of structural chromosome aberrations detected in peripheral blood cells was significantly higher when compared to that detected in animals raised on pasture containing no bracken fern. The application of sodium deoxycholate and Sephacryl-200 for the delipidation and separation of high density lipoprotein

Summary A method to remove lipids from human plasma high-density lipoprotein (HDL) was developed. The procedure required column chromatography on Sephacryl-200 in the presence of the bile salt sodium deoxycholate. The lipid free protein obtained retained the immunological properties of the native HDL.The author is grateful to Ms L. Gray for her excellent technical assistance and Dr M. A chromosome study ofScarites occidentalis (Coleoptera,Caraboidea)

Summary Scarites occidentalis has 2n=41 (females 2n=42), n=19+X₁X₂Y. This multiple sex-chromosome system and other karyotypic characteristics suggest that this species andS. buparius share a recent common ancestor in which the sex trivalent was probably originated.Thanks are due to Dr N. Alopecia in laboratory animals induced by a polyampholyte,polyethylene alanine

Summary Polyethylene alanine, a polyaminoacid, was found to induce a characteristic alopecia when administered topically or systemically to suckling mice, or newborn guinea-pigs or rabbits.Acknowledgments. Summary The site of sex pheromone release inMusca domestica andGlossina morsitans is the legs. Unicellular glands restricted to the tarsi and to the tibia are proposed as the source of the pheromone. On the mechanism of killing ofTrypanosoma cruzi by human polymorphonuclear leukocytes

Summary The temperature-dependence of some processes involved in the killing of sensitizedT. cruzi epimastigotes by human polymorphonuclear leukocytes (PMN) was determined. The rate of the reactions was related to the temperature of incubation according to the Arrhenius equation and the apparent energies of activation (Ea) were calculated. The Ea values separated these complex reactions into two groups: one with Ea of about 10 kcal/mol for the phagocytosis of the parasites and the release of lysosomal enzymes by PMN, and the other with Ea of about 22 kcal/mol for the cytotoxicity against sensitizedT. cruzi, the rate of oxygen consumption by PMN, and the lysis of the parasites with added hydrogen peroxide.This work was supported by research grants from CONICET and SUBCYT, Argentina, and UNDP/Word Bank/WHO Special Programme for Research and Training in Tropical Diseases.
