The effect of phenazine methosulfate on the activity of dehydrogenases as studied by starch gel electrophoresis

Résumé L'effet de la phenazine methosulfate (PMS) sur l'activité de plusieurs enzymes oxydatifs a été étudié. En tant que transporteur d'électron, la PMS a un rôle important sur la coloration des isoenzymes séparés par électrophorèse en gel d'amidon. En ce qui concerne la déshydrogénase lactique, un excès de PMS provoque une coloration diffuse, alors que dans le cas de la déshydrogénase succinique on observe l'apparition de «bandes négatives» qui n'ont pas encore été décrites.

Identification of human serum proteins binding iron,copper, and thyroid hormones by starch gel electrophoresis

Résumé On a analysé au moyen d'électrophorèse sur gel d'amidon les protéines qui, dans le sérum humain, lient le fer, le cuivre et l'hormone thyroïdienne, en utilisant comme témoins des protéines purifiées et la radioactivité. Le fer est lié par la -globuline qui, dans certaines familles, paraît se diviser en deux composants. Le cuivre est lié par la céruloplasmine, qui émigre dans la position ₂ rapide. La protéine principale liant la thyroxine est la préalbumine.     

Separation of serum proteins in different amphibian species by polyacrylamide gel electrophoresis

Zusammenfassung Mittels Polyacrylamid-Gel-Elektrophorese wurden die Serumproteine von 11 Amphibienarten in 15 bis 20 Fraktionen aufgetrennt. Das elektrophoretische Muster wurde mit demjenigen des menschlichen Serums verglichen und erwies sich als artspezifisch.

Esterases of the flounder (Platichthys flesus,Pleuronectidae, Teleostei): Development of an identification protocol using starch gel electrophoresis and characterization of loci

Summary Fish esterases are among the most difficult enzymes to identify using starch gel electrophoresis because of the many loci that are simultaneously active, and especially because of duplication phenomena, satellite bands, and stain trails. In an attempt to simplify and clarify electropherograms, various staining and inhibitory methods were tested on esterases from the flounderPlatichthys flesus. A range of migration and staining buffers and substrates were used, as well as chemical inhibition and heat inactivation. A combination of the methods made it possible to distinguish and characterize the five presumed esterase loci.     

Two-dimensional electrophoresis of rat serum esterases in cellulose acetate and acrylamide gradient gel

Zusammenfassung Isoenzyme von Serumesterasen wurden zuerst mit Zelluloseazetat und dann mit einem Gradientgel von Polyakrylamid mit zweidimensionaler Elektrophorese bei der Ratte untersucht. Ein neues Band, das eine starke Albuminesteraseaktivität aufweist und unempfindlich gegen 10^–3 M Physostigmin ist, wurde somit gefunden. Von 17 Esterasebändern waren 7 empfindlich auf Physostigmin, und 3 wurden auch durch Iso-OMPA gehemmt, während 284-C-51 gar keinen Effekt zeigte.

A starch gel electrophoretic study of the hemolymph proteins of some bermuda crustacea

Zusammenfassung Haemolymphe Proteine von 10 dekapoden Krebsen aus den Bermudas wurden mit Hilfe der «Starch-Gel»-Elektrophorese untersucht. In acht Arten erwiesen sich die zwei langsamsten Proteinkomponenten als Kupfer-positiv, in zwei Arten wurde nur eine einzige Kupfer-positive Bande ermittelt. Die Esteraseaktivität war mit den meisten Kupfer-positiven Banden verbunden, wobei 10^–4 Eserinsulfat keine Hemmung der Aktivität herbeiführte. Diese und weitere Versuche legen die Vermutung nahe, dass bei einigen Crustaceen zwei Molekülarten von Haemocyan vorkommen.

Rapid quantitative evaluation of serum LDH isoenzyme patterns after agar gel electrophoresis

Resumen Las isoenzimas de la LDH se separan por electroforesis en gel de agar, y luego de colorear con una sal de tetrazolio, se cuantifican empleando una técnica rápida de elución; los resultados no difieren significativamente de los obtenidos por cuantificación densitométrica.     

Tissue- and stage-specific RNA patterns in Drosophila as revealed by gradient formamide gel electrophoresis

The in vitro labeled RNAs in the accessory glands from male adults, and in the salivary glands and fat body from developing larvae of Drosophila melanogaster were separated on linear gradient acrylamide gels in formamide. The patterns appear to be tissue- and stage-specific, and several lines of evidence indicate that the visualized radioactive bands include both hnRNA and messages.     

Identification of proteins in human prostate tumor material by two-dimensional gel electrophoresis and mass spectrometry

Protein patterns in cells collected from benign prostatic tissues and prostate carcinomas were analyzed using two-dimensional polyacrylamide gel electrophoresis and mass spectrometry. Polypeptide expression was evaluated by computer-assisted image analysis (PDQUEST). Proteins expressed by prostate tumors were identified via in-gel digestion and subsequent matrix-assisted laser desorption/ionization mass spectrometry. In addition to cytoskeletal and mitochondrial proteins, a 40-kDa protein was identified as prostatic acid phosphatase (PAP). PAP expression decreased approximately twofold between benign and malignant tissue. Increased expression of heat shock protein 70 and decreased expression of tropomyosin 1 were also observed in the malignant tissue. The analysis of prostate material by two-dimensional gel electrophoresis and mass spectrometry shows that particular proteins are of interest as markers of disease. Further studies on polyacrylamide gel electrophoresis of water-soluble proteolipid-protein from bovine brain white matter

Zusammenfassung Wasserlösliche, proteolipide Proteine wurden von der weissen Hirnsubstanz durch Isolierungsmethoden vonTenenbaum undFolch ² gewonnen. Ohne Lyophilisation ergab die Disk-Elektrophorese bei pH 8.3 zwei Banden.
