文章以酿酒酵母(Saccharomyces cerevisiae)诱变菌株HXS-02为出发菌株,以菌株的生长、总硒和有机硒质量浓度及产率为指标,考察无机硒添加方式及还原型辅酶Ⅱ(NADPH)或果糖的添加对酵母富硒培养的影响。结果显示:菌株富硒培养时加入亚硒酸钠的适宜时间和适宜质量浓度分别为8 h和62.5 mg/L,在此条件下继续培养至24 h,菌株的干质量浓度和富硒能力最好,总硒产率和有机硒产率分别达到(8 760.7±50.6)μg/L和(8 573.5±46.5)μg/L;NADPH和果糖的添加可显著影响菌株的生长和富硒能力,在无机硒添加的同时向培养基中添加NADPH至浓度为0.20 mmol/L或果糖至质量浓度为6 g/L后,菌株的总硒产率和有机硒产率与未添加的菌株相比,分别增加了32.6%~42.6%和38.1%~51.5%。以上研究表明,适合的无机硒添加方式并辅以添加适量的NADPH或果糖,可以有效促进酵母转化无机硒为有机硒的能力,提高富硒酵母总硒和总有机硒的产率。  相似文献

将酵母接种在添加亚硒酸钠的培养液中进行驯化,获得富集有机硒的酵母,以柑子汁为主要原料,用富硒酵母酿制富硒保健酒.采用正交试验得到的最佳生产工艺条件为:富硒酵母接种量为12%,加糖量7%,发酵温度36℃,发酵时间为4d.所得产品有机硒含量可达12~17μg/m1,是一种理想的富硒营养保健品.  相似文献

文章以复原乳为原料,利用富硒青春双歧杆菌和干酪乳杆菌发酵制备一种新型的富硒酸奶。在单因素试验基础上,利用中心组合试验优化得到富硒酸奶的最优发酵条件如下:奶粉添加量为12%,蔗糖添加量为5%,低聚异麦芽糖添加量为1%,亚硒酸钠添加量为61.8μg/kg ,酪蛋白酸钠添加量为1.4%,接种量为4.5%,发酵温度为40℃,发酵时间为7 h。该富硒酸奶呈乳白色,凝乳完全,无乳清析出,组织细腻均匀,奶香纯正,酸甜适中,其有机硒的质量比为29.2μg/kg。在25 d (4℃)冷藏期间,pH值、乳酸菌数、蛋白质、脂肪和非脂乳固体质量分数呈不同程度下降趋势,但均符合国家标准的要求,有机硒的质量比相对稳定。  相似文献

本实验以亚硒酸钠(Na2 SeO3)为硒源对黑木耳进行富硒发酵,以获得富硒黑木耳发酵液,为有机硒多糖等开发利用提供依据.用3种不同碳氮源培养基26℃于200rpm进行液体发酵,在不同时间测定黑木耳菌丝体多糖含量和生物量,确定了最优培养基(每升)为葡萄糖20g,豆饼粉5g,硫酸镁0.5g,磷酸二氢钾2g.在优选培养基中通过添加不同浓度的亚硒酸钠对黑木耳进行富硒发酵.结果与不添加亚硒酸钠黑木耳发酵液相比,在硒浓度为10-50mg/L时,黑木耳菌丝体的硒含量和生物量及发酵液多糖含量均随硒浓度的增加而提高;在50mg/L时,硒含量、生物量和多糖含量达最高,分别为10.48μg/g、9.37g和L20.3g/L;当亚硒酸钠浓度高于50mg/L时,菌丝体硒含量降低,生物量、多糖含量亦下降.结果表明,黑木耳具有较好富硒能力,但培养基中亚硒酸钠的浓度会影响发酵液中多糖含量和黑木耳菌丝体的富硒效果.与不添加亚硒酸钠黑木耳相比,添加适量亚硒酸钠有利于黑木耳菌丝对硒的吸收利用,并可促进黑木耳生长和多糖含量的提高;但过量添加,则可抑制黑木耳生长并降低多糖含量和对硒的利用率.  相似文献

以谷氨酸棒杆菌作为原始出发菌株,通过常压室温等离子体诱变技术,确定高产琥珀酸等离子体诱变的物理参数和最佳诱变条件,采用在培养基中添加溴甲酚紫作为酸碱指示剂的方法,测定发酵液前后在波长590 nm处的吸光值,高通量筛选出在该波长处吸光值变化较大的突变株.结果表明:在诱变菌株的存活率为8%的条件下获得的突变株的正突变率达到22%.用40 g/L的葡萄糖对突变株进行摇瓶分批发酵,筛选得到诱变菌E7产琥珀酸15.6 g/L,乳酸22.8 g/L,该菌具备良好的产琥珀酸与有机酸的能力.  相似文献

为了提高Monacolin K的产量,以紫红曲霉M2为出发菌株进行诱变育种(紫外诱变、常压室温等离子体ARTP诱变),通过构巢曲霉对峙培养、高效液相色谱HPLC等方法筛选高产Monacolin K的优秀突变菌株。结果显示:相对于原始出发菌株M2,两种方法均能够高效筛选到红曲菌株,对6株高产Monacolin K诱变菌株进行5次传代培养,发现诱变菌株产Monacolin K的能力均有下降;但紫外诱变菌株Z-4和ARTP诱变菌株M-43的Monacolin K产量分别下降2.83%和1.97%,表现为良好的遗传稳定性,说明Z-4和M-43具有潜在应用价值。  相似文献

利用设施无土栽培,以亚硒酸钠作为外源硒,研究不同亚硒酸钠浓度对五个品种洋葱鳞茎的产量、总硒含量、无机硒含量以及有机硒含量的影响.结果表明,当亚硒酸钠的浓度为50 mg/kg时,品种奥林匹克的鳞茎有机硒含量最高,为2.08 mg/kg,产量为2241.7 g/m2,产量和有机硒含量的交互效应与其它品种的各处理存在极显著差异.另外,随着亚硒酸钠溶液浓度的升高,洋葱鳞茎的有机硒含量随之降低.  相似文献

对一株拮抗猕猴桃叶枯病的美丽短芽孢杆菌NF2进行He-Ne激光诱变育种,通过测定菌株的生长曲线确定诱变时的菌龄为摇瓶培养8h,此时菌体没有产芽孢;分别设置不同的诱变强度和时间,确定最佳的诱变条件为5mW,10min,此时菌株的拮抗效果提高了20.63%;SPSS软件分析诱变后菌株拮抗能力可稳定遗传至少15代。He-Ne激光诱变是一种安全有效的微生物育种技术,低剂量激光辐射更有利于菌株诱变效果的提高。  相似文献

从贵州乌当区富硒土壤中筛选亚硒酸钠还原菌并进行菌种鉴定,测试菌株硒素耐受能力及还原生成红色单质硒特性。结果表明:菌株X1704经16S rDNA序列分析鉴定为粘质沙雷氏菌(Serratia marcescens);菌株对亚硒酸钠有较强耐受能力,其最低抑制浓度(MIC)高达3500 mg/L;硒胁迫作用下,菌体的生长受到一定程度的抑制,稳定期也相应延迟;菌株能把96.8%的亚硒酸钠还原为红色单质硒。  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
