Steady-state absorption spectroscopy, circular dichroism, and resonance Raman spectroscopy have been used to investigate the thermal stability of LH2 complex isolated from purple photosynthetic bacterium Rps. palustris. The results show that: 1) upon increasing the temperature, a transition from B800 and B850 to free bacteriochlorophyll (B780) happens; 2) a gradual decrease and disappearance of CD signal in visible region occur; 3) a shift of the frequency, belonging to C=C and C-C stretching vibration, to higher wavenumber takes place. It is suggested that LH2 complex can be dissociated in the presence of B800, B850 and carotenoids simultaneously. Single-exponential fitting on the dynamic decay curves gives the apparent time constants of hundreds of minutes for various pigments.  相似文献

Leymus racemosus is highly resistant to wheat scab (Fusarum head bright). The transfer of scab resistant gene from L. racemosus to Triticum aestivum is of great significance for broadening the base of wheat resistance. In the present study, the pollen of T. aestivum-L, racemosus monosomic addition line with scab resistance was treated by irradiation with 1200 R ^60Co-γ-rays prior to pollinating to emasculated wheat cv. Mianyang 85-45. Nine plants with a telocentric chromosome 7Lr#1S were observed in M1, and one ditelosomic substitution line 7Lr#1S was selected from selfcrossing progenies and confirmed by chromosome C-banding and GISH. Furthermore, a co-dominant EST-SSR marker CINAU 31 was employed to identify this substitution line. A pair of chromosome 7A of common wheat were found to be replaced by a pair of telocentric chromosome 7Lr#1S, and further investigation showed that chromosome configuration of the substitution line at MI of PMCs after GISH was 17.50Ⅱ^w + 2.19Ⅱ^w + 0.42Ⅱ^7Lr#1S + 1.08 Ⅰ^7Lr#1S + 0.69 Ⅰ^w. Two telocentric chromosomes paired as a bivalent in 59.7% of PMCs. Abnormal chromosome behaviors of telocentric chromosomes were observed in part of PMCs at anaphase I and telophase I, including the moving of two telocentric chromosomes to the same pole, lagging and earlier separation of their sister chromatid. All these abnormal behaviors can be grouped into three distinct types of tetrads according to different numbers of 7Lr#1S in their daughter cells and various micronucleus in some tetrads. However, due to the high transmission frequency of the female and male gametes with a 7Lr#1S, 84% of the selfcrossing progeny plants had ditelosomic substitution. The substitution line showed high resistance to wheat scab in a successive two-year test both in the greenhouse and field; hence, the line will be particularly valuable for alien gene mapping, small fragment translocation induction and telosomic cytological behavior analysis.  相似文献

The aim of this study is to explore whether RT-5, one novel active ginsenosides from Ginseng, has protective effects against cisplatin(CDDP)-induced nephrotoxicity in vivo. One model of acute renal failure induced by CDDP in rats and one model of xenograft tumors of human cervical cancer in nude mice are established. Four groups are assigned: control, CDDP, RT-5, CDDP plus RT-5, in which the RT-5 is administered via oral gavage 24 h before CDDP intraperitoneal injection. The significant increase in blood urea nitrogen and creatinine are induced by CDDP treatment at 6 mg/kg, which are attenuated by pre-treated with RT-5 at 10 mg/kg. RT-5 could ameliorate CDDP-induced morphological damages in kidney by PAS staining, and reduce tubular apoptosis evaluated by TUNEL staining. Pretreatment with RT-5 notably inhibits CDDP-induced oxidative stress in kidney tissues. Interestingly, RT-5 does not interfere with the in vivo anti-cancer effects of CDDP against the growth of xenograft tumors in nude mice. These data suggest that co-treatment RT-5 with CDDP might attenuate the following nephrotoxicity without inhibiting its anti-tumor efficiency, which could provide one novel strategy for cancer treatment in clinics.  相似文献

目的:寻找一次染毒条件下,腹腔注射阿霉素诱导小鼠骨髓细胞微核率出现峰值的时间-剂量条件。方法:将150只小鼠随机分为5组,每组30只,雌雄各半。腹腔注射,1次给药,对照组给生理盐水,处理组的阿霉素剂量分别是2.5 mg/kg、5 mg/kg、10 mg/kg和20 mg/kg。染毒18 h、24 h、30 h、48 h和72 h后取小鼠骨髓进行微核分析,评价阿霉素对小鼠的遗传毒性作用。结果:理想的腹腔注射诱导的小鼠骨髓细胞微核率峰值的条件就是:一次染毒,阿霉素剂量在10%LD50即2.5 mg/kg至80%LD50即20 mg/kg的范围内剂量越大微核率越高,取样时间在30~48 h间微核率最高。  相似文献

以熔融萘作稀释剂,用3,4-二氨基苯甲酸测定Pt是一种有效的分光光度法.Pt与3,4-二氨基苯甲酸(DBA)在90 ℃生成蓝绿色的配合物,在pH 10.0~12.5范围内,配合物定量地被萃取进入熔融萘中.有机相溶解于无水CHCl3中,在715 nm处以试剂为空白测定其吸光度,在Pt浓度为(0.1~2.0)×10-6 mol/L范围内服从比耳定律.摩尔吸光系数和桑德尔灵敏度分别为1.2×106 L/mol*cm和 0.000 41 mg/cm2.实验确定了操作条件,考察了各种干扰离子的影响,并测定了混合样品中的Pt含量.  相似文献

A new method to calculate the formation enthalpies and free energies of compounds based on standard electromotive force of cation, φ0, and free energies of formation, ΔG2980, or formation enthalpies, ΔH2980, of chloride was proposed. A linear relationship between φ0 and ΔG2980, φ0 and ΔH2980, and ΔG2980 and ΔH2980 was found respectively. The parameters used for thermodynamic calculation were calculated. The calculated values of the formation enthalpies and the free energies of formation of platinum compounds, such as Pt3(PO4)2 and Pt(PO3)2, were given for the first time. The mechanism of eliminating platinum inclusions in phosphate laser glass by POCI3 gas bubbling was interpreted from thermodynamic aspect.  相似文献

研究高分子顺铂(PGA-Asp-CDDP)腹腔给药昆明鼠,1h后取出脏器检测其铂含量.石墨炉原子吸收光谱测定获得的铂浓度标准曲线,其相关系数为0.997 5,特征浓度为2.4μg·L-1;并考察了消解液用量和消解温度对测定铂浓度的影响;回收率实验表明该消解方法在测定铂浓度上具有可行性和可靠性.在此基础上,昆明鼠的心脏、肝脏、脾脏、肺和肾脏的铂含量检测结果分别为(5.46±0.89)、(28.65±13.45)、(45.30±4.07)、(16.90±4.74)和(76.31±2.59)μg·g-1.  相似文献

Mercury(II) is an important factor in hepatotoxicity that can enter the body through marine diets and amalgams.In the present study,the protective effect of the Eriobotrya japonica flower extract(EJFE) on HgCl 2-induced hepatotoxicity was investigated.Five mg/kg of mercuric chloride in drinking water was given to rats either with saline or EJFE(100 and 200 mg/kg as intraperitoneal(IP)) for 30 d.The mercury levels in different groups of liver tissues of the rats were measured with flameless atomic absorption spectroscopy(F-AAS).Also,mercury accumulation in the liver of the rats was modeled by using a parallel chemical kinetic model.The results showed that HgCl 2-induced oxidative damage led to a significant decrease in glutathione(GSH) and the total antioxidant capacity(TAC) levels,and to a significant increase in lipid peroxidation level.Accumulated mercury was 14.47% more in the livers of the stress groups than in those of the control groups(P<0.001),whereas the amount of Hg was adjusted to 13.49% and 13.93% in groups treated with 100 and 200 mg/kg of EJFE respectively,as compared with stress groups(P<0.001).HPLC analysis of EJFE revealed that hesperetin and gallic acid are the major antioxidants in EJFE.Results demonstrate that flowers of the Eriobotrya japonica cause a significant protection against HgCl 2 induced hepatotoxicity in all diagnostic parameters by strengthening the antioxidant defense mechanisms and they may have a therapeutic function in free radical mediated diseases.  相似文献

 制备抗癌药顺铂和卡铂在水溶液中的降解产物-羟桥双核铂(Ⅱ)化合物,研究其化学结构.以顺铂为起始原料,通过水合和313nm紫外光照射,分离出目标化合物.通过测定铂含量,ESI-MS,1H NMR和单晶X衍射分析测定其结构.所得到的化合物为以羟基搭桥的双核铂(Ⅱ)配合物,中心离子与2个氮原子和2个氧原子组成平面正方形,2个中心离子之间的距离为0.3095nm,有弱作用.2个配位平面的二面角为2.7°,几乎处于同一平面.2个硝酸根处于配位的外界.  相似文献

The present study aims at investigating the potential impacts of the nC60 complex with atrazine on the development of Japanese medaka (Oryzias latipes) embryos and chronic toxicity to D. magna. Although exposure to nC60 below 8 mg/L had no effects on the hatching rate of medaka embryos, it increased the deformity of hatched larvae. Moreover, the presence of nC60 significantly increased the hatching time of the embryos exposed to atrazine. 14-day exposure of D. magna to nC60 significantly reduced offspring production even at a concentration as low as 0.5 mg/L. This indicates that nC60 is able to reduce the ability of D. magna to produce offspring, and therefore would have an impact on population. For atrazine, no significant difference in offspring production was observed. The nC60 complex with atrazine was also found to significantly decrease the reproduction of D. magna, which was similar to the result of exposure to nC60. These results suggest that nC60 would have potential risks to aquatic organisms. Therefore, much attention should be paid to their associations with other contaminants for fullerene’s risk assessment.  相似文献

采用维生素B1、维生素B5、DHA、胆碱等原辅料,制成新型老年配方液态奶,达到预防阿尔茨海默等疾病的特殊生理功效。通过单因素实验对不同影响因素进行考察,运用正交试验找出较优液态奶配方。结果表明,较佳配方维生素B1添加量为1.7mg/kg,维生素B5添加量为11mg/kg,DHA添加量为300mg/kg,胆碱添加量为700mg/kg。较佳配方生产的产品中,维生素B1含量为1.04mg/kg,维生素B5含量为8.6mg/kg,DHA含量为253mg/kg,胆碱含量为547mg/kg,均能达到有效改善脑力健康的日摄入量。  相似文献

Fullerenen-propylamine platinum, rhodium complex have been prepared from C60 via amination withn-propylamine, then reacting respectively with potassium chloroplatinite or rhodium chloride. The two noble metal complexes can effectively catalyze hydrosiiylation of olefins with triethoxysilane. 研究桂枝茯苓丸在人卵巢癌耐药性裸鼠肿瘤模型中对凋亡的诱导作用,探讨其可能的作用机制。采用实时无标记细胞分析系统检测SKOV3/DDP对顺铂和紫杉醇的耐药性,建立SKOV3/DDP卵巢癌耐药性BALB/c裸鼠肿瘤模型,模型裸鼠随机分为桂枝茯苓丸浓缩液16 g.kg-1.d-1(高)、8 g.kg-1.d-1(中)、4 g.kg-1.d-1(低)剂量组、PI3K抑制剂LY294002组、低剂量桂枝茯苓丸LY294002组(联用组)及对照组,给药10天后剖取肿瘤组织,采用流式细胞术检测肿瘤细胞的凋亡率,采用荧光定量PCR检测肿瘤组织中MTDH、PTEN mRNA的相对表达。结果表明,SKOV3/DDP细胞对顺铂和紫杉醇的耐药倍数分别为4.57和4.36倍。随着桂枝茯苓丸浓缩液剂量的增加,肿瘤细胞的凋亡率逐渐增加,上调PTEN mRNA和下调MTDH mRNA的作用逐渐增强,各组之间差异具有显著性(P < 0.01)。LY294002和低剂量桂枝茯苓丸联用后,抑制凋亡的作用强于二者分别单用的凋亡抑制作用,下调MTDH mRNA和上调PTEN mRNA的作用强于其他各组(P < 0.01)。说明在SKOV3/DDP卵巢癌耐药性BALB/c裸鼠模型中,桂枝茯苓丸能够剂量依赖性地诱导耐药性肿瘤细胞的凋亡,其机制可能与上调PTEN mRNA和下调MTDH mRNA有关。  相似文献

探讨了呋喃二烯的体内外抗肿瘤作用及作用机理.采用MTT法检测呋喃二烯对10种人肿瘤细胞的体外增殖抑制作用,研究发现呋喃二烯对体外培养的多种肿瘤细胞具有很强的抑制和杀伤效应,其IC50在0.98-4.81μg/mL.并对敏感的白血病细胞HL-60进行初步的作用机制研究,其形态学和流式细胞分析试验均表明它能有效诱导细胞调亡.整体动物试验结果可证实呋喃二烯乳注射液能明显延长荷腹水癌S180小鼠的生存期,生命延长率分别为80mg/kg(91.2%),30mg/kg(82.4%),10mg/kg(70.59%).  相似文献

研究艾拉莫德对脂多糖(LPS)诱导的小鼠系统性炎症和多器官损伤的保护作用。小鼠腹腔注射LPS 5 mg/kg建立系统性炎症和多器官损伤模型。试验结果显示,与模型组相比,阳性药强的松组(prednisone)和T614中高剂量组动物死亡率、临床症状、脏器系数及组织病理学结果均显著改善;同时血液生化指标(ALT、AST、ALP、BUN、CREA和UA)显著降低(均P0.01),血液学指标(WBC、RBC、PLT和RET)均与空白对照组动物水平相当。试验结果表明T614对LPS诱导的小鼠系统性炎症和多器官损伤具有保护作用。  相似文献

为研究双酚A(bisphenol-A,BPA)暴露可能对成年小鼠神经行为的影响,将成年小鼠暴露于BPA(0.04,0.4,4和40 mg/(kg·d)12周后,发现BPA(0.04~4 mg/(kg·d))可降低雄鼠血清和脑内的睾酮水平,但对雌鼠血清和脑内的雌二醇水平没有影响.旷场、明暗箱、镜子迷宫和高架十字迷宫等行为模型的检测结果显示,BPA暴露12周对小鼠的活动性和探究意愿没有显著影响,但明显增加雌鼠却减少雄鼠在高架十字迷宫中进入开放臂的次数和开放臂停留时间,表明成年期BPA暴露可加剧雄鼠而减轻雌鼠的焦虑情绪.这些结果提示,长期BPA暴露可性别特异性地影响成年小鼠的焦虑状态,脑内性激素水平的改变可能与此有关.  相似文献
