通过定点突变对麦芽糖诱导型启动子Pglv-M1进行改造,提高启动子启动能力。分析Pglv-M1的Sextama-35区与Pri-bmow-10区,通过对这些区域模拟随机突变,软件打分后获得最高分的新片段。将新启动子与载体p HCMC04-sva连接,并在枯草芽孢杆菌Bacillus subtilis 1A857中诱导表达,每12 h取粗酶液进行酶活测定。研究结果表明:除突变启动子Pglv-35以外,其余突变体启动效果均有所下降,有突变体构成的表达载体Pglv-35-pHCMC04-sva诱导表达后,得到的单位粗酶活力最高点出现在36 h处,较突变前提高1.31倍。PglvM1作为诱导型启动子Pglv的改造产物,其各个区域的相互影响较为紧密。通过对启动子关键区域单独或同时突变,仅获得一组启动能力有所提高的启动子Pglv-35,这为今后针对启动子的思考、研究方向提供参考。  相似文献

MYB转录因子在调控植物生长发育和逆境响应方面发挥着重要的作用.通过克隆获得了核桃的1条R1-MYB类转录因子EFM基因(命名为JrEFM1),利用生物信息学和实时荧光定量RT-PCR(RT-qPCR)技术,分析在不同非生物及植物激素处理下JrEFM1的表达规律,探究了JrEFM1的基本生物学功能.结果显示,JrEFM1的编码区长为1 320 bp,编码蛋白含439个氨基酸,分子量为48.322 KDa,理论等电点为9.26.与葡萄、番薯等具有较近的进化关系.其启动子包含干旱胁迫(MBS)、热激响应(HSE)、水杨酸(SA)、玉米素(O2-site)和赤霉素响应(GARE-motif)等相关元件.对JrEFM1在干旱,冷害,热激,ABA,JA,SA处理下的表达情况进行分析,发现JrEFM1可被这些处理不同程度地诱导,同时根和叶表现出不同的转录水平.表明JrEFM1可响应逆境胁迫,并与激素信号通路相关;JrEFM1可作为核桃逆境响应机制研究及抗逆育种的优良候选基因.  相似文献

Sterile and semi-fertile F1 plants were obtained by intergeneric sexual hybridization between paternal Brassica oleracea var. alboglabra (genome CC, 2n=18) and maternal Sinapis alba (genome SS, 2n=24), BC1 plants were obtained by backcrossing between paternal B. oleracea and maternal semi-fertile F1 plants. Genomic in situ hybridization (GISH) combined with dual-colour fluorescence in situ hybridization (dcFISH) showed that sterile F1 plants contained 21 chromosomes consisting of one B. oleracea chromosome set and one S. alba chromosome set, belonging to expected hybrids, and semi-fertile F1 plants contained 30 chromosomes consisting of two B. oleracea chromosome sets and one S. alba chromosome set. It is obvious that the semi-fertile F1 plants belong to unexpected hybrids. 1―3 trivalents were detected at meiotic metaphase I of semi-fertile F1 pollen mother cells (PMCs). Different separation ratios of S chromosomes were detected at anaphase I. A monosomic alien addition line (MAAL) was identified by GISH-dcFISH from BC1 plants; it contained 19 chromosomes consisting of 18 C chromosomes and 1 S chromosome. At meiotic metaphase I, 9 divalents from B. oleracea and one univalent from S. alba could be detected. Sometimes, one putative C-S trivalent could also be detected. The achievement of B. oleracea-S. alba monosomic alien addition lines lays a foundation for gene introgression, location and cloning.  相似文献

Transgenic tobacco plants carrying CrylAc, Crylle or both genes were obtained. In the leaves of transgenic plants carrying both genes, the contents of CrylAc and Crylle proteins were 0.173% and 0.131% of the total proteins, respectively. CrylAc protein content was 0.182 % and Cry1 le protein content was 0.124% of the total proteins in the leaves of transgenic plants containing only one Bt gene. Fresh leaves of transgenic tobacco and wild-type plants were used for the insect bioassay against wild-type and Cry1Ac-resistant cotton bollworm (Helicoverpa armigera). The bioassay results showed that transgenic plants carrying both genes were significantly more toxic to wild-type and CrylAc-resistant cotton bollworm than those carrying CrylAc or Crylle alone. This study indicates that the higher toxicity of transgenic tobacco plants carrying both genes is caused by the cooperative function of both Bt proteins, thus providing a potential way to delay the development of insect resistance to transgenic crops.  相似文献

以NCBI中收录的不同物种ISL1蛋白的相关序列信息为基础,进行生物信息学分析和功能预测。其中大鼠的ISL1基因长12307 bp,编码349个氨基酸。所编码蛋白质不含信号肽,可能是存在于内质网的亲水性蛋白。结构预测分析发现ISL1蛋白二级结构存在较多无规则卷曲和双锌指的空间结构,证实其发挥转录因子功能的结构基础。同源性比对分析显示,大鼠ISL1基因编码蛋白与人、奶牛、小鼠、金仓鼠、灰狼的ISL1蛋白高度同源,达到100%;与原鸡、斑马鱼、非洲爪蟾同源性也较高,分别达到99%、98%及97%,这说明了ISL1蛋白序列的稳定性和保守性。通过对大鼠ISL1蛋白的生物信息学分析和功能预测,为进一步探讨其分子调控机制提供重要的参考依据。  相似文献

In order to identify the genes associated with glioblastoma differentiation, some ESTs, expressed differentially in the control cell and the differentiated human glioblastoma cell line BT-325 induced by the all-trans retinoid acid, have been isolated by the method of DDRT-PCR. Of the 46 ESTs sequenced, 19 are from new genes. A full-length 1 535-bp cDNA, termed geneGDR1, has been isolated from the human cDNA library using the probe designed according to one of the novel ESTs, HGBB098. The open reading frame ofGDR1 gene encodes a putative protein containing 334 amino acid residues. Blast against the current GenBank DNA and protein sequence database did not reveal significant homology with any known proteins. RT-PCR shows thatGDR1 mRNA level increased in the differentiated BT-325 cells after being treated with RA. The different expression patterns ofGDR1 mRNA in human tissues have been detected through the multiple tissue Northern blot hybridization.  相似文献

Mitochondrial cytochromeb genes of about 450 bp fragments from 3 proturan species, 5 collembolan species and 2 dipluran species have been sequenced. The number of nucleotide substitutions and Kimura 2-parameter distances have also been calculated, and a series of molecular phylogenetic trees reconstructed by using parsimony and distance methods. The proturan, collembolan and dipluran species have evolved monophyletic groups. The results suggest that Protura and Collembola are sister groups, while Diplura is more or less demonstrating a closer phylogenetic relationship to the pterygotan insects. The phylogeny and their systematic position of protura and other groups are also discussed.  相似文献

Common carpCyprinus carpio genomic DNA repetitive sequence CR1 has been DIG-labeled and hybridizedin situ against chromosomes of red common carp (Cyprinus carpio L. Xingguo red var. ). It is found that the repetitive sequence CR1 is mainly localized at the centromeric regions of chromosomes of the red common carp. The application of the chromosomalin situ hybridization technique on fish and the relationship between CR1 repetitive sequence distribution and its function have been discussed.  相似文献

研究Tup1基因对酿酒酵母(Saccharomyces cerevisiae)细胞耐高糖性状的影响。利用Cre/loxP系统对单倍体细胞中的转录因子Tup1基因进行敲除,单倍体细胞复倍后研究基因缺失菌株的性状变化。结果表明,Tup1基因缺失虽然减弱了菌株的呼吸能力,但却增强了菌株在高浓度葡萄糖培养基中的生长和发酵能力。Tup1基因可能通过调节酿酒酵母细胞呼吸强度来调控细胞的高糖应激反应。  相似文献

Two set ofAndrias davidanus fertilized eggs and its seedlings were got by means of artificial reproduction. The sections of the embryo ofAndrias davidianus were made by histological method. The expression of homologous sequences of Quox-1 gene has been studied by immunohistochemical method with special antibody. Results show that the expression of homologous sequences of Quox-1 gene was detected in the epidermis of blastula and gastula. 将细胞色素P450 55a1基因克隆到镰刀菌素植物超表达载体pCAMBIA1302中,构建了pCAMBIA1302-cyp55a1-gfp植物超表达载体,以水稻日本晴为遗传转化的受体对象,通过农杆菌介导侵染方法进行了遗传转化.结果表明:成功构建了细胞色素P450 55a1基因超表达载体,获得了多个细胞色素P450 55a1基因超表达的阳性植株,并以RT-PCR技术分析了阳性植株中细胞色素P450 55a1基因的表达水平.  相似文献

OsNHX1 gene (Na+/H+ antiporter gene ofOryza sativa L.) was introduced into Poplar 84K withAgrobacterium tumefaciens- mediated transformation. PCR, Southern and Northern blot analysis showed thatOsNHX1 gene was incorporated successfully into the genome of Poplar 84K and expressed in these transgenic plants. Salt tolerance test showed that three lines of transgenic plants grew normally in the presence of 200 mmol/L NaCl, while the Na+ content in the leaves of the transgenic plants grown at 200 mmol/L NaCl was significantly higher than that in plants grown at 0 mmol/L NaCl. The osmotic potential in the transgenic plants with high salinity treatment was lower than that of control plants. Our results demonstrate the potential use of these transgenic plants for agricultural use in saline soils.  相似文献

以非洲菊(Gerbera jamesonii)切花为材料,采用四因素[1-甲基环丙烯(1-MCP)、柠檬酸(CA)、蔗糖、 8-羟基喹啉(8-HQ)]三水平的正交试验设计,通过比较各项形态指标和生理指标的变化情况,探讨了各配方的保鲜效果.结果表明, 100 mg/L CA+12 mg/L 1-MCP+6%蔗糖+300 mg/L 8-HQ和200 mg/L CA+10 mg/L 1-MCP+6%蔗糖+200 mg/L 8-HQ保鲜液处理,能增加鲜质量、增大花径、增加花瓣蛋白质质量比;降低花瓣丙二醛含量,延缓花瓣细胞膜透性的增加,且均比CK组延长了其3d瓶插寿命;提高了切花的观赏品质.  相似文献

The cry1Ah gene was one of novel insecticidal genes cloned from Bacillus thuringiensis isolate BT8. Two plant expression vectors containing cry1Ah gene were constructed. The first intron of maize ubiqutinl gene was inserted between the maize Ubiquitin promoter and cry1Ah gene in one of the plant expressing vectors (pUUOAH). The two vectors were introduced into maize immature embryonic calli by microprojectile bombardment, and the reproductively plants were acquired. PCR and Southern blot analysis showed that foreign genes had been integrated into maize genome and inherited to the next generation stably. The ELISA assay to T1 and T2 generation plants showed that the expression of CrylAh protein in the construct containing the ubil intron (pUUOAH) was 20% higher than that of the intronless construct (pUOAH). Bioassay results showed that the transgenic maize harboring cry1Ah gene had high resistance to the Asian corn borers and the insecticidal activity of the transgenic maize containing the ubil intron was higher than that of the intronless construct. These results indicated that the maize ubil intron can enhance the expression of the Bt cry1Ah gene in transgenic maize efficiently  相似文献
