The relationship between juvenile hormone and ecdysteroid titers during the early stage of the last larval instar of the silkworm,Bombyx mori, was examined in this study. When larvae were fed 20-hydroxyecdysone-supplemented mulberry leaves throughout the last larval instar, 100% underwent supernumerary larval molting instead of metamorphosis. The application of juvenile hormone mimic during the early last instar did not induce supernumerary larval molting, but did delay metamorphosis. Temporal and quantitative ecdysteroid titer measurements revealed that in normal larvae the titers maintained very low levels (3–12 ng/ml) during the early stage of the last instar; however, in 20-hydroxyecdysone-treated larvae, levels ranging from 24 to 45 ng/ml were detected, and a major peak (246 ng/ml) was observed on day 6. These results show that very low ecdysteroid titer levels during the early stage of the last larval instar may play an important role in initiating decreases in juvenile hormone titers as well as in directing metamorphosis.  相似文献

Controlled experiments on the metamorphosis of marine invertebrate larvae require artificial inducers. These inducers can be used for studying the involvement of known signal transduction pathways in settlement and metamorphosis. The ability of the tumor-promoting phorbol ester TPA (12-O-tetradecanoylphorbol-13-acetate) to induce metamorphosis in planulae of the Red Sea soft coral speciesHeteroxenia fuscescens, Xenia umbellata, Dendronephthya hemprichii, Litophyton arboreum andParerythropodium fulvum fulvum, and the stony coralStylophora pistillata, was examined by using various concentrations of TPA. The chemical induced metamorphosis in all six species. The effect was unspecific and concentration-related. For all the corals except forX. umbellata the highest mean percentages of metamorphosis were obtained with 8.1×10–7–10–9 M TPA. ForX. umbellata, the percentage of metamorphosis was lower, and was obtained within a wider TPA concentration range. The present results, along with previous studies on Hydrozoa and Scyphozoa, demonstrate that TPA is the first common artificial inducer for these classes of Cnidaria. TPA is known to activate the enzyme protein kinase C (PKC) and therefore plays an important role in studying the phosphatidylinositol signal transduction system. Evidence for the involvement of this pathway in triggering metamorphosis has already been reported for Hydrozoa and Scyphozoa. Our results suggest that PKC is also involved in initiating metamorphosis in Anthozoa.  相似文献

Highly active metabolites have been detected in the hemolymph of the lepidopteranSpodoptera exigua infected with the mycopathogen,Beauveria bassiana. A combination of phenyl sepharose and CM ion exchange chromatography was utilized to extract the active metabolites from infected hemolymph samples. The active in vivo metabolites, having a molecular mass greater than 10 KDa, were thermolabile and were inactivated by proteinase K. These metabolites were characterized by their ability to disrupt metamorphosis, killing treated larvae at the wandering or pupal stage. Additionally, injection ofS. exigua larvae with active samples caused a reduction in the number of filopodial-producing hemocytes. The biological activities and biochemical properties suggest that novel compounds are produced duringB. bassiana mycosis.  相似文献

Two pteridine-containing bromophysostigmine alkaloids, urochordamine A and B, which were isolated from ascidians as larval metamorphosis promoters, were converted to more polar compounds, urochordamine A and B, respectively, when left standing in protic solvents. These four compounds promoted larval metamorphosis of the ascidianHalocynthia roretzi in the order A>A>B>B, and induced metamorphosis of the pediveliger larvae of the musselMytilus edulis galloprovincialis.  相似文献

Summary The problem of arrested or delayed development was examined in the sugar cane borer,Diatraea saccharalis. It was found that the insect can either enter diapause or exhibit a period of delayed metamorphosis according to the photoperiod conditions prevailing. We have observed the development characteristics ofD. saccharalis and conclude that a distinction should be made between a delayed metamorphosis phase and a diapause stage.This work was supported by a grant from NSERC.  相似文献

Summary Macroscopic anatomical investigations on the brain of the Blue Whale (Sibbaldus musculus Linnaeus, total brain weight without dura: 6500 g), show that this species of cetacea has a degree of cerebralization between that of the primitive Southern Right Whale (Eubalaena australis Desmoulins) and that of the Humpback Whale (Megaptera novaeangliae Borowski). Of the Balaenopteridae examined to date, the Blue Whale has the lowest cerebralization.

Laboratory experiments were conducted to induce neoteny inHynobius retardatus, which had been reported to propagate in larval forms like axolotl. A large number of newly hatched larvae were reared in an aqueous solution of thiourea (TU) and sodium perchlorate (SPC) in order to arrest the metamorphosis. Gonadal development in the metamophosis-arrested larvae was compared with that in normally metamorphosing and metamorphosed controls. Metamorphosis-arrested male larvae produced morphologically mature spermatozoa approximately 4 months after hatching, when the gonads in the controls began to differentiate into testes, or to show the premeiotic proliferation of germ cells. Possible endocrine controls of these phenomena are discussed.  相似文献

Summary Lactate dehydrogenase zymograms of mature testes ofHymenochirus boettgeri show in addition to the five isozymes composed of LDH-A and LDH-B subunits, a second 5-band system which is due to isozymes formed between LDH-A and a third subunit, LDH-C. These testis-specific LDH-C isozymes appear around 6 months after metamorphosis indicating that their expression is correlated with sexual maturity as is generally the case in mammals. This is the first report of a testis-specific LDH isozyme in a lower tetrapod; such isozymes have hitherto only been reported in mammals and in the pigeon.This study was supported by a grant from the Fond National Suisse de la Recherche Scientifique to H.R.K. Summary We examined the role of thyroid hormone in mediating morphological integration between cranial cartilage and bone during anuran metamorphosis. Exogenous T3 applied to premetamorphic tadpoles (Bombina orientalis) via intracranial implants of plastic micropellets precociously induced typical metamorphic changes in both tissues, but also dissociated the relative timing of developmental events between them. Morphological integration between the two primary cranial tissues is achieved in part by each tissue responding independently to endocrine factors and does not reflect a tight developmental coupling between them.  相似文献

Summary Larvae ofSalamandra salamandra quadrivirgata Dürigen and tadpoles ofXenopus laevis Daud, treated with a percutaneous application of thyroxin, show a more intensive calcification of their skeletal system after a precocious metamorphosis. The calcified area of bones is increased; additional centres of calcification can be distinguished in quite a number of other skeletal elements. By thyroxin-soaked agar implants in the hindlimbs ofAmbystoma mexicanum, deposition of calcium salts is furthered in the bones of the leg treated, as well as in the adjacent pelvic girdle, thus demonstrating that the thyroid hormone exerts a direct effect upon calcification. It is supposed that during normal metamorphosis the thyroid gland plays an equal part in the process of calcification in the skeletal system.  相似文献

Summary L-Canavanine, 2-amino-4-(guanidinooxy)butyric acid, and L-arginine incorporation into de novo synthesized proteins was compared in six organisms. Utilizing L-[guanidinooxy14C]canavanine and L-[guanidino14C]arginine at substrate saturation, the canavanine to arginine incorporation ratio was determined in de, novo synthesized proteins.Caryedes brasiliensis andSternechus tuberculatus, canavanine utilizing insects;Canavalia ensiformis, a canavanine storing plant; and to a lesser extentHeliothis virescens, a canavanine resistant insect, failed to accumulate significant canavanyl proteins. By contrast,Manduca sexta, a canavanine-sensitive insect, andGlycine max, a canavanine free plant, readily incorporated canavanine into newly synthesized proteins. This study supports the contention that the incorporation of canavanine into proteins in place of arginine contributes significantly to canavanine's antimetabolic properties.  相似文献

Summary Ecdysteroid titers were determined in the hemolymph of adult freshwater prawn,Macrobrachium rosenbergii, during various stages of the molt cycle, using radioimmunoassay. A comparison was made between hormone levels in males which undergo normal molt and dominant males of the population which periodically commit cheliped autotomy. The latter is a recently discovered phenomenon whereby a male voluntarily sheds off both its long claws at molt, once the ratio between body length and combined claw length reaches a ceiling level of about 13. The components of individual Dufours glands excised fromAnoplolepis custodiens workers were analysed by GC-MS. In addition to then-alkanes andn-alkenes previously reported2 in these glands, primary alcohols (C19-C22), secondary alcohols (C20-C23), 2-ketones (C20-C23) and possibly carboxylate ethyl esters (C19 and C21) were identified as components of these glands. It seems possible that these high-boiling compounds are used by the workers in laying trails on the hot sandy surfaces of their characteristic habitat and in lining of the inner walls of nests, but no standard compounds have been available to us for any behavioral studies.  相似文献

Summary Approximately 180 small and mainly acrocentric chromosomes are present inGeotria australis (Geotriidae) from the southern hemisphere. It has been suggested that a common sex pheromone composition may account for interspecific sexual interactions observed with certain moths in the Arctiidae. In this study, it is demonstrated that the sex pheromones released by females of the Scarlet Tiger Moth,Callimorpha dominula L., and the Cinnabar Moth,Tyria jacobaeae L., have similar activities and elute at the same retention time on analysis by coupled gas chromatography (GC)-electrophysiology with males from each species. Peak enhancement on GC, chiral GC and coupled GC-mass spectrometry using authentic compounds show that the sex pheromone for bothC. dominula andT. jacobaeae is (3Z,6Z,9S,10R)-9,10-epoxyheneicosa-3,6-diene.  相似文献
