The activity of membrane-bound dipeptidyl aminopeptidase IV (EC 3.4.14.5) was found to be markedly reduced in the Fischer 344 rat strain compared with that in the Wistar strain. Analysis of membrane proteins by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Ouchterlony immunodiffusion revealed the specific absence of this enzyme molecule in the Fischer 344 strain. 相似文献

Summary The specific activity of dipeptidyl peptidase IV (DPPIV E.C. 3.4.14.-) in the plasma membrane of Morris hepatoma 9121 or hepatoma 7777 was 3.5% and 2.9%, respectively, of that in the plasma membrane of rat liver. The enzyme activity in the serum of hepatoma-bearing rats was 141% (hepatoma 91219) and 162% (hepatoma 7777) of the normal value. cytochemical investigation showed that the DPP IV activity was almost completely absent from the hepatoma cell plasma membrane and was not sequestered within these cells. Indirect immunofluorescence staining with a polyclonal antibody directed against DPP IV indicated that the loss of activity was due to the absence of DPP IV molecules in the plasma membrane. The possibility that the enzyme is transferred from the membrane into the serum as a result of structural alterations is discussed. 相似文献

Aminopeptidase activities, assayed as arylamidase activities, were investigated in selected tissues of 1, 6, 12 and 24-month-old rats. The enzyme activities were found to have a heterogeneous distribution and age-related changes were observed. The highest levels of soluble arginyl-aminopeptidase activity were detected in brain homogenate at all the studied ages, whereas membrane-bound activity presented the highest levels in brain and kidney in the four ages tested. Aspartyl-aminopeptidase activity was detected mainly in the particulate fraction of kidney at all four ages. In 1, 6 and 12-month-old animals, soluble aspartyl-aminopeptidase activity was also higher in the kidney than in the rest of the tissues, whereas in the group of 2-year-old rats, the highest levels were found in both kidney and liver. Age-related changes were observed in all the studied tissues and for all the assayed enzymatic activities. In general, the maximal levels were detected in both the youngest and the oldest animals, and the minimal ones in 6 and 12-month-old rats. However, in the adrenals, the soluble and membrane-bound arginyl-aminopeptidase activity was higher in 6-month and 2-year-old rats than in 1-month and 12-month-old rats. These changes may reflect the functional status of the susceptible endogenous substrates of aminopeptidases. 相似文献

Summary DPP IV was localized in the meningeal lamellae of the spinal cord sheaths of the rat by light and electron microscopy. A membrane-bound reaction product of DPP IV was found in the internal, intermediate and external meningeal lamellae which delineated the CSF-filled meningeal spaces. The cells of the marginal glia displayed heterogeneous localization of the reaction product for DPP IV. DPP IV distribution in the spinal cord sheaths suggests its possible participation in the interactions of the meningeal cells with the neuropeptides in cerebrospinal fluid. 相似文献

The involvement of the renin-angiotensin system in the regulation of cell proliferation in the rat endometrium 总被引:1,自引:0,他引:1
Oestrogens are known to enhance angiotensin biosynthesis by increasing the elaboration of its precursor, angiotensinogen.
On the other hand, we found that inhibition of angiotensin-converting enzyme (ACE) suppressed the proliferative response of
the rat anterior pituitary gland to oestrogens. To answer the question whether the angiotensin system is involved in the control
of the cell proliferation of the uterine epithelium, the effects of an ACE inhibitor, enalapril maleate, and of angiotensins
II and IV, alone or together with losartan, an antagonist of angiotensin receptor type 1 (AT1), on endometrial epithelial
cell proliferation have been studied. The experiments were performed on ovariectomized female Wistar rats. In the first experiment
the animals were injected with a single dose of oestradiol benzoate or received an injection of solvent only. Half of the
oestrogen-treated rats were injected additionally with enalapril maleate (EN, twice daily). The incorporation of bromodeoxyuridine
(BrDU) into endometrial cell nuclei was used as an index of cell proliferation. It was found that oestradiol alone dramatically
increased the BrDU labelling index (LI) of endometrial cell nuclei, and this effect was partially blocked by the simultaneous
treatment with EN. In the second experiment, the animals were injected intraperitoneally with angiotensin II (AII), angiotensin
IV (AIV) or saline, alone or together with losartan. It was found that AIV induced an increase in the LI in uterine epithelium,
and this effect was not blocked by the simultaneous treatment with losartan. The increase in LI in uterine epithelium was
also observed in the rats treated with AII and with losartan. These findings suggest an involvement of angiotensin IV in the
control of uterine epithelium cell proliferation.

Summary Dopamine beta-hydroxylase (DBH) enzyme activity was associated in rat superior cervical ganglion with tetrameric DBH-A (294,000 D) and dimeric DBH-B (147,000 D) and in rat adrenal gland with DBH-A and a novel molecular form of DBH, defined as DBH-C, with a molecular weight of 125,000 D. Pretreatment of the rats with cycloheximide markedly reduced DBH activity without altering the molecular heterogeneity. 相似文献

Summary We produced a new type of antiserum to thyrotropin-releasing hormone (TRH) in rabbits. The immunogen is TRH-BSA, the production of which is based on the formation of an amide bond using carbodiimide (EDC). The specificity of the antiserum was assessed by enzyme immunoassay (EIA) and immunohistochemistry. When using the anti-TRH serum for immunohistochemistry in rat hypothalamus, new magnocellular groups were detected in the ventrolateral parts of the posterior hypothalamus and the dorsal parts of the third ventricle. Colchicine treatment was found not to be necessary to visualize perikarya containing TRH. 相似文献
