Serum-induced stimulation of snRNA synthesis in mouse 3T3 fibroblasts

Small nuclear RNAs (snRNAs) from quiescent and serum-stimulated 3T3 cultures, labeled with [3H]uridine [( 3H]U), were electrophoresed in polyacrylamide-urea slab gels and revealed by staining with ethidium bromide and by fluorography. Judged by labeling with [3H]U, synthesis of 7S and U1-U6 RNAs was very low or absent in quiescent cultures. The serum-induced transition of 3T3 cells from a resting to a growing state was accompanied by an early, apparently sequential stimulation of snRNA synthesis; stimulated synthesis of 7S, U1, U2, U3, U4 and U6 RNAs coincided in time with serum-induced stimulation of 45S pre-ribosomal RNA (pre-rRNA) and heterogeneous nuclear RNA (hnRNA) synthesis.     

Inhibitory action of bradykinin on release of the adrenergic transmitter in the isolated lapine kidney

Summary In the isolated lapine kidney perfused with tyrode solution and prelabeled with [³H] norepinephrine, bradykinin (10 ng/ml) decreased the overflow of tritium elicited by sympathetic nerve stimulation both in the presence and absence of indomethacin. These observations indicate that bradykinin acts at presynaptic sites reducing release of the adrenergic transmitter in the isolated lapine kidney.This study was supported by American Heart Association Grant 77-803, and U.S. Public Health Service Grant HL-16134 from the National Heart and Lung Institute.     

Binding of [3H]GABA and [3H]muscimol to subcellular particles of a neurone-enriched culture of mouse brain

Summary Binding of [³H]GABA and [³H]muscimol, indicative of GABA-receptors, has been demonstrated in a neurone-enriched culture of embryonic mouse brain using a ligand-binding technique. Evidence is provided for the existence of different populations of GABA-receptors.This study was supported by C.N.R.S., ATP N^o 3356 and by Commissariat à l'Energie Atomique, Departement of Biologie. Mechanism of age-dependent involution in embryonic chick notochords

To study the possible mechanism of the age-dependent involution of the notochord, isolated mesenchymefree notochords of chick embryos were cultured in vitro and compared with their counterparts in vivo. Two different aspects were evaluated: (1) DNA synthesis measured by [³H]thymidine incorporation and visualized by autoradiography and (2) cell death quantified by counting the number of pyknotic nuclei. The results demonstrate that [³H]thymidine uptake by notochords shows an age-dependent decrease in vitro as well as in vivo. The number of [³H]thymidine-labelled notochord cells, however, is higher in vitro than in vivo. At the same time, there is an age-dependent increase in pyknosis in the notochord in vivo and in vitro. So, during the aging process, the number of both pyknotic nuclei and of [³H]thymidine-labelled nuclei suggest a high turnover of notochord cells in vitro. From these results, we can conclude that the process of involution in aging notochord seems to be controlled by a programmed intrinsic process, which might be influenced partially by the microenvironment in vivo.     

The role of glycine in the biosynthesis of a terpene

Zusammenfassung Mit doppelt markiertem Glycin konnte gezeigt werden, dass das Kohlenstoffatom der Methylgruppe viel besser als das Kohlenstoffatom der Karboxylgruppe in die Isoprengruppe des Ophiobolins B ausCochiobolus miyabeanus eingebaut wird. Die Radioaktivität wird in Ophiobolin B eingebaut, wenn die folgenden Verbindungen verwendet werden: [3-¹⁴C]-Serin, [¹⁴C-Methyl]-Sarcosin, [¹⁴C-Methyl]-Methionin, und [¹⁴C] Formate.

Multiple daily amphetamine administration decreases both [3H]agoinst and [3]antagonist dopamine receptor binding

Summary Multiple daily amphetamine injections in rats decreased both [³H]agonist as well as [³H]antagonist striatal dopamine receptor binding. Concurrently, these animals exhibited a decrease in striatal dopamine concentration and, paradoxically, an enhancement of behavioral responsivity.This study was supported by PHS grant MH32990 to I.C. and DA0156805 to D.S.I. Rat testes interstitial cell nuclei exhibit three distinct receptors for retinoic acid

Summary Purified nuclei from rat testes interstitial cells were incubated with an equimolar complex of [³H]retinoic acid and purified cellular retinoic acid-binding protein (cRABP) and with ATP. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and radiofluorographic analysis of the nuclear fractions indicated the presence of 3 highly labeled receptors for retinoic acid which were distinct from cRABP. These data demonstrate that retinoic acid binds to 3 novel nuclear acceptors of which cRABP does not appear to be a part.Scientific contribution No. 1021, The Storrs Agricultural Experiment Station, The University of Connecticut, Storrs (Connecticut 06268, USA).     

Autoradiographic studies on RNA synthesis and transport in the Salivary gland ofLygaeus sp. (Hemiptera-Lygaeidae)

Summary Autoradiographic studies using³H-uridine in the salivary gland of adultLygaeus sp. were carried out. The gland cell nuclei, particularly the multiple nucleoli, are the sites of incorporation of the label exhibiting RNA synthesis. The labelled molecules (RNA) are transported to the cell cytoplasm and then into the gland lumen in which no turnover of the radioactivity is observed.I am grateful to my teacher, Prof. P. S. Drosophila melanogaster does not dealkylate [14C]sitosterol

Summary Drosophila melanogaster was unable to dealkylate and convert [¹⁴C]sitosterol to cholesterol and no evidence was found for conversion of [¹⁴C]desmosterol to cholesterol. Therefore,D. melanogaster is incapable of dealkylating and converting C₂₈ and C₂₉ phytosterols to cholesterol.The authors thank O. J. Duncan III, and H. S. Symonds for technical assistance. Inhibition by tetanus and botulinum A toxin of the release of [3H]noradrenaline and [3H]GABA from rat brain homogenate

Summary Rat brain homogenate was preloaded with [³H]noradrenaline or [³H]GABA and stimulated with high K⁺. Tetanus toxin and botulinum A neurotoxin partially prevent the evoked [³H]noradrenaline release in the same range of toxin concentrations starting below 10^–10M. In contrast, release of -amino butyric acid (GABA) is much more sensitive to tetanus than to botulinum A toxin.     

Increase of interferon antiviral activity by exogenous cyclic adenosine-3′∶5′-monophosphate (cAMP)

Summary Some effects of cAMP on replication of Semliki Forest Virus in chick embryo fibroblast cell cultures are described. Depending on concentration, the incorporation of [³H]-uridine into viral RNA or the formation of plaqueforming units is inhibited; the highest concentration tested was 8 mM. Cyclic AMP has an effect of its own and increases the Interferon action in the lower concentration ranges of Interferon (up to 1 unit/ml). The effect of cyclic AMP is fast, needs no induction and is also visible in late phases of viral replication. However, these experiments do not establish a causal relation between cAMP and Interferon.Work supported by the Swiss National Science Foundation, grants 3.1050 and 3.540.     

Autoradiographic localization of testosterone-3H in the female rat brain and estradiol-3H in the male rat brain

Zusammenfassung Eine halbe Stunde nach der i.v. Injektion wurde Testosterone-³H von den Nerven- und Glialzellen innerhalb des Gehirns der weiblichen Ratte und Estradiol-³H innerhalb des Gehirns der männlichen Ratte aufgenommen. Zwei Stunden nach der Injektion war die Aufnahme in hohem Masse nur noch in gewissen Teilen des hypothalamisch-limbischen Systems vorhanden.

Brain factor fromGalleria mellonella (Lepidoptera) stimulating silk gland activity

Brain extracts from day 1–4 last instar larvae ofGalleria mellonella (Lepidoptera) stimulate RNA synthesis in cultured silk glands from day 3 last instar larvae. When the fibroin-synthesizing posterior parts of silk glands were incubated for 3 h in vitro in the presence of brain extract (0.1 brain equivalent), [³H]-uridine incorporation into RNA was stimulated more than twofold. The stimulating effect of brain extract showed a dose response relationship. It is suggested that the heat-resistant and protease-sensitive brain factor is a peptide.     

Cytogenetic changes induced by 1-(N1-methylhydrazinomethyl)-N-isopropyl benzamide in Ehrlich ascites tumor cells

Zusammenfassung Trägermäuse von Ehrlich-Ascites-Tumoren zeigen nach Behandlung mit 1-(N¹-Methylhydrazinomethyl)-N-Isopropyl-Benzamid (MIH) eine sofortige Zellteilungshemmung. Diese lässt sich nach s.c. MIH-Injektion von 200 mg/kg bis auf 72 h ausdehnen. Der MIH-Behandlung widerstehende Zellen (nach 5 Tagen) waren durch 2 neue metazentrische Chromosomen ausgezeichnet.

On the synthesis of serine and homoserine samples asymmetrically labelled with tritium and deuterium in the hydroxymethylene group

Summary (1 R) [1-³H,²H₁] 3-Phenylpropanol, the key intermediate in the synthesis of (4 R) [4-³H,²H₁] D, L-homoserine and of the (4 S)-isomer, is obtained from (1 S) [1-²H₁] 3-phenylpropanol and (1 RS) [1-³H] ethanol upon incubation with yeast alcohol dehydrogenase and NAD⁺; under similar conditions 2-phenylethanol undergoes very small exchange with [1-²H₂] ethanol.     

The role of glycine in the biosynthesis of steroids

Zusammenfassung Während der Biosynthese von Cholesterol mit homogenisierter Rattenleber wird [2-¹⁴C] des Glycins viel besser eingebaut als [1-¹⁴C].Saccharomyces cerevisiae produziert radioaktives Squelen (ausser Ergosterol mit Radioaktivität des Ringsystems) mit [2-¹⁴C] Glycin und mit [3-¹⁴C] Serin, aber nicht mit [1-¹⁴C] Glycin.

9,10-Dihydroergotamine: Production of antibodies and radioimmunoassay

Summary Antibodies against 9,10-dihydroergotamine (DHE) were produced by immunizing rabbits with a conjugate of 6-nor-6-carboxymethyl-9,10-dihydroergotamine and bovine serum albumin. A highly specific and sensitive radioimmunoassay for DHE has been developed.We are greatly indebted to Dr.T. Fehr for the synthesis of 6-nor-6-carboxymethyl-9,10-dihydroergotamine, and to Dr.E. Schreier for providing us with 9,10-dihydroergotamine-[13-³H]-base of high specific activity.     

Interaction of galectin-1 with caveolae induces mouse embryonic stem cell proliferation through the Src, ERas, Akt and mTOR signaling pathways

Galectins have the potential to provide a promising alternative for unveiling the complexity of embryonic stem (ES) cell self-renewal, although the mechanism by which galectins maintain ES cell self-renewal has yet to be identified. Galectin-1 increased [³H]-thymidine incorporation as well as cyclin expression and decreased p27^kip1 expression. Src and caveolin-1 phosphorylation was increased by galectin-1, and phospho-caveolin-1 was inhibited by PP2. In addition, inhibition of caveolin-1 by small interfering RNA and methyl-β-cyclodextrin (Mβ-CD) decreased galectin-1-induced cyclin expression and [³H]-thymidine incorporation. Galectin-1 caused Akt and mTOR phosphorylation, which is involved in cyclin expression. Galectin-1-induced phospho-Akt and -mTOR was inhibited by PP2, ERas siRNA, caveolin-1 siRNA and Mβ-CD. Furthermore, mTOR phosphorylation was decreased by LY294002 and Akt inhibitor. Galectin-1-induced increase in cyclin expression and decrease in p27^kip1 was blocked by Akt inhibitor and rapamycin. In conclusion, galectin-1 increased DNA synthesis in mouse ES cells via Src, caveolin-1 Akt, and mTOR signaling pathways. The effect of the phytoalexins,lubimin, (−)-maackiain,pinosylvin, and the related compounds dehydroloroglossol and hordatine M on human lymphoblastoid cell lines

Summary We have tested the effect of the phytoalexins lubimin, (–)-maackiain and pinosylvin and the related compounds dehydroloroglossol and hordatine M on the growth of the human lymphoblastoid cell lines Molt and Raji. (–)-maackiain, pinosylvin and dehydroloroglossol showed significant growth inhibitory action on the cells. Suppression of [³H] thymidine and [³H] leucine uptake was tested and noted in pinosylvin and dehydroloroglossol. The phytoalexins and related compounds are widespread in plants and provide a potential source of antineoplastic substances.We would like to acknowledge the assistance of J. Significance of nitrate reductase in nitrate assimilation in free-livingRhizobium cultures

Summary An in situ method for measuring nitrate reductase (NR) in free-livingRhizobium cultures has been developed. Arginine, threonine, valine, glutamic acid, tyrosine and tryptophan were found to promote NR synthesis.
