Structure and assembly of the 20S proteasome   总被引:3,自引:0,他引:3  
The barrel-shaped 20S proteasome is one of the two components of a larger 26S particle, the multicatalytic 2000-kDa protease complex. The proteolytic sites are located in the inner chamber of the 20S particle and are only accessible via narrow entrances. This paper reviews the current knowledge concerning proteasome formation, proteolytic activities, structural aspects and assembly. Eukaryotic proteasomes are made up by four rings each of which contains seven different subunits occurring at fixed positions. While the outer rings contain α-type subunits, the inner ones comprise β-type subunits. The current assembly model for eukaryotic 20S proteasomes is based upon the detection of 13S and 16S intermediates, respectively, in addition to previous findings with archaebacterial and eubacterial proteasome assembly. The available data suggest a cooperative assembly of the α-type and β-type subunits into half proteasome-like complexes followed by dimerization into proteasomes. During or after dimerization of half proteasomes, the β-type subunits are processed. The prosequence of the β-type subunits is essential for the assembly process and prevents protease activity of immature proteasomes.  相似文献

Résumé Nous avons étudié la distribution du RNA 5S dans le foie du serpent sud-américainCrotalus durissus terrificus. Nos résultats ont montré que le RNA 5S de ce reptile est associé à la sous-unité ribosomique 60 S. D'autre part, la mobilité electrophorétique de ce RNA de faible poids moléculaire est la même que celle d'Escherichia coli et de pupe d'Apis mellifera L.

Summary A structure similar in topography and morphology to the carotid labyrinth of amphibians has been found in 4 catfishes, namely,Clarias batrachus, Heteropneustes fossilis, Rita rita andMystus seenghala. The present finding of the presence of the carotid labyrinth in fishes is the first report, and the results are discussed in the light of the amphibious habit of these catfishes.We are grateful to Prof. U.S. Srivastava for providing laboratory facilities and for encouragement. Summary The activity of the pure 10R (=natural) and 10S enantiomers of juvenile hormone III (JH III) was determined in 3 different bioassays, and the relative binding affinity of the 2 enantiomers to the haemolymph JH-binding protein of the cockroachNauphoeta cinerea was measured. In theGalleria wax test, a local morphogenetic assay, the 10R enantiomer was 5240 times more active than, the 10S enantiomer, 1Galleria unit corresponding to 0.42 pg of 10R-JH III as compared to 2.2 ng for 10S-JH III. In a systemic morphogenetic assay with the cockroachNauphoeta cinerea 380 times less 10R enantiomer was necessary in order to induce detectable juvenilisation (58 ng 10R and 22 g 10S) and in a systemic gonadotropic assay withNauphoeta cinerea 255 times less 10R was needed to induce vitellogenin synthesis in 50% of the insects (6.7 ng 10R and 1710 ng 10S). In the JH-binding protein assay 10R-JH III had an affinity for the JH-binding protein (lipophorin) which was approximately 46 times higher than that of 10S-JH III.  相似文献

DNA probes containing the repeated rDNA region ofDrosophila melanogaster (coding for e.g. 28S and 18S rRNA) hybridized in situ to distinct regions of two heterologous mitotic chromosomes of the honeybee, identifying the nucleolus organizing regions (NORs). The method allows a rapid establishment of a physical map ofApis mellifera using other DNA probes ofDrosophila. This is the first report on well-defined chromosomal markers in the honeybee.  相似文献

Summary Haemolymph ofPalamnaeus bengalensis contains normally 12 and during moulting up to 15 free amino acids, being in maximal number at the end of ecdysis. Aminoacidaemia is most pronounced during the pharate stage. Tyrosine appeared for tanning of the cuticle. Taurine and methionine were not present.Acknowledgment. The author is thankful to Dr.S. C. Summary Ontogeny of light response in the compound eyes of the fly,Sarcophaga bullata was investigated. Summary Ectopic eyes were produced on the fleshfly,Sarcophaga bullata by transplantation of imaginal eye discs. Summary Nuclear polyhedrosis virus (NPV) material of the armywormMythimna (Pseudaletia) separata was administered in the form of PIBs and free virus rods to the larvae of silkwormBombyx mori. Routes used for administration were topical, intrahaemocoelic and oral. The larvae were treated with following concentrations: 10×105 PIBs/L, 10×106 PIBs/L, 10×107 PIBs/L, 10×108 PIBs/L. In all the 3 experiments, the larvae showed neither any signs and symptoms, nor mortality due to polyhedrosis. Thus it appears that the NPV ofM. (P.) separata is safe for the silkwormB. mori.Acknowledgments. Summary The metabolism of the trichothecene 4-monoacetoxyscirpenol by intact gut tissue was determined in the fungus-feeding Nitidulid,Carpophilus hemipterus (L.) and the non-fungus-feeding caterpillars, the fall armyworm,Spodoptera frugiperda (J. E. Smith) and the corn earworm,Heliothis zea (Boddie). The primary metabolite was the hydrolysis product scirpentriol. The amount of metabolism by theC. hemipterus larvae was ca 10 times that of the caterpillars on a per mg protein basis, suggesting metabolic adaptation for feeding on fungi that may contain mycotoxins.Acknowledgment. Summary Allantoinase, an enzyme in the purine-urea cycle, was found inEudistylia vancouveri (Polychaeta). The enzyme had a pH optimum at 7.6 TheK m was 0.012M allantoin, and the Arrhenius energy of activation was 12.6 to 14.6 kcal/mol.Supported by the U. S. Summary Males ofDiprion similis respond to both the (+)-2R,3R,7R and (–)-2S,3S,7S enantiomers of its sex pheromone, erythro-3,7-dimethyl pentadecan-2-yl propionate. Summary The attraction ofS. scolytus to the components of Multilure is described. 4-Methyl-3-heptanol is an attractant which is synergised bya-cubebene. Multistriatin appears to be an inhibitor. A combination of 4-methyl-3-heptanol and cubeb oil is more effective as a bait forS. scolytus than Multilure.We thank Mr A. K. Hughes and Mr J. Summary Resilin is present in the endocuticle of the flexible abdominal intersegmental cuticle of the, physogastric queen of the termite,Odontotermes obesus (Rambur). Summary Males ofPodisus fretus (Hemiptera: Pentatomidae) release a long-range attractant pheromone containing linalool (49.0%), (E)-2-hexenal (34.5%), benzyl alcohol (12.0%), nerolidol (2.0%),-terpineol (1.1%), and traces of several other compounds. The eastern yellowjacket,Vespula maculifrons (Hymenoptera: Vespidae), is attracted to artificial pheromones forP. fretus and for the sympatric species,Podisus maculiventris.The authors thank Mr T.J. Summary DNA probes to identify the genes encoding toxins in enteric bacteria have been developed. Use of these probes reduces the number of animals required for toxicity testing, as suspect bacteria can be directly tested for the presence of toxin. We have augmented the gene probes available by developing probes against theEscherichia coli enterotoxin LTII and shiga toxin fromShigella dysenteriae 1.The LTII gene fromE. coli 357900 was identified and characterised and a suitable internal probe was obtained. The LTII gene was found not to be common among enterobacteriae from various geographical locations. Isolates predominately of animal origin from Nigeria and Thailand hybridized with the probe.The shiga toxin gene was isolated fromS. dysenteriae 1 by a combination of in vivo and in vitro methods. An internal probe was identified and used against different serogroups ofShigella andE. coli isolated. The probe was found to hybridize withS. dysenteriae 1 isolates and also someS. flexneri andS. sonnei strains. Representatives were tested for toxin production and found to produce toxin at low levels.  相似文献

Summary The volatile oil present in the secretion of a scale insect,Ceroplastes ceriferus Anderson (fam. Coccidae), has been found to consist of a number of sesquiterpenes which are enantiomeric with respect to those fromC. rubens Maskell, which infests the same host tree.
