Beta-blocking drugs and human platelet aggregation in vitro

Riassunto L'effeto inibente di farmaci -bloccanti sull'aggregazione di piastrine umane indotta in vitro mediante ADP è risultato proporzionale alle dosi dei farmaci impiegati. La seguente scala gerarchica di potenza è disposta in ordine decrescente di attività: pindolol, K 4423, propranolol, INPEA.     

In vitro enhancement of human platelet aggregation by somatostatin

Summary Very low concentrations of somatostatin (S-14) strongly potentiate the in vitro aggregation induced by collagen, ristocetin and arachidonic acid, but not that induced by ADP or epinephrine, in both human platelet rich plasmas and gel-filtered platelet preparations. Desensitization phenomena may be induced either by repeated addition of S-14 or long lasting contact between S-14 and platelets.

The effect of several diuretics on adenosine-diphosphate induced platelet aggregation in vitro

Résumé L'agrégation des plaquettes, induite par l'ADP exogène, fut observée par turbidimétrie en mesurant les variations de la densité optique. Le méralluride, le méersalyl et le PCMB préincubés avec les plaquettes de rat et de cobaye préviennent l'agrégation, tandis que l'acide éthacrynique n'a aucun effet. Par contre, le chlorire méthylique de mercure la provoque en l'absence d'ADP.     

ADP-induced platelet aggregation in vivo after exclusion of different circulatory districts.

The effect of the ADP infusion on the basal platelet count was studied in controls and in rats submitted to the exclusion of the following circulatory districts: splenic, renal, cerebral and of a hind-limb. After these exclusions the ADP-induced thrombocytopenia was less marked than the controls.     

Effect of sympathectomy on platelet aggregation and blood coagulation in rats

Summary It is shown that the ability of blood platelets to aggregate in partially and completely sympathectomized rats is significantly lower than in intact animals. The blood clotting system of sympathectomized rats is hyperactive. The sympathectomy-provoked changes may be due to the increased content of adrenaline in the blood.     

Defective ristocetin and bovine factor VIII-induced platelet aggregation in normal rats

Riassunto Le piastrine di 3 diversi ceppi di ratti, contrariamente alle piastrine umane e di cavia, non sono aggregate (sia in vitro che in vivo) nè dalla ristocetine nè dal fattore von Willebrand bovino, porcino o umano, quest'ultimo reso aggregante previo trattamento enzimatico. Tale insensibilità, di cui si è potuta escludere l'origine plasmatica, potrebbe derivare dall'assenza, sulle piastrine di ratto, di un recettore per il fattore von Willebrand (acido sialico?); potrebbe essere cosi spiegata anche la mancata risposta delle piastrine alla ristocetina. Questi risultati suggeriscono che le piastrine di ratto possono essere un utile modello sperimentale per lo studio della sindrome di Bernard-Soulier nell'uomo.

The release of platelet factor 4 during platelet aggregation and the possible significance of this reaction in hemostasis

Résumé Nous avons constaté que le facteur plaquettaire 4 est libéré au début de l'aggrégation des plaquettes par l'ADP et l'adrénaline. Ce phénomène a pu être mis en évidence en étudiant l'activité antihéparinique et l'activité paracoagulante (coagulation non enzymatique de complexes solubles de monomères de fibrine) du facteur plaquettaire 4.     

The in vitro effects of several progestogens,estrogens and non-steroidal compounds with estrogenic activity on adenosine diphosphate induced guinea-pig platelet aggregation

Résumé Des disperisions aqueuses de progestogènes et estrogènes stimulent l'agrégation des plaquettes de cobaye produites par ADP in vitro. Le diéthylstilbestérol et l'hexestrol qui ne sont pas des stéroïdes, mais possèdent une activité estrogénique, ont un effet inhibitoire sur l'agrégation.
