Summary Amino acids contained in extracts of adultEutetranychus orientalis were separated and determined quantitatively by 2-dimensional paper chromatography. 14 amino acids were identified. Asparagine, ornithine, histidine, lysine, aspartic acid, serine and glycine were the major components of the free amino acid pool, comprising 83.94% of the total content.  相似文献

A C-terminally elongated form of peptide histidine isoleucine amide (PHI) was isolated from porcine intestine based on its effect on cAMP production in IMR-32 cells. The structure was determined by amino acid sequence analysis of tryptic fragments and by mass spectrometry. The peptide has 42 amino acid residues like those described from human, rat and mouse, but the amino acid sequence of the C-terminal extension of pig PHI is unique. Unlike the other peptides, it has a C-terminal Ala and it differs at five positions from the human form and at six positions from the rat form, while the human and the rat forms differ by only two substitutions. To avoid confusion arising from different C-terminal residues, a unifying nomenclature is proposed: PHI-27 for the hormone and PHI-42 for the elongated product.  相似文献

Summary 19 free amino acids were separated and quantified from the hemolymph of 5th instar larvae of the mothAchaea janata. Proline, histidine, threonine/glutamine/asparagine, lysine, valine and serine were the predominant amino acids in the hemolymph. Changes in amino acid concentration are discussed in relation to metabolic and other physiological activities.Acknowledgment. Y.P. Summary Five 3,4-dihydroxyphenylalanine (DOPA)-containing peptides have been isolated from hemocytes of the ascidian,Halocynthia roretzi. Three of them were composed of DOPA, proline, phenylalanine, histidine and arginine in different ratios, while the other two contained only DOPA and an unidentified amino acid. DOPA-containing peptides were found to exist in only one type of hemocyte.  相似文献

Summary Free amino acids in the haemolymph ofSpodoptera littoralis full-grown larvae infected with a nuclearpolyhedrosis virus were compared with those in the haemolymph of normal insects. Amino acids were separated by 2-dimensional paper chromatography and quantified colorimetrically. Most of the amino acids in the haemolymph of diseased full-grown larvae decreased markedly in concentration but proline, lysine, aspartic acid and histidine occurred in greater concentration in the haemolymph of diseased full-grown larvae than in the haemolymph of healthy insects.  相似文献

The catalytic action of serine peptidases depends on the interplay of a nucleophile, a general base and an acid. In the classic trypsin and subtilisin families this catalytic triad is composed of serine, histidine and aspartic acid residues and exhibits similar spatial arrangements, but the order of the residues in the amino acid sequence is different. By now several new families have been discovered, in which the nucleophile-base-acid pattern is generally conserved, but the individual components can vary. Covalent immobilization of yeast cells by an activated diamine spacer is accompanied by increased levels of cell wall proteins, lipids, amino sugars, amino acids and acid phosphatase leakage, and by altered composition of mannoproteins. The observed changes in cell wall composition are attributed to the effect of cell-solid surface contact.  相似文献

Phosphoproteins of Chicken bone have been extracted by 0.5 M EDTA pH 7.5. Their amino acid composition was similar to that of phosphoproteins of other calcified tissues. The crude EDTA extract contained 80 to 90% proteins, only 70% of the total organic phosphorus was bound to the proteins. We have studied and identified two phosphorylated components. o-phosphoserine and o-phosphothreonine have been identified by amino acid analysis at pH 1.7 from a partial acid hydrolysate, and confirmed by the liberation of the parent amino acids after total hydrolysis. In addition, phosphorus was found equimolecular to both of them. The presence of these phosphorylated groups is important for an understanding of the role of these proteins in the mechanism of mineralization.  相似文献

Summary The concentration of free amino acids in the haemolymph of non-diapausing and diapausing pupae ofHeliothis armigera was investigated. 20 amino acids were detected in the haemolymph of the studied stages. Asparagine, glutamine, cystine, ornithine, histidine and valine were the predominant free amino acids at all stages. The diapause resulted in increased levels of most of the amino acids.  相似文献

The amino acid histidine is an excellent buffer and is therefore included in several organ preservation solutions used in transplantation medicine. However, when used at concentrations as in these solutions, histidine has a marked injurious potential. Therefore, we here assessed the mechanism of histidine-induced cell injury and searched for ways to use the buffering power of histidine but avoid histidine toxicity. When cultured hepatocytes were incubated in HTK solution or in modified Krebs-Henseleit buffer containing 198 mM L-histidine at 37°C, most cells lost viability within 3 h (LDH release 86 ± 7% and 89 ± 5%, respectively). This injury was accompanied by marked lipid peroxidation, and was strongly inhibited by hypoxia, by the antioxidants trolox, butylated hydroxytoluene and N-acetylcysteine and by the membrane-permeable iron chelators 2,2′-dipyridyl, 1,10-phenanthroline, LK 614, LK 616 and deferoxamine. Thus, histidine-induced cell injury appears to be mediated by an iron-dependent formation of reactive oxygen species. D-Histidine, imidazol and L-histidine methyl ester also elicited marked injury, while the N-substituted derivatives Nα-acetyl-L-histidine and tert-butyl-oxycarbonylhistidine and histidine-containing dipeptides showed almost no toxicity. Histidine toxicity, its iron dependence and the superiority of Nα-acetyl-L-histidine were also evident during/after cold (4°C) incubations. Therefore, we suggest the addition of iron chelators to histidine-containing solutions, and/or replacing histidine with Nα-acetyl-L-histidine in organ preservation solutions. This enzyme has the same molecular weight as the Porcine isoenzymes but its isoelectric point is more basic. Summary Free amino acid composition of the intestinal contents, intestinal cells and hemolymph has been determined in larvae of the mothPhilosamia cynthia. From the hemolymph/lumen concentration ratio, an active transport could be inferred for neutral and basic amino acids. The values of cell/lumen and hemolymph/cell ratios suggested that the active step in the transport mechanism could be localized at the luminal pole of the enterocyte for neutral amino acids (except aromatic amino acids) and at the basolateral pole of the enterocyte for basic amino acids (except arginine).This work was supported by grants from Italian Consiglio Nazionale delle Ricerche and from Ministero della Pubblica Istruzione, Rome. Summary Sedimentation coefficient, intrinsic viscosity, amino acid composition and myosin binding capacity of actin from calf's heart were determined. The results are in fair agreement with values published for actin from other sources. No influence of strophantin-g or strophantin-k on the velocity of polymerization and the viscosity and thixotropy of f-actin could be detected.  相似文献

Three antibacterial proteins were isolated from acid extracts of channel catfish (Ictalurus punctatus) skin by cation exchange chromatography and reverse-phase high-pressure liquid chromatography. The molecular masses of the proteins were 15.5, 15.5 and 30 kD as determined by SDS-polyacrylamide gel electrophoresis. Mass spectrometry, amino acid composition and amino acid sequence data suggest that the most abundant protein is closely related to histone H2B. The H2B-like protein was inhibitory to Aeromonas hydrophila and Saprolegnia spp., which are important bacterial and fungal pathogens of fish.
