利用量子化学密度泛函理论(DFT)中的杂化泛函B3LYP对三个系列60个喹啉环取代的喜树碱衍生物(CPTs)进行了构型优化,在6-311+G(d,p)基组下计算出相应的电子结构参数;采用逐步多元回归方法对各组化合物建立了几个新的分子描述符与抗癌活性之间的定量结构-活性关系(QSAR)模型,探讨了影响化合物抗癌活性的主要结构因素及其作用机理.结果表明,所得QSAR模型具有良好的预测能力,影响药物活性的主要因素有分子极化率和原子净电荷以及及最高占据分子轨道能量.根据所建最佳QSAR可以较准确地预测喜树碱类衍生物的抗癌活性,这有助于未来设计合成新型高效抗癌喜树碱类似物以及改进和完善其作用机理.  相似文献

应用量子化学MP3方法对电子结构进行优化,并用MNDO方法计算了21种二吡啶苯并氮杂酮化合物的电子结构,并对其抑制HIV-1病毒活性进行了QSAR研究,结果得到了一个多元线性方程.二吡啶苯并氮杂草酮化合物的活性与7位上的净电荷Q7、偶极矩DIPO以及C环上的净电荷Qc之间有较大的关系.当7位上的正电荷越多则活性越大,分子的偶极矩越小则活性越大,当C环上的净电荷总和越负则活性越大.相关分析的数据还表明,在影响活性大小的诸多因素中,Q7的影响最大.其次是logP与DIPO.据此,可以预测并指导合成有关的抗HIV-1的药物.  相似文献

用量子化学密度泛函(DFT)及分子力学(MM )方法,计算了一系列抗肿瘤喹唑啉衍生物分子的电子结构、几何结构及分子性质(广义结构参数),并通过回归分析,筛选出主要因素,建立定量构效关系(QSAR)方程.发现该类化合物分子的抗肿瘤活性与一些广义结构参数有密切关系,特别是苯环18位碳上取代基(R)的体积与表面积的比值r18,嘧啶环与苯环(C环)间的距离d,以及次低未占据分子轨道(NLUMO)的能量NL与抗肿瘤活性显著相关,可通过取代基的选择与计算来筛选药效最佳的先导物.QSAR方程的拟合相关系数(R2)及交叉验证相关系数(q2)分别为0.851 6和0.714 6,表明所得模型具有可信的预报能力,可用以精确预测该系列化合物活性,并为设计抗肿瘤活性更高的喹唑啉衍生物及作用机理分析提供理论指导.  相似文献

从贵州省黔东南地区土壤分离到一株能产生杀菌活性物质的菌株ZY105,其代谢产物对多种植物病原真菌均具有较强的抑制活性,抗菌谱较广.其中对烟灰霉病菌、油菜菌核病菌和黄瓜灰霉病菌的抑菌效果最好,抑制率达到85%以上,通过稳定性的研究,无论是在连续传代过程中,还是不同温度条件保存下,培养特征和抑菌活性基本没有变化.发酵液中抗菌物质对热及酸、碱也都比较稳定.  相似文献

运用Hamiltonian PM3算法,计算了36种卤代芳烃化合物的26个量子化学参数,在此基础上通过偏最小二乘法得到了卤代芳烃化合物在缺氟沉积物中还原脱卤速率的QSPR模型.该模型可用于解释卤代芳烃化合物还原脱卤的机理.结果表明,卤代芳烃化合物中整个分子的最负的原子净电荷(q^-)、键序最小的碳卤键中碳原子的单中心项电子—核吸引能(ENC)、该碳—卤键上碳原子的净电荷(qc)、分子最低未占据轨道能(EIumo)值越大,该卤代芳烃化合物还原脱卤的速率常数越小;而前线轨道间隙能(△E)、键序最小的碳卤键中碳原子的单中心项电子—电子推斥能(EEC)、相对分子质量(Mr)、整个分子的最正的原子净电荷(q^ )、平均分子极化率(α)值越大,该卤代芳烃化合物还原脱卤的速率常数越大.  相似文献

采用量子化学从头算法,计算10个大黄素类似物分子结构参数,用逐步回归分析法筛选合适结构参数作为网络输入节点,建立大黄素类化合物抗乳腺癌(MCF-7)细胞活性的BP神经网络模型。研究表明,大黄素类化合物抗乳腺癌(MCF-7)细胞活性与分子中A环电荷和第3条最低空轨道能量相关,神经网络模型经留一法交叉验证,具有可信的预报能力。  相似文献

采用Chemoffice10.0中的MOPAC/AM1半经验算法,计算13个无机氢化物的6种量子化学参数:偶极距(μ)、分子电子能(EE)、分子最高占据轨道能(E homo)、分子最低未占据轨道能(E lumo)、分子总能量(TE)和非氢原子的净电荷(q),利用SPSS19.0软件的主成分分析模块和BP神经网络模块对样本数据集进行预处理,将得到的样本数据集输入人工神经网络,构建无机氢化物pK a的主成分-反向传播神经网络(PCABPNN)QSPR模型.结果表明:利用SPSS19.0软件快速实现PCA-BPNN模型取得了非常满意的结果,而且模型的相关系数高,所得结果均优于传统的多元线性回归方法.  相似文献

应用量子化学半经验的MNDO方法计算了40个取代芳烃化合物的电子结构,探讨了化合物电子结构与其对发光菌毒性的关系.结果表明:(1)当化合物的分子极化度P、分子体积V、疏水参数logP较大时具有较大的水溶性和脂溶性,因而化合物的毒性较大;(2)当最高占据轨道(HOMO)能级和最低空轨道(LUMO)能级较低时,化合物容易接受电子具有较大的氧化性,取代芳烃的毒性也随之增大;(3)取代基的吸电子能力越强,吸电子基个数越多,苯环的正电性越大,化合物的毒性越大.得到的QSAR方程为:-logEC50=-2.230 O.202P-0.366EHOMO,据此可预测取代芳烃化合物对发光细菌的毒性.  相似文献

应用PM3量子化学方法和分子连接性指数拓扑理论,进行了氯代苯酚类化合物对水生物发光细菌的半致死量之间的定量结构-活性相关(QSAR)研究,QSAR模型表明,化合物分子体积效应对毒物靶分子的作用呈一定的相关性,体现在其与^1X^v和分子的平均极化率α呈正相关性,而与分子的最低空轨道ELUMO呈负相关性,所得的最显著的相关模型为-logEC50=-1.82532 0.04678α。  相似文献

用中药决明子的乙醇提取物及氯仿提取物对核盘菌(Sclerotinia)、镰刀菌(Fusarium)、柑桔绿霉(Penicillium)、棉花炭疽病菌(ColletotrichumgossypiiSouthw.)等10余种植物病原菌进行抑菌活性筛选研究。结果表明,中药决明子乙醇提取物与氯仿提取物对镰刀菌、弯孢菌、油菜菌核病菌、金黄色葡萄球菌和棉花炭疽病菌都有一定的抑菌作用,其中对油菜菌核病菌和棉花炭疽病菌的抑制效果较好,抑菌率达53%和62%。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
