Differentiation antigens in keratinocytes]

By means of indirect immunofluorescence with human sera it has been demonstrated that cytoplasmic components of the basal cell layer showed antigenic differences as compared to the upper cell layers. Moreover there were differences whether the cells were involved in a mucous, a parakeratinized or an orthokeratinized differentiation.     

Detection, by direct lymphocyte immunocytotoxicity, of antigens in cells transformed by Rous virus]

The hamster cells transformed by the Rous Sarcoma Virus (V.S.R.) evidenced surface antigenic alterations that were detected by a method of cellular mediated immunocytotoxicity. Immune hamster lymphocytes were added to tritiated proline prelabeled target cells. These lymphocytes were able to lyse specifically the V.S.R. transformed cells.

Purification and study of equine infectious anemia virus antigens, produced in vivo or in cultured cells]

The G and C antigens of equine infectious anemia virus have been produced in vivo and in infected cell culture, then they have been purified. Their physico-chemical properties have been determined, particularly their molecular weight, their sedimentation coefficient, their density and the iso electric point.     

Demonstration of Yersinia pseudotuberculosis in in cultivated soils]

The bacteriological analysis of 37 pools of cultivated soils collected in a limited area in the Parisian district permits the isolation of 14 strains of Y. pseudotuberculosis: 9 strains are of serotype II, 3 of serotype I, 1 of serotype III, and of serotype IV. This diversity contrasts with the prevalence of serotype I in infections of man and animals and the scarcity of type III and IV in these species. The abundance of our isolates is in favour of a large distribution of the germ in this substratum, which can be considered as a potential contamination source.

Immunization of hamsters with normal pancreas extracts prevented growth of transplantable pancreatic carcinomas.

Demonstration and evolution of chorionic gonadotropin in ewes]

Occurrence of ovine Chorionic Gonadotropin (oCG) is demonstrated in placenta and amniotic fluid with the use of a radioreceptor assay (corpus luteum membranes) in ewes. Identification of oCG is possible as early as 15th day of pregnancy. It should be secreted at a constant rate, and its maximum concentration is recorded on the 130th day. This hormonal factor might be one of the major components accountable for high progesteronemia observed during the 100 last days of pregnancy.     

Demonstration of erythroblastic differentiation after 14 days in blood in vitro during Vaquez's disease without addition of erythropoietin]

Leucocytes of normal individuals and patients with polycythemia vera were isolated from the peripheral blood by Ficoll-Hipaque density gradient centrifugation and cultured in vitro suing the bovine plasma clot culture technique with a minor modification: the addition of fresh normal serum. After 14 days in the presence of sheep erythropo?etin (3U/ml) erythropo?etic bursts containing between 3 and 10 subcolonies were observed in normal and polycythemia vera cultures. Blood leucocytes of patients with polycythemia vera rise to these erythropo?etic bursts without addition of erythropo?etin to the culture. This behavior was never observed in the blood of normal individuals. These results indicate that in polycythemia vera commited erythro?d stem cells of high proliferative capacity closely resembling the murine erythro?d burst forming unit have an abnormal sensitivity to erythropo?etin as well as the immediate precursors of the proerythroblasts. The culture of these cells from the peripheral blood offers some practical advantages.     

Cytotoxicity of human T-lymphocytes sensitized by Epstein-Barr virus. Role of HLA antigens at the surface of target cells infected by the virus]

Peripheral T-lymphocytes from patients with infectious mononucleosis are cytotoxic towards target cells from Epstein-Barr virus-genome carrying human lumphoblastoid lines. Thus, these T-cells appear to be sensitized to viral coded determinants. Daudi cells, that carry EBV-genome, but lack HLA antigens are resistant to specific cytolysis in this model. These results suggest direct HLA involvement in the target structure recognized by birus-sensitized cytolytic T-lymphocytes in human.     

Effects of parathyroid hormone and calcitonin on carbonic anhydrase location in osteoclasts of cultured embryonic chick bone

Summary The ultrastructural location of carbonic anhydrase has been examined in osteoclasts of cultured embryonic chick metatarsi. In untreated cultures and in those to which parathyroid hormone (PTH) was added, the enzyme was found in close association with the plasma membrane of the ruffled border. After brief calcitonin treatment (10 min) the ruffled border disappeared and the association of carbonic anhydrase with the plasma membrane was diminished. The results indicate that the hormones employed act directly on embryonic bone to alter osteoclast structure and the intracellular location of carbonic anhydrase.Acknowledgments. Outer mitochondrial membranes catalyze the transfer of N-acetyl-glucosamine into polyprenic and proteinic endogenous acceptors. Different effectors (nucleotides, EGTA) are used, suggesting that there are two separate metabolic ways.

Zusammenfassung Es wurde untersucht, ob die Erythroblasten im menschlichen Knochenmark spezifische und unspezifische Antigene, entsprechend den Erythrozyten, besitzen. Diese Eigenschaft wurde auch bei den unreifsten basophilen Erythroblasten beobachtet.     

Demonstration of dopaminergic receptors in human pituitary adenomas secreting prolactin]

3H Domperidone binding on cellular membranes from human prolactin adenomas demonstrates the presence of two dopaminergic binding sites. The mean value of the dissociation constant (Kd) for five adenomas is of 0.29 +/- 0.14 nM for the first site and of 4.19 +/- 1.56 nM for the second site. The maximal number of binding sites (Bmax) varies from one adenoma to another. The binding is completely displaced at 30 nM of tritiated Domperidone by apomorohine, a specific dopaminergic agonist.     

Demonstration of N-acetylgalactosaminyltransferase activity in the murine lymphoma YC8]

The N-acetyl-galactosaminyltransferase activity has been studied in the biological fluids of YC8-lymphoma bearing Balb/c mice. It is enhanced during tumor development from 1 to 30 times in peritoneal fluids and from 1 to 10 times in sera. This activity is nil in urines. Optimal requirements for activity have been determined. Results suggest the existence, during tumor process not only of an enhancement of enzymatic activity, but also of a new molecule synthesis, molecules which are endogenous acceptors for the enzyme.
