鲍氏层孔菌是一种珍贵的药用真菌,本文首次以发酵培养物的免疫增强活性为指标,同时结合发酵生物量、胞外多糖含量等指标,研究发酵时间、初始pH值、碳源、氮源、无机盐、装液量、温度等因素对鲍氏层孔菌发酵的影响.结果表明在培养基初始pH 6.0,碳源为蔗糖,氮源为豆粉,无机盐为MgSO4,装液量为40 mL/150 mL三角瓶,26℃培养7 d时,培养物的免疫增强活性、生物量及胞外多糖含量达到最佳.  相似文献

本实验研究了不同碳源、氮源、温度、发酵时间、接种量、种龄、起始pH值等因素对枯草芽孢杆菌(BaciHus subtilis)B1⑥发酵产多糖的影响,并对发酵影响因素进行了优化实验.结果表明,在发酵过程中,玉米和豆粕粉分别是最佳的碳源和氮源.无机盐的最佳水平为:磷酸二氢钾0.15%,磷酸二氢钠0.1%,MnSO4 0.1%、氯化钙0.2%.其他适宜的发酵条件是:发酵时间5天,接种5%,装液量100ml/300ml,起始pH为8.0.  相似文献

产辅酶Q10热带假丝酵母菌株C-1发酵过程优化   总被引:1,自引:0,他引:1  
该文以热带假丝酵母(candida tropicalis)C-1为CoQ10的生产菌,通过发酵过程的优化,重点考察了碳源、氮源、接种量、酸碱度和装液量等对生物量和CoQ10产量的影响。发现氮源对产物合成影响最大,其次分别是酸碱度、装液量和碳源。优化后的发酵条件是:碳源,葡萄糖26g/L+蔗糖26g/L;氮源,酵母膏7.5g/L+蛋白胨7.5g/L;pH为6.5,装液量为40mL,发酵时间为30h,培养温度为30℃,CoQ10的得率为10.8mg/L。此外,加入适量的维生素B1、烟酸和铁离子等有利于CoQ10微生物细胞的生长和CoQ10产量的提高。  相似文献

从土壤中分离到一株产几丁质酶的细菌LL-C.该菌经16S rDNA序列同源性分析及形态培养特征、生理生化特征分析,鉴定为Luteibacter rhizovicinus,属于黄单胞菌科(Xanthomonadaceae).以不同发酵时间、碳源、氮源、起始pH值、培养基装液量和培养温度为因素,考察LL-C菌株产几丁质酶的优化条件.结果表明,最有利于该菌产几丁质酶的条件:碳源为胶体几丁质、氮源为(NH4)2SO4,培养基起始pH值为4.5,培养基装液量为30 mL,培养温度为32 ℃,振荡培养时间为7 d,此优化条件下,摇瓶产酶活力为115.02 μkat·L-1.  相似文献

对一株磷细菌的发酵条件进行了研究。其中,单因素发酵试验表明,碳源以2%的淀粉最佳,氮源以0.1%的NH4CI最好。最适培养温度为37℃,最适初始pH为7.0,最佳振荡频率为200r/min,装液量为30/250mL,并进行了中试罐发酵分析,测定了罐发酵过程中的菌量、pH、还原糖及总氮的变化情况。  相似文献

从烟草根际土壤中成功筛选出10株解钾细菌,并选择其中四种透明圈明显且生长量较大的菌种进行了解钾活性测定,其中解钾细菌GJ07的发酵液中K~+离子质量浓度高出空白对照组54.5%.对GJ07进行16S rDNA的鉴定,确定其为巨大芽孢杆菌.采用批量发酵培养,通过改变碳源、氮源、碳氮比、温度、pH值、装液量等研究了解钾细菌GJ07生长的优化控制条件.结果表明蔗糖和蛋白胨为GJ07生长的最佳碳源和氮源(碳氮质量比为10:1),最适宜生长温度为35℃,最适宜初始pH值为7.00,最佳装液量为25 mL.  相似文献

本试验以具有纤维素酶活力的黑曲霉诱变菌株为材料,研究了碳源、氮源、碳氮比、培养基起始pH值、接种量及发酵时间对该菌株产植酸酶活力的影响。结果表明,该菌株最适产酶pH为6.0;最佳接种量为10%(v/v);最适碳源和氮源分别为麸皮和黄豆粉,且它们的最优比例为5∶1,最适发酵时间为60h。在最优的发酵条件下,筛选菌株产纤维素酶活可达168U/g。  相似文献

以一株γ-聚谷氨酸高产菌枯草芽孢杆菌B-115为实验菌株,分别考察碳源、氮源种类及浓度、前体物添加量、生长因子和发酵条件对γ-聚谷氨酸产率的影响.优化结果显示:碳源是6.5%的玉米糖化液,氮源是0.4%的普通蛋白胨,前体物谷氨酸钠的添加量为4%,生长因子种类及添加量分别为0.15%硫酸镁、0.006%硫酸锰、0.8%磷酸二氢钾、1.0%氯化钠、0.03%氯化钙;发酵条件为初始pH值6.5,接种量2%,装液量50 mL/250 mL1,50 r/min3,7℃培养84 h;γ-聚谷氨酸的产率可从57.85 g/L提高到68.30 g/L.  相似文献

为了筛选到蝉拟青霉优良的培养基及摇瓶发酵条件,通过对碳、氮源筛选的单因素实验,筛选出葡萄糖为碳源、蛋白胨为氮源;在此基础上以筛选的碳源、氮源及无机盐K2HPO4,MgSO4.7 H2O为考察因素,以菌丝体生物量为指标,利用正交实验筛选出生物量较高的培养基组合.结果表明,最优培养基按质量分数为葡萄糖3%,蛋白胨1.5%,K2HPO40.1%,MgSO4.7 H2O0.05%.适宜的摇瓶发酵条件为培养温度25~27℃,培养基起始pH6~7,接种量6%,摇床转速为150 r/min,500 mL三角瓶最适装液量为100 mL,发酵周期为7 d.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
