Histochemical demonstration of differences in AMP deaminase activity in rat skeletal muscle-fibres

The histochemical method for AMP deaminase confirms that rat slow-twitch fibres contain less AMP deaminase activity than fast-twitch fibres. However, the enzyme appears similarly localized in all fibre types.     

Changes in activity levels of AMP deaminase and adenosine deaminase in the Indian apple snail,Pila globosa (Swainson) during starvation and aestivation stress

Summary Activity levels of AMP deaminase and adenosine deaminase have been studied in hepatopancreas, foot and mantle tissues ofPila globosa with reference to starvation and aestivation. The activity levels of both enzymes were decreased in all the tissues of aestivating snails while in starved animals AMP deaminase activity was increased, in contrast to the decreased adenosine deaminase activity.Acknowledgments. AMP deaminase, 5'-nucleotidase and adenosine deaminase have been estimated in skeletal muscle and myocardial tissue in normal rats and in rats subjected to experimental myocardial infarction or hypothermia. A difference in the enzyme distribution was found between the right and left ventricles in the normal rat. A decrease in the activity of 5'-nucleotidase and an increase in the activity of adenosine deaminase were observed in infarcted myocardial tissue. The activity of all 3 enzymes was found to be depressed in the myocardium in rats subjected to hypothermia. These results are discussed in relation to adenosine production and its beneficial effects.

Beta-adrenergic-stimulated adenylate cyclase activity in normal and EBV-transformed lymphocytes

Summary Beta-adrenergic-associated cyclic AMP accumulation was studied in intact lymphocytes before and after transformation with Epstein-Barr virus into immortal cell lines. Although a marked reduction in isoproterenol-stimulated cyclic AMP synthesis was observed in transformed cells, forskolin-stimulated cyclic AMP accumulation was preserved. A parallel loss of¹²⁵-iodocyanopindolol binding sites suggests that the reduction in beta-adrenergic-stimulated AMP synthesis is due to receptor down regulation.     

Occurrence of sepiapterin deaminase in the normal type silkworm,Bombyx mori

Summary Sepiapterin deaminase was prepared from the normal strain of the silkworm, Bombyx mori. After inhibition experiments, this enzyme was found to have the same properties as that isolated from the lemon mutant strain. Several new inhibitors and their K₁ values are described for the deaminase.The authors wish to thank Dr Wm. Veronal-acetate pretreatment completely inhibited the ATPase reaction in these red fibres but not in type I fibres of the gastrocnemius. The former are type II red muscle fibres and hence are unlike type I red, the so-called slow-twitch muscle fibres.

Adenosine promoted accumulation of adenosine 3',5'-monophosphate in rabbit vagus nerve.

Desheathed rabbit vagus nerve has been found to form cyclic AMP when incubated with adenosine. This accumulation of cyclic AMP is inhibited by theophylline but not by antiadrenergic agents, anticholinergic agents or local anaesthetics. Depolarizing media are not able to promote cyclic AMP accumulation in this preparation.     

Effect of prostaglandins and dibutyryl cyclic AMP on the morphology of cells in primary astroglial cultures and on metabolic enzymes of GABA and glutamate metabolism

Prostaglandins (PGE1) and dibutyryl cyclic AMP (dBc AMP) induce similar morphological changes in astrocytes obtained in primary cultures. PGE1 and dBc AMP increased 2 enzymes of GABA and glutamate metabolism, GABA-T and AAT, but did not modify GDH and GLN-S. Prostaglandins probably affect the cAMP content of glial cells and act in the same way as dBc AMP on glial cell differentiation.     

Immunocytologic differentiation of hypophyseal innervation in carp using anti met-enkephalin and anti-alpha-endorphin serums]

The immunocytological reactions with anti-met-enkephalin and anti-alpha-endorphin reveal two types of fibres in the intermediate lobe of the Carp. Their pathways through the adenohypophysis and the localization of their terminals are different. The anti-met-enkephalin positive fibres terminate mainly in a zone close to the proximal pars distalis; however some fibres terminate deeper in the intermediate lobe. The anti-alpha-endorphin positive fibres penetrate mainly in the central and ventral part where thye are more numerous than the former. It seems that the anti-met-enkephalin positive fibres belong to the gomori positive preoptico-hypophyseal tract, whereas the anti-alpha-endorphin positive fibres are gomori negative.     

Effect of pyrophosphate and orotidine monophosphate on cytosine deaminase regulatory properties

The maximal velocity of the reaction (Vmax) and the half-saturation constant (K0.5) values of the S. typhimurium cytosine deaminase were altered in the presence of its effectors, pyrophosphate and orotidine monophosphate. From the kinetics of orotidine monophosphate inhibition of cytosine deaminase, it was characterized as a mixed-type noncompetitive inhibitor.

Role of adenosine A1 and A2 receptors in the regulation of aldosterone production in rat adrenal glands

To investigate the roles of adenosine A1 and A2 receptors in the regulation of aldosterone production, we examined the effects of adenosine and adenosine agonists (N6-cyclohexyl adenosine; selective adenosine A1 receptor agonist and 5'-N-ethylcarboxamine adenosine; selective adenosine A2 receptor agonist) on aldosterone and cyclic AMP production in rat adrenal capsular cells. Neither adenosine nor 5'-N-ethylcarboxamine adenosine caused significant effects on basal aldosterone or cyclic AMP production. Also, adenosine (10(-3) M) showed no consistent effects on aldosterone and cyclic AMP production induced by ACTH. On the other hand, N6-cyclohexyl adenosine exhibited a significant inhibition of basal aldosterone and cyclic AMP production at doses of 10(-4) M and 10(-3) M; furthermore, 10(-3) M N6-cyclohexyl adenosine inhibited aldosterone and cyclic AMP production stimulated by ACTH. These results suggest that adenosine A1 receptors are coupled to and inhibit adenylate cyclase and may be involved in the inhibition of aldosterone production.     

Degeneration of unmyelinated axons in the dental root canal induced by 6-OH-dopamine

Summary The pulpal nerve fibres of feline incisors were examined ultrastructurally after i.v. administration of 6-OH-dopamine. The presence of degenerating unmyelinated fibres at this site provides conclusive morphological evidence that sympathetic fibres enter the dental pulp.     

Effects of TSH and cyclic AMP on the human thyroid cells cultured in a chemically defined medium

Summary In a serum-free, chemically defined medium human thyroid cells elongated remarkably and resembled fibroblastic cells. They retained the cyclic AMP response to TSH and the supplement of medium with TSH or dibutyryl cyclic AMP permitted the preservation of epithelial nature by the cells. Cyclic AMP of the cells of epithelial nature was higher than those of fibroblastic appearance.     

Elaunin fibres in the basement membrane of sweat gland secretory coil are rich in disulfide-groups.

Disulfide-groups of elaunin fibres of sweat gland basement membrane are demonstrated a) by thiosulfation/aldehyde-fuchsin staining or thiosulfation/Alcian Blue + 0.8 M MgCl2 staining, and b) by identifying SH-groups after reduction with sodiumthioglycollate. Elaunin fibres share this staining behaviour with "elastic fibre microfibrils" and with oxytalan fibres.     

Cyclic AMP is a likely mediator of ovulation in the tsetse fly.

Ovulation in tsetse flies is normally induced by mating, but virgins can be stimulated to ovulate with an injection of dibutyryl cyclic AMP, cholera toxin (a cyclic AMP generator), or aminophylline (a phosphodiesterase inhibitor). Thus, elevation of cyclic AMP is a likely link in the events leading to ovulation.
