Maintenance of human embryonic stem cells on gelatin

Matrigel is routinely used as a coating material in the feeder-free culture system of human embryonic stem cells （hESCs）. However, matrigel is costive and inconvenient to use. In this study, the possibility of using gelatin as an alternative coating material was investigated. The results showed that, after trypsinization, hESCs were maintained undifferentiated on gelatin. These hESCs expressed pluripotent markers, formed teratoma and maintained a normal karyotype. As measured at passage 10, the hESCs expressed a high level of Oct4 on both gelatin and Matrigeh hESCs growing on gelatin formed AP-positive colonies in similar size and number to those growing on Matrigel （P〉 0.05）. Moreover, hESCs growing on gelatin contained a comparable percentage of SSEA-4-positive cells to those growing on Matrigel （95.1% vs.94.3%, P〉 0.05）. H-1 hESCs were maintained undifferentiated on gelatin for 20 passages and remained the stable normal karyotype. This gelatin-based culture protocol may allow us to propagate hESCs in large scale, with less cost.     

带HSV－tk基因的重组腺病毒杀伤离体肺癌细胞的研究

带有单纯疱疹病毒脱氧胸苷激酶基因（ＨＳＶ－ｔｋ）的腺病毒结合ｇａｎｃｉｃｌｏｖｉｒ（ＧＣＶ）对分裂细胞有很强的杀伤作用．在感染复数（Ｍ．Ｏ．Ｉ,ＭｕｌｔｉＰｌｉｃｉｔｙｏｆＩｎｆｅｃｔｉｏｎ）达到１０００时对人体肺腺癌细胞Ａ５４９的杀伤几乎达到１００％．四种人体肺癌细胞株（Ａ５４９,ＬＡＸ,ＳＰＣ,ＳＫＹ）对带ＨＳＶ－ｔｋ的腺病毒（ＡＤＶ／ＲＳＶ－ｔｋ）的杀伤作用表现出不同的敏感性．另Ａｃｙｃｌｏｖｉｒ（ＡＣＶ）和ＧＣＶ对感染了重组腺病毒ＡＤＶ／ＲＳＶ－ｔｋ的细胞都有一定杀伤作用,但杀伤效果有很大差别;就Ａ５４９而言,ＧＣＶ的杀伤作用比ＡＣＶ高７～８倍．此外ＡＤＶ／ＲＳＶ－ｔｋ结合ＧＣＶ杀伤肿瘤细胞时有“旁观者效应”,即感染了ＡＤＶ／ＲＳＶ－ｔｋ的细胞与未感染细胞混合后,后者也明显地遭到杀伤．     

Lectin-induced expression of DR antigen on human cultured follicular thyroid cells

HLA-DR antigens, the human equivalent of mouse I region-associated or Ia products, are polymorphic cell surface sialoglycoproteins involved in initiation of the immune response. Their expression is normally restricted to B lymphocytes, macrophages, dendritic and other antigen-presenting cells and vascular endothelium and possibly some cells of the mucosa lining body cavities. HLA-DR expression can be modified during cell differentiation; B lymphocytes become negative on maturing to plasma cells and human T lymphocytes acquire these antigens when activated in vitro or in vivo. We report here that human thyroid follicular cells which are normally negative for HLA-DR molecules, can be induced to express these antigens when cultured with phytohaemagglutinin (PHA), concanavalin A (Con A) or pokeweed mitogen (PWM). These lectins exert their action directly on the thyroid cells with no concomitant mitogenic effect.     

Preliminary study on human fibroblasts as feeder layer for human embryonic stem cells culture in vitro

To avoid the direct contact with mouse cells and possible heterogeneous pathogen in future application ,we need to replace mouse embryonic fibroblasts with human fibroblasts as the feeder layer to maintain human embryonic stem cells growth in the undifferentiated state,We Success-fully use human fibroblasts derved from aborted fetus and adult prepuces as feeder layer to maintain human embryonic stem cells growth ,During the passage and growth on this feeder layer,the human embryonic stem cells can keep their undifferentiated state.     

Generation of tetraploid complementation mice from embryonic stem cells cultured with chemical defined medium

Mouse embryonic stem cells(mESCs)derived from inner cell mass(ICM)of pre-implantation embryos,can maintain undifferentiated state when cultured in N2B27 medium supplemented with GSK3inhibitor CHIR99021 and MEK inhibitor PD0325901(‘‘2i’’)and leukemia inhibitor factor(LIF).Compare to conventional culture medium,all components of this medium are defined.With the N2B27 medium,‘‘2i’’and LIF,mESCs can contribute to the germline of the chimeric embryos,however,whether the‘‘all-ES cells’’mice can been generated by tetraploid complementation is unclear yet,while the tetraploid complementation serve as a golden standard to assess the pluripotency of ES cells.Here,our study showed that mESCs derived and cultured with the N2B27 complete medium could generate fertile mice by tetraploid complementation.In addition,the survival rate of tetraploid complementation mice produced by inbred mES cell lines is higher than the conventional culture condition,and increased the percentage of Oct4 positive cells contrast to conventional medium either.Therefore,the N2B27 medium supplemented with‘‘2i’’and LIF is an alternative choice forthe derivation and long-term culture of mouse embryonic stem cells.     

利用人胚胎干细胞评价反式维甲酸的胚胎发育毒性

目前,药品安全、食品安全和环境污染等问题受到国人的广泛关注,也受到政府的特别重视。医用化学品、食品添加剂和环境污染物等通过各种途径进入体内,可能产生胚胎发育毒性;因此,建立新的高通量、高灵敏的胚胎发育毒性检测和评价十分重要。研究尝试利用全反式维甲酸(all-trans retinoic acid,RA)对人胚胎干细胞H9的细胞毒性和分化抑制,评价RA的胚胎发育毒性。通过CCK8检测不同浓度下RA对人胚胎干细胞H9和鼠胚胎成纤维细胞3T3的细胞存活百分数和半数增殖抑制浓度(50%inhibitory concentration,IC50),发现处理5 d和10 d,RA对人胚胎干细胞H9的IC50分别是9.23μg/mL和7.20μg/mL,明显低于3T3细胞。通过实时定量PCR检测不同RA浓度下,自然分化20 d的细胞中Nkx2.5、α-MHC、ACTC1和TNNT2基因的表达,发现0.3μg/mL及其以上浓度RA显著抑制H9细胞向心肌细胞分化。RA对H9细胞α-MHC、TNNT2和ACTC1的半数抑制分化浓度(ID50),分别为0.16、0.07和0.05μg/mL,ACTC1和TNNT2的ID50明显低于α-MHC的,提示ACTC1和TNNT2可能更适合作为人胚胎干细胞分化抑制的评价的指标。     

氧化石墨烯对人肺腺癌细胞的毒性产生的剂量效应

氧化石墨烯(GO)具有比表面积大、亲水性、溶液分散性好等独特的理化性质,而日趋广泛应用在生物医学。目前,在其低剂量下暴露细胞的生物安全性方面的报道很少,本实验选取GO,并对其理化性质表征。制备成浓度为0μg/m L、2μg/m L、4μg/m L、8μg/m L、16μg/m L、32μg/m L使其暴露24 h在体外培养的人肺腺癌细胞A549,检测胞内超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT的活性,以及胞内的蛋白含量,荧光染色检测细胞产生的活性氧(ROS)的含量。MTT检测24 h、48 h、72 h细胞活性。结果表明,GO暴露24 h,8μg/m L对A549的活性具有最高促进作用。胞内SOD、CAT活性显著升高,蛋白含量增加,胞内ROS、MDA的含量降低。结论是GO对A549细胞活性具有双重性,低浓度下下调细胞内ROS,降低GO对A549的氧化损伤,从而提升细胞活性;高浓度下对细胞产生氧化损伤,抑制细胞活性。
