将重组质粒pPIC9K-apoAⅠ-milano用BglⅡ酶切后转化Pichia pastoris GS115菌株,筛选得到G418高抗性转化子,经甲醇诱导和摇瓶发酵,用SDS-PAGE与Western blot检测上清液,发现有明显的重组人载脂蛋白AⅠ米兰突变体(rAIM)表达,但其分子量比正常蛋白小.对酵母工程菌的摇瓶发酵条件进行优化后,rAIM表达水平得到提高, 约为70 mg/L发酵液,其中正常大小的rAIM所占的比例约为80%.采用冷丙酮沉淀法初步纯化了酵母工程菌发酵液中的rAIM,并验证其有磷脂结合能力.  相似文献

以表达人Cu,Zn-SOD的重组毕赤酵母为出发菌株,通过摇瓶实验研究发酵初始pH值、诱导剂浓度、诱导温度和培养基组成等因素对目的蛋白表达水平的影响.实验结果表明,培养基组分对目的蛋白表达水平的影响最大.与YPG培养基相比,菌株在FM22培养基(含0.15%组氨酸及PTM4)中目的蛋白表达量提高5.5倍.在优化摇瓶发酵条件下(诱导剂浓度1%,诱导温度27℃,FM22培养基(含0.15%组氨酸及PTM4),初始pH值为6.0),发酵液上清酶活水平为895 U/mL,较初始提高8.5倍;以摇瓶实验结果指导5 L罐发酵,得到13 539 U/mL酶活,为摇瓶试验的15倍.  相似文献

对黄色短杆菌突变株BM2610生产L-异亮氨酸的摇瓶发酵条件进行了研究.单因素优化了菌株BM2610的发酵培养基与培养条件,确定了发酵培养基的最佳碳氮源、最适起始pH、装液量及接种量等,进一步通过响应面分析方法优化了发酵培养基的组成配比.结果在优化条件下,BM2610摇瓶发酵平均积累L-异亮氨酸16.11 g·L~(-1),比未优化前提高了126.3%.  相似文献

本报道了以玉米为主要原料,采用红曲霉菌,通过液体深层发酵生产红曲色素的最佳培养基组成及其相关工艺条件的选择。摇瓶结果表明,按最佳培养基配方,以8-10%的接种量,在32℃经过72-96hrs的摇瓶培养,发酵液红色素色价基本稳定在120u/ml。最高达150-160u/ml,湿菌体收率为培养液的12%,湿菌体色价为800-1000u/g。  相似文献

采用滤纸片法测定菌丝体生物量以及苯酚-硫酸法测定不同培养期的桑木层孔菌多糖含量和发酵液中残糖含量,研究了摇瓶培养条件下不同培养期的桑木层孔菌多糖含量的动态积累规律.结果表明,桑木层孔菌在摇瓶中培养12d时,获得的菌丝体最多,干重达13.580g/L;培养8d时得到最多的胞外、胞内多糖,含量分别达4.585g/L和32.465mg/g;培养到12d时发酵液中残余糖量最少,为4.744g/L.该实验为进一步研究和开发利用桑木层孔菌多糖奠定了实验基础和科学依据.  相似文献

通过摇瓶试验,研究含盐量为7 000 mg·L-1、石油含量分别为10 000 mg·L-1,30 000 mg·L-1,50 000 mg·L-1条件下,空白对照组(不添加微生物)、细菌组、真菌组分别在5,12,19,26,33 d时对石油的降解效果,并对比分析了降解效果.结果表明,经过33 d后,空白对照组中石油含量未变化,细菌组石油含量为10 000 mg·L-1一组的石油降解率最高,达到了65.69%,真菌组石油含量为30 000 mg·L-1一组石油降解率最高,达到了45.84%.可见,在较高的含盐量条件下,细菌和真菌对石油的降解也仍然有显著效果.  相似文献

采用液体摇瓶培养法,对茶薪菇液体发酵的培养基pH、最适温度、摇瓶装量、摇瓶转速及不同液体培养基配方进行了探讨,结果表明,茶薪菇液体发酵的培养基初始pH为6.0-6.5,最适温度为25℃,摇瓶装量为50-70ml/250ml三角瓶,摇瓶转速为120-150rpm,最适培养基配方为以麸皮为主要成分的培养基。  相似文献

为了提高兰花炭疽病拮抗菌巨大芽孢杆菌(Bacillus megaterium)1-12菌株的芽孢形成率及芽孢数量,在摇瓶发酵基础上对芽孢形成的主要影响因素进行了考察.通过单因素实验和正交实验对1-12菌株产芽孢条件进行了分析,确定了摇瓶发酵最佳条件:培养基组成(质量分数)为玉米粉1%,大豆蛋白胨1%,CaCl2·2H2O·"0.1%,MnSO4·H2O 0.05%;培养液初始pH 6.0;培养条件为种龄20 h,装瓶量30 mL/250 mL,接种量8%,温度30 ℃,摇瓶转速200 r/min.在此条件下,B. megaterium 1-12菌株发酵液中的芽孢数量为1.92×109个mL-1,芽孢形成率达到了95%.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
