对长白白眉蝮蛇蛇毒类凝血酶的理化及酶学性质进行系统研究. 结果表明, 该酶为一分子量为38 000, 等电点为450 并对温度有很高稳定性的糖蛋白, 该酶的最适p H 值为80,其活力可被 D F P 和 P M S F抑制.  相似文献

用恒电位法在电化学聚合吡咯的同时,将酪氨酸酶固定在导电聚吡咯膜内,制成一种灵敏、稳定的酪氨酸酶电极。讨论了溶液p H 值和聚合电位对酶固定化的影响,对酶分子嵌入前后吡咯膜的 S E M 图和 C V 曲线进行了分析和比较。该电极响应对甲苯酚的线性范围为5 .0 ×10 - 8 ~1 .0 ×10 - 6 mol· L- 1 ,最适p H 值为6 .6 ,酶反应表观上遵循 Michaelis_ Menten 动力学,表观米氏常数为2 .2 ×10 - 5 mol· L- 1 。  相似文献

绿色木霉926液体达发酵终点(6~7d)后,所得纤维素酶中CMC酶活最高可达20U/ml,滤纸酶活最高可达5.5U/ml.发酵液纤维素酶中CMC酶组分经硫酸铵沉淀,SephadexG—100柱层析后可提纯6倍左右.紫外光谱分析表明:纤维素酶及其组分在280nm附近有强的吸收峰;红外光谱分析表明:纤维素酶在O—H,N─H,C—N,C=O有吸收带;CMC酶学研究表明,CMC酶作用的最适反应PH为5.0,最适反应温度为60℃,常温下稳定的pH范围是4.5~6.0.  相似文献

类产碱假单胞菌的谷氨酸脱氢酶水平受氮源调节,以氮为唯一氮源时酶水平有明显提高,研究表明,该酶的最适反应温度为50℃,催化氢化反应和脱氨反应的最适pH值分别为8.0和8.8,金属离子及嘌呤核苷酸对酶活力的影响不显著。  相似文献

以水稻为材料根施系列浓度的Na2SeO3,试验对稻苗生长以及对苗中GSHPx的影响.结果表明,低浓度的Na2SeO3(005~10mg/L)能促进稻苗生长,最适浓度为05mg/L;高浓度(≥20mg/L)则对稻苗产生毒害.稻苗GSHPx活性高度依赖于Se的存在,揭示出该酶可能是一种依赖于Na2SeO3的诱导酶.GSHPx能有效地抵抗稻苗的Se中度中毒,其酶活较对照提高958%;对于严重Se中毒的稻苗有一定的抵抗作用,其酶活较对照提高692%.  相似文献

从高效降解真菌Z58培养物中通过硫酸铵分级沉淀和SephadexG100分子筛层析提取和纯化降解酶,研究了该降解酶对乐果的降解特性,Z58菌株的乐果降解酶为胞内酶,主要分布于细胞膜组分,该酶对乐果酶促降解最适pH为75,最适温度为40℃,其米氏常数Km为059%.  相似文献

从菊芋根际土壤中分离出一株菊粉酶(InulinaseE.C.3.2.1.7)产生菌,经物理和化学诱变得高酶活黑曲霉突变株AJ1958.该菌株在培养基(0.07g/mL菊芋提取液1000mL,豆饼粉5g,麸皮5g,KCI0.7g,FeSO4·7H2O0.01g,MgSO4·7H2O0.5g,PH5.0)中,于30℃振荡培养3d,酶活力可达64μmol·min-1.酶水解反应的最适PH为3.0~4.0,最适温度为60℃,在PH2.5~5.5的范围内和75℃以下较稳定.适宜条件下,10h内0.05g/mL的菊粉溶液中之底物几乎100%被酶水解.产物糖中果糖质量分数为93%,葡萄糖质量分数为5.6%.该突变株产生的菊粉酶具有较满意的水解性和热稳定性.  相似文献

本文研究了嗜碱芽孢杆菌( Bacillus alkalophilic) N T T33β- 甘露聚糖酶的产生条件和酶学性质.其产酶的最佳碳源为 1% 槐豆角,最佳氮源为 1% 蛋白胨+ 0.2% 酵母膏,发酵培养36h 产酶量最高(达 61.3u/m l).甘油、葡萄糖、甘露醇等对产酶有强的阻遏作用.经分离纯化后测得纯酶的分子量为38.9 K D,等电点 p I为3.0,酶反应最适 p H 为 9.0~10.0,最适温度为80℃,稳定 p H 为6.0~9.0,稳定温度为60℃以下,金属离子中 Cu2+ , Ba2+ , M g2+ , Al3+ , Co2+ 对该酶有一定的激活作用,而 Ag+ , Hg2+ , Mn2+ 对该酶有明显抑制作用  相似文献

研究了一株从白酒发酵现场分离得到的K123菌株普鲁兰酶的合成及其部分酶性质.结果是K123菌株的异淀粉酶在培养液中的大量积累发生在稳定期后期,菌的最适生长温度为35℃,产酶的最适温度则为30℃;菌的最适生长pH和产酶的最适pH均在7.0~7.5;产酶的最佳碳源为可溶性淀粉,有效碳源是糯米淀粉、糊精、支链淀粉、玉米面、茁霉多糖、麦芽糖,而葡萄糖则抑制产酶;产酶的最佳氮源是蛋白胨、酪素水解物,无机氮源则使得K123菌株的产酶量略低于有机氮源.通气量对菌体的生长影响较大,但菌体的生长达稳定期后,通气量的改变对菌的产酶影响不大.K123菌株所产的异淀粉酶水解茁霉多糖的产物为麦芽三糖,不水解动物淀粉,是普鲁兰酶(pululanase).该酶反应的最适温度为50℃,最适pH5.8,受Ca2+,Mn2+等离子激活,受Cu2+,Zn2+等离子抑制;该酶的热稳定性较差,50℃以上迅速失活;pH稳定性较好,中性,微酸性条件下,冰箱中可长期保存.该酶应用于固态白酒发酵可提高出酒率平均4%以上.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
