The ultrastructure of blood capillaries and venules are studied in rat skin. After i.v. injection of histamine the luminal surface of the endothelial cells show protrusions of variable size. Numerous gaps have been found in the capillary wall, specially in the venules. These alterations are not observed in the animals that were treated with calcium dobesilate before the administration of histamine. In these cases the capillary structure are indistinguishable of the controls. The possible effects of the histamine and calcium dobesilate on the cell coat and cell junctions of the endothelial capillary cells are discussed. 相似文献

Serglycin is a proteoglycan found in hematopoietic cells and endothelial cells. It has important functions related to formation
of several types of storage granules. In connective tissue mast cells the covalently attached glycosaminoglycan is heparin,
whereas mucosal mast cells and activated macrophages contain oversulfated chondroitin sulfate (type E). In mast cells, serglycin
interact with histamine, chymase, tryptase and carboxypeptidase, in neutrophils with elastase, in cytotoxic T cells with granzyme
B, in endothelial cells with tissue-type plasminogen activator and in macrophages with tumor necrosis factor-α. Serglycin
is important for the retention of key inflammatory mediators inside storage granules and secretory vesicles. Serglycin can
further modulate the activities of partner molecules in different ways after secretion from activated immune cells, through
protection, transport, activation and interactions with substrates or target cells. Serglycin is a proteoglycan with important
roles in inflammatory reactions.

In unoperated fasted rats, feeding raised the serum gastrin concentration, reduced the gastric mucosal histamine content and activated the gastric histidine decarboxylase. The reduction of gastric histamine and activation of histidine decarboxylase was induced also by the injection of pentagastrin. In antrectomized rats, feeding failed to produce these effects. Injection of pentagastrin, however, still lowered gastric histamine and activated gastric histidine decarboxylase. Thus, antral gastrin seems to be an obligatory mediator of the postprandial activation of histidine decarboxylase and mobilization of histamine. 相似文献

In the presence of Zn2+ (0.3 mM), carbachol (10–6 M) or histamine (10–5 M) induced the phasic response in guinea-pig taenia caeci while the tonic response was markedly inhibited. However, when the muscles were kept in Zn2+-containing medium following the first stimulation with either carbachol or histamine, neither application of carbachol nor of histamine elicited another phasic contraction. Caffeine (25 mM) did not induce contraction in the presence of Zn2+. After the washing out of caffeine in the presence of Zn2+, however, the muscle did then develop the phasic response on the application of carbachol or histamine. In conclusion, Zn2+ did not affect the carbachol or histamine-induced Ca2+ release from the storage sites. However, when Zn2+ was continuously present, Ca2+ was not supplied to the storage sites. Furthermore, carbachol and histamine mobilized a common cellular Ca2+ store, but they activated Ca2+ release channels different from the ones activated by caffeine in the Ca2+ storage sites. 相似文献

Heat-shock protein 60 (Hsp60) is a highly conserved stress protein which has chaperone functions in prokaryotes and mammalian
cells. Hsp60 is associated with the mitochondria and the plasma membrane through phosphorylation by protein kinase A, and
is incorporated into lipid membranes as a protein-folding chaperone. Its diverse intracellular chaperone functions include
the secretion of proteins where it maintains the conformation of precursors and facilitates their translocation through the
plasma membrane. We report here that Hsp60 is concentrated in apoptotic membrane blebs and translocates to the surface of
cells undergoing apoptosis. Hsp60 is also enriched in platelets derived from terminally differentiated megakaryocytes and
expressed at the surface of senescent platelets. Furthermore, the exposure of monocytic U937 cells to Hsp60 enhanced their
phagocytic activity. Our results suggests that externalized Hsp60 in apoptotic cells and senescent platelets influences events
subsequent to apoptosis, such as the clearance of apoptotic cells by phagocytes. 相似文献

An increased production of histamine has been demonstrated during mixed cultures between allograft donor and recipient lymphocytes. This phenomenon could result from the action of a non-dialysable factor released by recipient cells in the presence of donor cells. This factor is able to increase histamine production from normal spleen cells. Mast cell activation involves the rapid release of inflammatory mediators, including histamine, from intracellular granules. The cells are capable of regranulation and multiple rounds of activation. The goal of this study was to determine if there are changes in the content of pre-formed mast cell mediators after a round of activation. After 24 h, the histamine content of bone marrow-derived mast cells (BMMC), but not that of peritoneal mast cells, exceeded the amount in resting cells. Accumulation of histamine in BMMC peaked at 72 h of activation, and returned toward preactivation levels by 96 h. The increase in histamine content was accompanied by an increase in the gene expression of histidine decarboxylase. No increases in beta hexosaminidase or murine mast cell protease-6 were observed. These findings indicate that BMMC respond to activation by increasing total cell-associated histamine content. This increase may be important to the response of these cells upon subsequent exposure to antigens. 相似文献

Oriented cellulose deposition is critical to plant patterning and models suggest microtubules constrain cellulose synthase
movements through the plasma membrane. Though widespread in plants, urochordates are the only animals that synthesize cellulose.
We characterized the distinctive cellulose microfibril scaffold of the larvacean house and its interaction with house structural
proteins (oikosins). Targeted disruption of cytoskeletal elements, secretory pathways, and plasma membrane organization, suggested
a working model for templating extracellular cellulose microfibrils from animal cells that shows both convergence and differences
to plant models. Specialized cortical F-actin arrays template microfibril orientation and glycosylphosphatidylinositol-anchored
proteins in lipid rafts may act as scaffolding proteins in microfibril elongation. Microtubules deliver and maintain cellulose
synthase complexes to specific cell membrane sites rather than orienting their movement through the membrane. Oikosins are
incorporated into house compartments directly above their corresponding cellular field of expression and interact with the

The smooth muscle relaxation induced by nitroglycerin is hypothesized to be mediated by an increase in the cytoplasmic concentration of guanosine 3′,5′-monophosphate (cGMP) and subsequent dephosphorylation of the 20-kilodalton myosin light chain (MLC). We investigated this hypothesis in procine coronary arterial smooth muscle stimulated with histamine (3 μM) or K+ (30 mM). Stimulation of [32P]Pi-labeled muscle with histamine or K+ for 2 min resulted in a four- or 6.2-fold increase, respectively, in the incorporation of32P into MLC. After 48 min of exposure to histamine. MLC phosphorylation decreased to the basal level and the phosphorylation of desmin, synemin, and of three unidentified cytosolic proteins was increased. K+ stimulation resulted in a sustained increase of MLC phosphorylation but had no effect on the phosphorylation of desmin, synemin, or the three unidentified cytosolic proteins. Application of nitroglycerin (1 μM) 48 min after histamine stimulation inhibited the phosphorylation of desmin, synemin, and the three cytosolic proteins. The sustained phase of histamine-induced contraction was also inhibited to a greater extent then the acute phase of histamine-induced contraction and both the acute and sustained phases of K+-induced contraction. These results suggest that MLC phosphorylation is required for both phases of K+-induced contraction, whereas phosphorylation of intermediate filament proteins is required for the sustained phase of histamine-induced contraction. Intermediate filament proteins, rather than MLC, may also be the target for the relaxant action of nitroglycerin during histamine-induced sustained contraction. 相似文献
