文章报道了赖草(Leymusscaline)花粉母细胞(PMC)的细胞融合现象。结果表明,细胞融合主要发生在间期、终变期。间期发生细胞融合主要是细胞质转移,因此不会产生明显的遗传学效应。终变期细胞融合主要是染色体的迁移,其细胞遗传学效应主要是造成供体和受体细胞的染色体数目变化,从而产生了非整倍体配子。文章还对细胞融合产生的原因进行了讨论。  相似文献

自1958年Okada首次表明紫外灭活的仙台病毒可以诱导体外培养细胞融合形成多核体以来。细胞融合技术得到了迅速发展和应用。以聚乙二醇(PEG)为代表的化学融合方法在生物、遗传、医药学等研究领域取得了可喜的成绩。自1978年报道电脉冲诱导细胞融合成功以来.Zimmermann及其同事将细胞电介质电泳引入诱导细胞间接触,创立了物理的实用的Zimmermann电融合法。现在,电融合法已由非特异性的细胞电融合发展为特异性电融合法。激光诱导细胞融合技术成为目前的研究热点.但此项技术尚处于发展初期,有待于进一步完善。本综述了PEG化学融合法.电融合领域的新进展和激光诱导细胞融合技术。  相似文献

活化雌三醇 (E3 )C3处的酚—OH ,与牛血清白蛋白 (BSA)偶联 ,制备得E3 的完全抗原。利用完全抗原免疫动物 ,采用单克隆抗体技术 ,经过细胞融合、克隆筛选、扩大培养、动物体内诱生等过程 ,最后获得了E3 的单克隆抗体。对纯化的单克隆抗体的性质进行表征 ,结果表明 :抗体的效价为 6 0× 10 -10 mol L ,抗体属于IgG1型 ,分子量为 16 80 0 0u ,单克隆抗体与固定化抗原亲和常数为4 7× 10 7L mol。  相似文献

体细胞电融俣技术因其融合效率高、对细胞无毒害作用而愈来愈普遍地用于细胞工程技术操作中。为了准确地确定实验参数,试验采用一个组的电融合仪和自制平行电极,以马苓薯和薯蓣为材料,研究了在不同原生质体密度和不同电场强度件下的细胞融合效率。试验结果显示,以交变电场频率1.0MHz、电压140Vpp.cm^-1处理原生质体20-40秒,尔后施加瞬时直流高压(120kv)脉冲1-2次,可以获得73.5%-76.7%的融合细胞,其中11.5-14.4%的融合细胞可发生正常分裂。在本试验设计范围内,电场强度对细胞融合频率的影响主要表现在融合细胞能否发生分裂,而初始融合频率在各处理间无显著差异。  相似文献

采用电激细胞融合的方法 ,对紫罗兰MatthiolaincanaR Br与桂竹香CheiranthuscheiriL 原生质体培养及电激细胞融合效率导入最适条件作了探讨。紫罗兰的子叶原生质体分裂初期培养最适条件 :培养基为 1 2MS。培养基组成中 ,蔗糖质量分数为 4%、甘露糖醇浓度为 0 6mol L。加入谷酰胺酶无机态氮源时 ,效果明显。次之 ,加入植物生长调节物质 1mg LBA也达到高分裂率。再者 ,添加玉米素时效果也甚好。根据原生质分离前的子叶在 5℃预处理分裂率最高 ,培养温度 2 4~ 30℃为宜。电激细胞融合 (singlepairs)产生效率高的条件为 :紫罗兰原生质体密度 1× 10个 mL ,高周波电强度 1MHz ,15 0V cm ,脉冲幅 70 μs;桂竹香原生质体密度 1× 10个 mL高周波电强度 1MHz,10 0V cm ,脉冲幅 30 μs。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
