An improved method for the isolation of eosinophilic leukocytes from normal human blood

Summary A relatively short and simple method for the isolation of the eosinophils from normal human blood is reported. With a recovery of about 45–55%, cell preparations, showing a degree of purity of 90–98%, are obtained. The isolated cells are morphologically intact and viable, as assessed by the trypan blue exclusion test and by active phagocytosis.The authors are indebted to the Schweizerischer National-fonds zur Förderung der wissenschaftlichen Forschung for financial support.

A simple enzymatic method for isolation of hepatocytes

Summary A new method is described for isolating hepatocytes by an enzymatic procedure: 0.05% bacterial collagenase in calcium-free Hanks' solution is injected into the liver tissue which is removed aseptically. The time required to prepare culture and risk of bacterial and fungal contamination is greatly reduced.     

A simple method for the collection of leucocytes from rat blood

Zusammenfassung Blutleukozyten von Ratten wurden mit menschlichem (normalem) Serum behandelt. Die Viabilität dieser Präparationen konnte dadurch demonstriert werden, dass die Zellen durch Phytohemagglutinin in «blast» Formen umgewandelt wurden.     

A simple method for blood exchange in mice

Summary Ease of transfusion and high long-term survival rate were obtained when whole blood was administered via the corpus cavernosum of the penis and removed from the orbital sinus of male C57Bl mice. When 2 volumes of blood are replaced approximately 14% of pre-transfusion red cells remain after hematocrit corrections are made. A rapid method for the fractionation of avian blood cell nuclei

Summary Red blood cell nuclei from avian anaemic blood have been fractionated by rate sedimentation on discontinous sucrose gradients into fractions which can be distinguished both by their RNA synthesizing ability and by their morphology as revealed by electron microscopy.Acknowledgment. Oxidation of bilirubin with chloranil. A simple method for preparing isomerically pure biliverdin

Summary Isomerically pure biliverdin IXa can be prepared in high yield through dehydrogenation of bilirubin IXa with chloranil-picric acid in chloroform containing t-butanol.     

A method for the isolation of ommochrome-containing granules from insect eyes

Summary A routine method for the preparation of pigment granules from insect eyes is reported. When viewed using an electron microscope, the isolated granules appear to be free from different structural elements, and there is very little contamination by membranous material. A rapid electrophoretic method for the separation of hydroxyproline from proline

Summary A rapid, sensitive procedure is described which uses custom made apparatus to separate radioactive imino acids. This technique provides a simple measurement of the distribution of newly synthesized collagen in different fractions.Acknowledgment. A method for the isolation of the parts of ciliates

Zusammenfassung Eine Methode für selektive Solubilisierung der Zellen und verschiedener Organellen von Ciliaten, welche auf einer speziellen Anwendung der MethodeMazia undDan zur Gewinnung isolierter Mitoseapparate von Seeigeleiern beruht, wird beschrieben. Die Ciliaten werden bei niedriger Temperatur in 40% Ethanol extrahiert und mit Digitonin behandelt. In salzfreier Digitoninlösung (1% aq) bleiben nur das Wimpernsystem und damit verbundene Strukturen erhalten und können in genügender Menge für chemische Analysen gewonnen werden. Durch Zugabe verschiedener Mengen von KCl ist es möglich, auch Zellkerne, Pellicula, usw. vor der Auflösung zu schützen und durch differentielle Zentrifugierung zu isolieren.

A simple apparatus for the rapid extraction and weighing of small animal brains

Summary The construction and use of an inexpensive apparatus for the rapid extraction and weighing of small animal brains is described.     

A simple method of obtaining multiple blood samples from the portal vein and the hepatic vein in the rat in vivo

A very simple and rapid technique for inserting a catheter in the portal vein and the hepatic vein in the anesthesized rat in vivo is described. The pointed, saline-containing PE tubing is frozen in liquid nitrogen, whereupon it is used as a 'needle' to insert the catheter into the blood vessel. Multiple blood samples can be obtained from the portal and the hepatic vein at the same time, so that in situ extraction of drugs by the liver can be measured in vivo, since hepatic blood flow is uninterrupted.     

A simple method for the preparation of diastereoisomeric phosphonodipeptides

Summary Diastereoisomeric mixtures of phosphonodipeptides, prepared from racemic dialkyl 1-aminoalkanephosphonates, were separated by ion-exchange column chromatography.
