分泌型Survivin真核表达载体的构建及其在HeLa细胞中的表达

目的:构建含6个组氨酸标签(His6)的小鼠存活素(Survivin)融合蛋白的真核表达载体,及其在真核细胞中的表达。方法:根据小鼠Survivin序列设计引物,经过RT-PCR克隆Survivin的cDNA并进一步构建分泌型Sur-vivin的真核表达载体,经测序鉴定后在大肠杆菌中扩增在HeLa细胞中表达;以金属离子螯合层析富集,再用免疫印迹法鉴定。结果:克隆到Survivin编码区全长序列,经DNA测序后证明与已报道序列相同。构建氨基端融合小鼠IL-2信号肽,羧基端带有His6标签的Survivin融合蛋白的真核表达载体,在HeLa细胞中转染成功并且表达;细胞上清经HisTrap HP柱层析富集后,进行免疫印迹分析,表明该融合蛋白以分泌型方式在HeLa细胞中表达。结论:成功克隆Survivin的cDNA,并构建其分泌型真核表达载体。     

DLL1-ICD基因真核表达载体的构建及转染鉴定

通过克隆DLL1-ICD(Delta-like1细胞内区域)的cDNA及测序鉴定,构建pEGFP-C1-DLL-ICD真核表达载体并进行细胞转染,为研究Notch信号通路与Neuritin的关系奠定基础。首先根据小鼠全长DLL1 cDNA序列,设计DLL-ICD PCR引物,以小鼠胎脑cDNA文库为模板,利用PCR技术扩增DLL1-ICD cDNA。将扩增出的基因片段克隆至pEGFP-C1载体,进行DNA序列测定。并进一步将测序正确的pEGFP-C1-DLL1-ICD真核表达载体转染HEK293细胞。结果显示:成功克隆了DLL1-ICD的cDNA片段,测序结果与基因文库中的DLL1-ICD序列一致,并在转染真核细胞后获得了较好地表达。表明pEGFP-C1-DLL1-ICD真核表达载体构建成功,并在293细胞中成功表达了DLL1-ICD-EGFP融合蛋白。研究结果为进一步探讨Notch信号通路与神经系统疾病的分子机制奠定基础。     

结核分枝杆菌Ag85B-ESAT6融合蛋白稳定表达细胞系的建立

建立了能够稳定表达结核分枝杆菌Ag85B-ESAT6融合蛋白的P815细胞系。将Ag85B和ESAT6基因分别克隆至真核表达载体pcDNA3,构建了Ag85B—ESAT6融合蛋白的真核表达质粒Ag85B-pcDNA3-ESAT6。在阳离子聚合物作用下,重组质粒转染与BALB／c遗传背景一致的P815（H-2^4）细胞。通过G418压力筛选后,得到1株阳性克隆细胞。经过RT-PCR检测到该细胞中有Ag85B-ESAT6融合蛋白mRNA表达,用间接免疫荧光可以在转染重组质粒的P815细胞膜上检测到较强的绿色荧光,证实P815细胞内有融合蛋白的表达。获得表达Ag85B-ESAT6融合蛋白的稳定细胞系。     

表达HIV-1复合多表位基因的p815细胞稳定株的建立和评价

建立稳定表达HIV-1p24MEG复合多表位基因的p815细胞克隆.设计引物,以HIV-1标准株全长cDNA序列为模板,PCR扩增获得p24基因片段;合成改造后的多个表位基因MEG并且与p24片段相连接,克隆入pcDNA3.1(+).在阳离子聚合物作用下重组真核质粒转染的p815(H-2d)细胞, 以G418压力筛选,RT-PCR检测mRNA表达,间接免疫荧光检测蛋白表达.通过PCR获得了HIV-1 p24片段,获得了与多表位基因连接后的p24MEG融合基因,成功构建了p24MEG基因的重组真核表达载体pcDNA3.1(+)/p24MEG.转染p815细胞后, RT-PCR检测到融合蛋白mRNA表达,间接免疫荧光显示 p815细胞内有融合蛋白的表达.结论:建立了稳定表达HIV-1p24MEG融合蛋白的p815细胞克隆,为评价多表位抗原p24MEG在BALB/c小鼠体内诱导的细胞免疫应答奠定基础.     

结核分枝杆菌HSP65-hIL-2融合基因稳定转染细胞系的建立

建立可稳定表达结核分枝杆菌HSP65-hIL-2融合蛋白的稳定转染P815细胞系. 在阳离子脂质体作用下,将HSP65-hIL-2真核表达质粒转染与BALB/c 遗传背景一致的P815细胞(H-2d).G418筛选阳性克隆,RT-PCR和间接免疫荧光法检测目的蛋白的转录和表达. 阳性克隆细胞经RT-PCR检测到HSP65-hIL-2融合基因特异性的mRNA表达;用鼠抗人的IL-2 mAb进行间接免疫荧光检测,可在转染的P815细胞浆中观察到较强的绿色特异性荧光,而未转染细胞则为阴性.成功获得稳定表达HSP65-hIL-2融合蛋白的稳定转染细胞系,为其疫苗的CTL研究提供了合适的靶细胞.     

人spindlin 1基因全长cDNA的克隆与定位

利用分子克隆技术和生物信息学手段,克隆并命名了人spindlin 1 基因. 此基因cDNA全长4375?bp,含有236～949 bp完整开放读框,预测编码27?ku 的膜内蛋白. 新基因在核酸序列上与鼠spindlin基因有96%同源,其编码氨基酸序列与鼠spindlin蛋白98%同源. 生物信息学分析表明该基因定位于9q22.1-22.3 区域, 基因组DNA全长为52.3?kb,由5个外显子和4个内含子组成,内含子和外显子之间的剪切完全符合GT-AG法则. 利用绿色荧光蛋白与spindlin 1基因的融合蛋白表达载体转染COS-7细胞表明spindlin 1表达的蛋白定位于胞核,且转染后的细胞中较多见胞核形态的改变,例如出现双核或巨型核.     

海岛棉ERF族转录因子EREB6基因克隆特征分析

中段密码子序列对目的蛋白在大肠杆菌中表达的影响

通过调整表达翻译肽链中段Asp—Pro—Pro三个氨基酸的密码子的序列,观察其对目的蛋白在大肠杆菌中表达的影响。用PCR方法分别在rhBMP4-m基因的5’端加上编码Asp-Pro—Pro酸水解肽的不同简并码序列,克隆入原核融合表达载体pRSET中进行诱导表达,SDS—PAGE分析,观察融合蛋白的表达效果。成功构建9个不同简并码序列的融合表达载体;经诱导表达分析,只有在两个脯氨酸的密码子均为CCG时,融合蛋白才能正常表达,说明改变表达肽链中段氨基酸密码子的碱基序列可以影响融合蛋白的表达效果。     

人白细胞介素32的真核表达、纯化及生物学活性鉴定

摘要:目的：构建人白细胞介素32（IL-32）和IgG4基因Fc段的融合基因真核表达载体IL-32-IgG4(Fc)-pOptiVEC,建立稳定转染的中国仓鼠卵巢细胞（CHO/DG44）克隆,并检测重组蛋白的表达及相应的生物学功能.方法：采用聚合酶链反应(PCR)从LPS激活的人CD4+T细胞cDNA中扩增出IL-32基因,并克隆到构建IL-32融合基因的真核表达载体 IL-32-IgG4(Fc)-pOptiVEC.经脂质体法转染CHO/DG44 细胞, 通过RT-PCR检测,筛选出实验组细胞,并对筛选的阳性克隆细胞再进行氨甲喋呤（MTX）加压筛选,利用ProteinG-Agarose亲和纯化培养上清中表达的融合蛋白,并通过SDS-PAGE、免疫印迹鉴定检测目的蛋白的表达,最后以 HeLa细胞为靶细胞测定重组蛋白的生物学活性.结果：构建了一个IL-32融合基因真核表达载体IL-32-IgG4(Fc)-pOptiVEC;筛选出了能稳定表达IL-32-IgG4(Fc) 融合蛋白的CHO/DG44细胞克隆; 亲和纯化后的IL-32-IgG4(Fc)融合蛋白在SDS-PAGE电泳后与预期分子质量大小相符,能够与羊抗人IgG-HRP 抗体特异结合,并能促进HeLa细胞的死亡.结论：成功构建了人IL-32融合基因真核表达载体IL-32-IgG4(Fc)-pOptiVEC,获得了能够表达具有生物学活性的IL-32-IgG4(Fc) 融合蛋白的CHO/DG44细胞克隆.     

棉花GhGAI1基因克隆及其功能的初步分析

DELLA蛋白是赤霉素信号通路中重要的蛋白质作用因子,负调节植物体中赤霉素的水平,同时受到生长素、脱落酸和乙烯的共同调节。根据已知的生物信息数据,本研究克隆了棉花GhGAl1基因,序列分析表明它编码DELLA蛋白。构建GFP与GhGAl1融合蛋白重组质粒,利用根癌农杆菌介导的花浸泡转基因方法转化拟南芥columbia生态型植株,进行过量表达实验。结果表明,GFP—GhGAl1融合蛋白定位于细胞核并且在施加赤霉素后迅速降解,表明该基因编码蛋白参与赤霉素信号通路。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。

橡胶颗粒沥青混合料低温抗裂性试验研究

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.
