The amino acid sequence was compared among the three allelic variants (allozymes) of sn-glycerol-3-phosphate dehydrogenase in D. virilis, which are detected by one-dimensional electrophoresis. The alpha GPDHf variant was different from the alpha GPDHm by only one substitution of 68-lysine for asparagine; alpha GPDHs differed from alpha GPDHm by substitution of 127-glycine for arginine. No electrophoretically 'silent' substitutions were found in a total of 352 amino acid residues.  相似文献

Latitudinal clines of malate dehydrogenase-1 (MDH-1) allozymes occur within honey bee populations on three continents: Europe, North America and South America. The North and South American populations are introduced and demonstrate that Mdh allelic clines were established within the last 150 years. The frequency of the medium electrophoretic allele increases in frequency with increasing latitude while the fast allele decreases with latitude on all the three continents. The clines are best explained by the average daily high temperature for July on all continents. These parallel clines provide evidence for selection on Mdh alleles in honey bees.  相似文献

Summary Aldehyde dehydrogenase (ALDH) activity is demonstrated in four strains ofD. melanogaster lacking active alcohol dehydrogenase (ADH-null mutants). In the four strains, ALDH activities are similar to those found in a wild strain. It is concluded that ADH-null flies are able to detoxify acetaldehyde. This finding is discussed in relation with the dual function of ADH proposed recently.This work was supported by a grant from the Medical Research Council of Canada (MRC 6920) to Dr F. Summary Proline is accumulated by overwinteringC. amonea larvae. Mortality averages over 50% by spring. PGMF in the population shifts from 70% in summer to less than 50% during winter, increasing again in summer. Product synthesis from stored glycogen may be mandated in dipteran larvae also using larval proteins for cold hardiness when completion of development is delayed until spring.Thanks are gratefully extended to Derek Lamport and Patrick Muldoon of the MSU/DOE Plant Biology Laboratory for the amino acid analyses. Summary A regulatory element tightly linked to theGpdh locus inDrosophila melanogaster has been isolated from a natural population. Flies homozygous for second chromosomes bearing the element,H31, have half the GPDH activity of normal homozygotes. Heterozygotes betweenH31 andF orS alleles exhibit dominance in GPDH activity. Heterozygotes betweenH31, F orS andDf(2L) GdhA have half the diploid level. The contribution of theS allele to the activity inS/H31 heterozygotes is more than four times that ofH31. Summary The effect of differentRhizobium loti strains on the biosynthesis of 2,3-diamino-butanoic acid and 2,4-diamino-3-methyl-butanoic acid in root nodules ofLotus tenuis, Anthyllis vulneraria andLupinus densiflorus has been investigated. Results suggest that biosynthesis isRhizobium strain dependent, that the bacteroid is the site of synthesis of the compounds and that their biosynthesis is confined to the symbiosis.  相似文献

Racemic 4,4,4-trifluoro-3-(indole-3-)butyric acid (TFIBA) has been synthesized and shown to inhibitAvena coleoptile elongation. (S)-(+)-TFIBA (fig. 1), which was prepared by an enzymatic method and markedly promotes root growth of Chinese cabbage, lettuce and rice plants, is a novel fluorinated plant growth regulator. Activity of the (S)-(+)-enantiomer of TFIBA was 10-fold greater than that of the (R)-(–)-enantiomer in the first two plant species and 5-fold greater in rice.  相似文献

To investigate the peculiar ionic specificity of KAAT1, an Na+- and K+-coupled amino acid cotransporter from Lepidoptera, a detailed analysis of membrane topology predictions was performed, together with sequence comparison with strictly Na+-dependent mammalian cotransporters from the same family. The analysis identified aspartate 338, a residue present also in the other cotransporter accepting K+ (CAATCH1), but absent in most mammalian transporters that have, instead, an asparagine in the corresponding position. Mutation of D338 in KAAT1 led either to non-functional transporters (D338G, D338C), or to an altered ionic selectivity (D338E, D338N), observable in uptake experiments and in electrophysiological properties. In particular, in D338E, the transport activity, while persisting in the presence of Na+, appeared to be completely abolished in the presence of K+. D338E also showed uncoupling between transport-associated current and uptake. The opposite mutation in the -aminobutyric acid transporter rGAT-1 (N327D) resulted in complete loss of function. In conclusion, aspartate 338 in KAAT1 appears to be important in allowing K+, in addition to Na+, to drive the transport mechanism, although other residues in different parts of the protein may also play a role in the complete determination of ionic selectivity.Received 23 September 2003; received after revision 11 November 2003; accepted 25 November 2003  相似文献

Summary A new natural substance has been identified in the rectal ampullae of gall-forming fruit flies. The substance was found to be the only volatile compound in the rectal ampulla of maleUrophora cardui andUrophora stylata. GC-MS methods were used to characterize its structure as 4-methyl-3Z,5-hexadienoic acid. Physiological parameters such as the amount of the acid at different ages and under different conditions were investigated. The biological significance of the new volatile as an arresting pheromone was tested in several bioassays. The arrestant function could not be established, but the results gave hints of a territorial function between conspecific males. The results are discussed with respect to gland morphology and predictions of communication models among fruit flies.  相似文献

Summary (16-D3)palmitic acid was applied topically to pheromone glands of the turnip moth. After incubation omega labeled analogues of fatty acid methyl esters and acetates were identified by gas chromatography with flame ionization detection and selected ion monitoring. The pheromone components (Z)-5-decenyl, (Z)-7-dodecenyl- and (Z)-9-tetradecenyl acetate were all biosynthesized from palmitic acid along a common pathway.  相似文献
