The variety of encryption mechanism and algorithms which were conventionally used have some limitations. The kernel operator library based on Cryptographic algorithm is put forward. Owing to the impenetrability of algorithm, the data transfer system with the cryptographic algorithm library has many remarkable advantages in algorithm rebuilding and optimization, easily adding and deleting algorithm, and improving the security power over the traditional algorithm. Library function call sequence is the direct reflection of a program's behavior. The relationship between program vulnerability and library calls is analyzed, and an intrusion detection method via library calls is proposed, in which the short sequences of library call are used as signature profile. In this intrusion detection method, library interposition is used to hook library calls, and with the discussion of the features of the library call sequence in detail, an algorithm based on information-theory is applied to determine the appropriate length of the library call sequence. Experiments show good performance of our method against intrusions caused by the popular program vulnerabilities.  相似文献

A pig BAC library was constructed with genomic DNA from a male Erhualian pig. After partial digestion with Hind III or BamH I the fragments obtained were cloned into the pBeloBAC11 vector. The library consists of 184320 clones which stored in 480 pieces 384-well plates (20 plates per superpool). A two-step 4-dimension PCR screening system was established to screen the positive clones. An average insert size of 128 kb was estimated from 105 randomly isolated clones, which indicates that the library is more than five times of genomic coverage. For the demonstration of the probability to pick out any unique genes or DNA markers from the library, 10 single-copy genes were screened out and the positive clones were yielded between 1 and 8 with an average of 3.6. Positive superpools were obtained for 32 microsatellite markers selected from different regions of pig genome. The number of positive superpools for each marker varies from 1 to 9 with an average of 4.78. This BAC library provides an additional resource for pig physical mapping and gene identification.  相似文献

Of ICE protease family, CPP32 (apopain or Yama) plays a central role in different apoptotic pathways. To study the molecules regulating CPP32, yeast two_hybrid system was used to identify proteins (peptides) that interact with CPP32. First, the CPP32 gene was cloned into plasmid vector pGBT9. The resulting recombinant plasmid was designated as pGBT9/CPP. The pGBT9 /CPP plasmid was transformed into the yeast strain HF7C, then the leukemia library was introduced. The transformation mixture was plated on medium lacking Trp, Leu and His in the initial screen. Colonies growing on the selection medium were further assayed for β galactosidase activity. Within 42 Trp +Leu +His + colonies only 5 turned blue in the presence of X_Gal. Plasmid DNA from 5 positive yeast colonies was prepared respectively and used to transform HB101 by electroporation. The transformation mixture was plated on medium lacking Leu to be selected for the library plasmid. Finally, only one library plasmid, designated as pY1, was determined to be truly positive by retransformation of pGBT9/CPP and the library plasmid into HF7C. The inserted cDNA of pY1 encodes a peptide of 15 amino acids, suggesting that it may be the domain interacting with CPP32.  相似文献

To examine the community structure of β-Proteobacterial ammonia-oxidizing bacteria (AOB) in prawn farm sediment, the 16S rRNA gene library was constructed with β-Proteobacterial AOB-specific primers. The library was screened by PCR-restriction fragment length polymorphism (RFLP) analysis and clones with unique RFLP patterns were sequenced. Two groups of β-Proteobacterial AOB, the Nitrosomonas and the Nitrosospira, were detected. The Nitrosomonas occupied an absolute dominant position, accounting for more than 90% of total clones in the clone library, while the Nitrosospira accounting for 5.48%. Nitrosomonas-affiliated clones were grouped into the Nitrosomonas marina and the Nitrosomonas sp. Nm143 clusters, and Nitrosospira-affiliated clones were grouped into the Nitrosospira cluster 1. No other clusters of β-Proteobacterial AOB were found. The results enriched our knowledge of AOB diversity in the prawn farm sediment and provided important foundational data for further functional studies of these microbes in mariculture environments.  相似文献

Thirty-two C-genome specific candidate bacterial artificial chromosome (BAC) clones were successfully screened from the BAC library by four-dimensional PCR method with the primer pairs of 75 simple sequence repeat (SSR) markers located in the nine C-genome linkage groups of Brassica napus. The screened 32 BAC clones have an average insert size of 114.2 kb with a range of 30-190 kb. They are the first set of C-genome BAC clones screened from B. napus genomic BAC library. The average insert size of this set of BAC clones presented that the constructed BAC library had a high quality. This set of BAC clones can be used as markers to identify individual chromosomes of B. napus C-genome.  相似文献

This report briefly describes the construction and characterization of a peach [ prunus persica (L.) Batch] Var. Jingyu bacterial artificial chromosome (BAC) library. The variety Jingyu has many important agronomic characters of stone fruits, and it is a main parent in Chinese peach breeding. After cloning of the high molecular weight peach DNA into pBeloBAC 11, we obtained over 22 000 recombinant clones. The BAC library has an average insert size of 95 kb and represents approximately 7 times peach haploid genome equivalents. After being screened with two randomly amplified polymorphic DNA markers, W4 and P20, which are linked to yellow flesh and nectarine genes of peach respectively, ten positive clones have been detected. This library is very useful for map-based cloning of peach genes and physical mapping of peach genome.  相似文献

A large human naive single chain antibody (scFv) library is constructed from 60 healthy donors via phage display technique.During the period,some methods are employed to optimize the diversity,such as multi donors,different annealing temperature,half-nest PCR.and assembly by two-way fusion PCR.In this study,78 electroporations resulted in 1010 library,diversity of which is assayed by enzyme fingerprint.The efficiency and diversity are all better than other researches.  相似文献
