Summary The early large birefringence signal and mechanical activity were studied together in isolated single fibres of frog skeletal muscle with double stimulation at short stimulus intervals (2–60 msec) at room temperature and at 4–6°C. In all fibres tested, extra tension and additional birefringence signal in response to the second stimulus appeared simultaneously and suddenly upon increasing the stimulus interval. The shape of the stimulus-interval versus tension-development curve makes it highly improbable that subthreshold calcium release occurs at shorter stimulus intervals; therefore, tension development reliably reflects Ca-release in these experiments. In contrast to the report by Suarez-Kurtz and Parker, birefringence signal and calcium release are shown not to be dissociated by double stimulation. This result supports the hypothesis that the early large birefringence signal is an intrinsic indicator of calcium release from the sr during EC-coupling in skeletal muscle. 相似文献

Summary In detergent-treated cardiac muscle fibers, forskolin, a potent activator of adenylate cyclases, inhibits tension development elicited with submaximal [Ca2+] and increases incorporation of32P into troponin-I. A similar reduced tension development has been observed after treatment with cAMP or the catalytic subunit of the cAMP-dependent protein kinase. It is concluded that these fibers still contain much of the enzyme cascade involved in evoking a contractile response to \-adrenergic stimulation.Acknowledgment. The authors thank Dr H. Exposure of voltage-clamped frog skeletal muscle fibres to silver caused a maintained inward current which could be carried by Ca2+, Mg2+ or Na+. Inorganic Ca2+ channel blockers and dithiothreitol (SH reducing agent) diminished this current, but a Na+ channel blocker did not. Summary Autoradiographic, stereological and histological studies have been carried out to determine the origin of muscle fibre splitting which supposedly occurs during muscle hypertrophy. The results obtained clearly indicate that the supposedly split fibres are a transient response probably derived from satellite cells and are not derived from pre-existing fibres by true splitting. Similarly, increases in muscle fibre size are not achieved by recruitment of satellite structures as indicated by lack of myonuclear recruitment.Acknowledgment. The excitation-contraction (E-C) coupling process in single twitch fibres from frog toe muscle was inhibited selectively by phenylglyoxal (PGO), a specific guanidyl modifying reagent. A new protein (31.5 kDa), which has PGO-binding ability and seems to play a key role in the E-C coupling process, was solubilized from transverse tubule membrane-junctional sarcoplasmic reticulum complexes (TTM-JSR) of frog skeletal muscles, using14C-PGO. The monoclonal antibody against this protein applied extracellularly inhibited the E-C coupling process of the single fibres. This protein appears to constitute the very first step of input for E-C coupling. It is considered to behave as an indispensable part of an electrometer to measure membrane potentials. Therefore, the name electrometrin is suggested for the new protein. 相似文献
