人骨唾液酸蛋白在巴斯德毕赤酵母中的高效分泌表达

以PCR方法扩增人bsp基因，与分泌型表达载体pPICZαA重组，重组质粒经电击转化毕赤酵母GS115菌株，获得的GS115／pPICZαA－hbsp表达菌株能高效分泌表达rhBSP.SDS-PAGE和Western blot分析结果表明，产物的分子质量接近66ku，能发生特异性抗原－抗体反应。     

两种杂合抗菌肽在毕赤酵母中的表达及活性测定

参照毕赤巴斯德酵母(Pichia pastoris)的偏好密码子,改造克隆杂合抗菌肽CA(1～7)M(4～11)和CB(1～7)M(4～11)基因,将改造后的基因克隆到pPICZα-A 载体中,构建分泌型表达载体pPICZα-A-CAM和pPICZα-A-CBM,转化宿主菌Pichia pastoris X-33,在甲醇的诱导下进行表达.实验结果表明,杂合抗菌肽获得表达,两种表达产物具有明显的抗菌活性.此外,本文还比较了二者的抗菌谱,分析了杂合肽的性质,优化了酵母电转化条件.     

Neuritin毕赤酵母分泌表达系统的构建

为构建Neuritin毕赤酵母分泌表达系统,进一步得到高纯度的具有天然活性的Neuritin蛋白提供实验基础。采用人工合成去除信号肽的neuritin基因并在N端引入His标签序列,两端引入Eco RⅠ和NotⅠ酶切位点,克隆至p PIC9K分泌型表达载体。重组质粒经PCR与测序鉴定正确后电击转化至毕赤酵母GS115感受态细胞中,经MD平板与G418平板筛选阳性重组子,抽提毕赤酵母基因组PCR鉴定重组质粒的插入。结果显示,p PIC9K-neuritin重组质粒经测序鉴定完全正确,电转后成功插入毕赤酵母基因组中。由此可知,成功构建Neuritin的毕赤酵母分泌表达系统。     

大黄鱼与美国红鱼抗菌肽Piscidin串联基因的构建、酵母表达及抗菌活性鉴定

采用Piscidin基因串联表达的方案,通过Eco31Ⅰ限制性内切酶定向连接方法合成Pseudosciaena crocea-Sciaenops ocellatus-piscidin (PSP)串联基因.以毕赤酵母(Pichia pastoris)的穿梭表达载体pPICZαA构建重组表达质粒pPICZαA-PSP,转化入毕赤酵母SMD1168菌株中.应用实时荧光定量PCR方法进行多拷贝克隆筛选,实现了PSP重组串联肽的诱导表达和纯化,并对其抑菌活性进行初步鉴定,为piscidin抗菌肽的生产应用奠定基础.     

共表达鸡IBDV VP2和IL-2基因的重组毕赤酵母的构建及免疫原性

通过重叠延伸PCR扩增IL2-VP2串联基因,克隆到pMD18-T载体上,亚克隆正向插入到毕赤酵母表达质粒pPICZαA中,经PCR和双酶切鉴定表明成功构建了酵母重组表达载体pPICZαA-IL2-VP2。将该重组质粒线性化后电击转化入感受态毕赤酵母X-33菌株,构建成分泌型表达IL2-VP2的酵母工程菌株。经甲醇诱导后,通过SDS-PAGE、Western-blot活性检测分析表明IL2-VP2基因长度为1.85kb,包含缺失了TAA终止密码子的IL-2基因和缺失了ATG起始密码子的VP2基因,两个基因之间以高度柔韧性的(Gly-Gly-Gly-Ser)编码核苷酸相连接,并在该体系中得到分泌表达,分子量约为50kD,且表达产物对传染性法囊病毒具有较好的反应原性。     

纳豆激酶基因在巴斯德毕赤酵母中的表达

构建pPICZαA-NK重组质粒，并转化入E.coli TOP-10中，得到转纳豆激酶基因工程菌，提取重组质粒经单酶切，双酶切，PCR分析及序列测定，证明克隆到载体pPICZαA上的外源基因即为纳豆激酶基因，将重组质粒pPICZαA-NK线性化后，分别转化毕赤酵母GS115与KM71，在含Zeocin^TM的选择性YEPD平板筛选到重组酵母，经检测重组酵母发酵上清液中具纳豆激酶溶纤活性，SDS-PAGE电泳结果表明外源蛋白的表达量占菌体蛋白的10%左右。     

天蚕抗菌肽B在毕赤酵母中的表达

Cecropins B是一种昆虫抗菌肽,具有分子量小,热稳定性、水溶性好、抗菌谱广等优点,更为重要的是抗菌肽对真核细胞几乎没有作用,仅仅作用于原核细胞和发生病变的真核细胞,是目前已知天然抗菌肽中活力最强的一种。根据毕赤氏巴斯德酵母(Pichia pastoris)偏好密码子,改造并化学合成天蚕素基因B,克隆到pPIC9K载体中,构建分泌型重组酵母表达载体CB-pPIC9K,转化Pichia pastoris受体菌KM71,在醇氧化酶(AOX)启动子调控下,Cecropin B获得表达,分子量约3.8KD,摇瓶发酵产率可达到200μg/mL,并对其抗菌活性进行了初步测定。结果表明我们表达的重组Cecropins B对G-菌有较好的抑菌活性,且具有较好的热稳定性。这些特点使得重组抗菌肽Cecropins B在食品防腐、疾病防治和动物饲料添加剂等方面显露出很好的应用前景。     

胸腺肽α1在毕赤酵母中的表达及纯化

采用基因工程菌发酵制备胸腺肽α1(Tα1).将人工合成的Tα1单体基因,连接到pPIC9K质粒上,构建表达载体pPIC9K-Tα1,转化毕赤酵母GS115,重组菌经甲醇诱导表达,取发酵液上清,过DEAE52、Sephade-xG-25柱纯化后获得目的蛋白.Tricine-SDS-PAGE和HPLC结果表明Tα1基因在毕赤酵母中得以表达,表达量为4.8mg/L.     

小鼠LEAP-2基因的克隆及其真核表达质粒的构建

从小鼠肝中提取总RNA,采用反转录聚合酶链式反应（RT-PCR）方法,获得了mLEAP-2基因编码区的cDNA,扩增出小鼠肝表达抗菌肽-2（mLEAP-2）成熟肽基因片段,重组入克隆载体pUCm-T,经DNA测序,该基因为120 bp,编码40个氨基酸。用限制性内切酶切下目的基因,插入毕赤酵母表达载体pPIC9中,构建成表达载体pPIC9-LEAP-2。重组毕赤酵母表达载体pPIC9-LEAP-2的结构,可望获得超量表达的高活性肝表达抗菌肽-2,为研制具有抗菌活性的新型基因药物奠定基础。     

重组人角质细胞生长因子(KGF-2)在毕赤酵母中的克隆与表达

利用RT－PCR技术，从体外培养的人胚胎成纤维细胞中扩增出人角质细胞生长因子（KGF－2）的cDNA及基因序列，并将其基因序列克隆入质粒pGEM-T Easy中，经测序与文献报道序列一致，将KGF－2基因切出后定向插入酵母分泌性表达质粒pPICZαA中，转化毕赤酵母菌株GS115，获得的重组体经甲醇诱导可表达出人角质细胞生长因子KGF－2，表达的KGF－2经Ni^2 柱亲和层析纯化后，有很好的生物学活性。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. 低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
