Reduced collagen biosynthesis in oral mucosa of beige (Chediak-Higashi) mice

Summary Oral mucosa of beige (Chediak-Higashi) mice had decreased levels of collagen synthesis and prolyl hydroxylase activity compared with normal animals. No significant difference was observed in non-collagen protein synthesis between the two groups. These results suggest that decreased collagen biosynthesis in oral tissues may be partially involved in the increased incidence of periodontal disease in the Chediak-Higashi syndrome.     

Fluorescence microscopical study of 5-hydroxytryptamine storage organelles in mepacrine-incubated blood platelets of beige mice (Chediak-Higashi syndrome)

Summary The number and fluorescence intensity of fluorescent granules (5-HT storage organelles) of mepacrine-incubated blood platelets of beige mice (Chediak-Higashi syndrome) are decreased compared to those of control mice platelets. This indicates both quantitative and qualitative changes of the 5-HT organelles, namely a reduced number and a reduced storage capacity, respectively.     

Imparied bone resorption of cultured calvaria from mice with abnormal lysosomal function (the Chediak-Higashi syndrome)

Summary Spontaneous bone resorption is reduced in cultured calvarial bones from mice with the Chediak-Higashi syndrome, as indicated by decreased mobilization of calcium from the bones to the medium. Although bone resorption in calvaria from mice with this disease can be stimulated by PGE₂ and 1 (OH)D₃, the amounts of mineral released after stimulation is also decreased.This investigation was supported by grants from the Swedish Association against Rheumatic Diseases, from the Royal 80 Year Fund of King Gustav V, and from the Swedish Medical Research Council (project No 14X-05426).     

Insulin secretion and (pro)insulin biosynthesis of cultured mice islets after glibenclamide loading in vitro

Summary Isolated islets from C57B1/6J mice exposed to 10 mmoles/1 glucose supplemented with 5 g/ml glibenclamide for 48 h released significantly less insulin in the subsequent short-time incubation thanuntreated controls (without glibenclamide), whereas insulin biosynthesis was not suppressed by glibenclamide treatment.Investigations were carried out as a part of the ForschungsprojektDiabetes mellitus und Fettstoffwechselstörungen, supported by the Ministry of Health of the GDR.

Reduced fertility in male mice following treatment with niridazole

Zusammenfassung Nach einmaliger i.p.-Injetion von 700 mg/kg Niridazol wurden männiiche Mäuse wiederholt mit unbehandelten Weibchen wöchentlich über 2 Monate hinweg gepaart und die Ova gesammelt. Es konnte eine starke Herabsetzung der Befruchtungsraten 3–4 Wochen nach Paarung festgestellt werden, was die Annahme einer Wirkung auf die meiotische Teilung der Spermatozyten und Spermatiden nahelegt. Nach der 7. Woche trat eine Normalisierung der Fruchtbarkeitsrate ein.

Adsorption of hageman factor (factor XII) on collagen

Résumé Nous avons trouvé que le facteur Hageman (factor XII), contenu dans le plasma humain citraté, pauvre en plaquettes, est adsorbé presque sélectivement par le collagène. Il est également possible d'éluer ce facteur au pH alcalin.     

Fibrin stabilizing factor (F XIII) and collagen polymerization

Summary Factor XIII induces a decrease and a modification in the collagen polymerization, probably due to the formation of transamidation bonds. This property has some importance in wound healing.     

Cyanobacterial symbiont biosynthesis of chlorinated metabolites fromDysidea herbacea (Porifera)

The tropical marine spongeDysidea herbacea contains large numbers of a symbiotic filamentous cyanobacterium identified on the basis of a detailed ultrastructural study asOscillatoria spongeliae. We report the flow-cytometric separation of the symbiont from the sponge cells, and demonstrate by chemical analyses that a unique group of polychlorinated compounds isolated from the whole sponge tissue is limited to the cyanobacterial filaments, whereas the accompanying sesquiterpenoids are found only in the sponge cells. This is the first demonstration that secondary metabolites ascribed to a sponge are localized in prokaryotic symbiont cells.     

Stabilisation of collagen by betel nut polyphenols as a mechanism in oral submucous fibrosis

Treatment of reconstituted collagen fibrils and pieces of rat dermis with the crude extract, purified tannins or (+)-catechin from betel nut (Areca catechu) increases their resistance to both human and bacterial collagenases in a concentration-dependent manner. These tanning agents may stabilise collagen in vivo following damage to the oral epithelium, and promote the sub-epithelial fibrosis which occurs in betel nut chewers.

Influence of oral glucose feeding on endotoxin lethality in mice

Occurrence and biosynthesis of 11 (R)-hydroxy-eicosatetraenoic acid (11-R-HETE) in the Caribbean soft coralPlexaurella dichotoma

Gorgonian soft corals from the Caribbean Sea are known to contain prostaglandin-like compounds as well as other products of arachidonic acid lipoxygenation, and the formation of the latter has been suggested to represent the first step in prostaglandin biosynthesis. Here we present evidence for the presence of 11-R-hydroxy-5Z, 8Z, 12E, 14Z-eicosatetraenoic acid (11-R-HETE), as well as of the enzyme responsible for its biosynthesis, in the Caribbean gorgonianPlexaurella dichotoma. Lipid extracts fromP. dichotoma were purified by conventional SiO₂ column chromatography followed by reversed phase high-pressure liquid chromatography (HPLC). These yielded a component having chromatographic and spectroscopic properties identical to synthetic 11-HETE. Electron impact mass spectrometric analysis of the acetoxy-, methyl ester derivative of the compound confirmed its identity as 11-HETE, while chiral phase HPLC of the methyl ester derivative showed that the stereochemistry of the alcoholic carbon atom wasR. Enzymatically active homogenates fromP. dichotoma were able to convert both unlabelled and [³H] arachidonic acid into 11-HETE. In vitro biosynthesis of the latter metabolite was also observed with homogenates of the Mediterranean gorgonianParamuricea clavata, another non-prostaglandin-containing soft coral, thus suggesting that 11-R-HETE production is not necessarily accompanied by prostaglandin formation in gorgonian corals.
