纳米金免疫层析法定量检测尿RBP

应用纳米金免疫层析技术(双抗夹心法)和柠檬酸三钠还原法建立一种对尿RBP的快速定量检测方法,并在此基础上研制出快速检测试纸条.该试纸条用纳米金免疫层析定量分析仪可在15 min内实现RBP定量检测,检测限为150μg/L(相当于150 ng/mL),检测范围为150～5000μg/L.与尿微量白蛋白(ALB)、转铁蛋白(TRF)、β2-微球蛋白(β2-MG)、尿纤维连接蛋白(FN)、溶菌酶(LZM)等肾脏疾病标志物无交叉反应.该方法特异性强、检测灵敏度高、范围广、简便快捷,在近端肾小管的损伤、糖尿病肾病等肾脏疾病的早期诊断及过程监控中具有较为广泛的临床应用.     

盐酸克伦特罗快速检测试纸条的应用探讨

采用免疫层析快速检测试纸条,检测盐酸克伦特罗标准品及各种猪尿样本,并与酶联免疫检测方法(ELISA)进行比对,结果表明,试纸条检测结果与ELISA符合性较好,适用于盐酸克伦特罗的现场快速检测.     

基于顺磁微粒的金黄色葡萄球菌肠毒素B免疫层析试纸条的制备

采用基于顺磁微粒标记抗体的免疫层析法,制备金黄色葡萄球菌肠毒素B(SEB)快速定量检测的层析试纸条。利用EDC和NHS交联活化顺磁微粒,对顺磁微粒包被抗体的体系进行优化;根据双抗夹心法原理制备免疫层析试纸条,对制备的SEB免疫层析试纸条进行测试研究。优化的顺磁微粒偶联抗体的缓冲体系为p H 9.0,5mmol/L硼酸,0.05%Tween-20;磁珠包被抗体量为125μg/2mg磁珠,封闭剂为0.25%的脱脂牛奶,磁珠保存液为5mmol/L硼酸、0.05%Tween-20+0.05%Triton X-100,0.01%Na N3,0.5%BSA,p H9.0;建立的SEB顺磁微粒免疫层析试纸条T线抗体浓度为1mg/m L,最低检测限为1ng/m L,磁信号检测显示,该试纸条在SEB浓度0.1~1 000ng/m L范围内具有很好的线性关系。SEB顺磁微粒免疫层析法具有简单快速、灵敏度高、可定量的特点,可应用于SEB的快速检测。     

基于适配体的胶体金色谱层析试纸条快速检测肌酸激酶同工酶

研究基于适配体的胶体金色谱层析试纸条快速检测肌酸激酶同工酶,用于揭示长期飞行下航天员潜在的心肌和肌肉损伤.利用SELEX方法筛选得到的肌酸激酶同工酶适配体,并优选出与肌酸激酶同工酶有较强亲和能力的核酸适配体应用于胶体金免疫层析试纸条,通过优化胶体金颗粒大小、缓冲液浓度等条件,成功建立了一种基于适配体的胶体金试纸法用于CK-MB的快速检测.实验结果表明,通过基于磁珠的核酸适配体筛选方法筛选出的与肌酸激酶同工酶有亲和力的核酸适配体,能够用于构建适配体-适配体夹心胶体金免疫层析试纸条并成功应用于82 nM以上的肌酸激酶同工酶的快速、可视化检测,同时,该免疫层析试纸条具有良好的特异性和稳定性,可用于未来空间医学即时检测.     

鸡传染性支气管炎病毒抗体胶体金免疫层析试纸条的研制

[目的]建立一种快速、灵敏的胶体金免疫层析法(Colloidal-gold immunochromatography assay),用以检测禽类早期感染鸡传染性支气管炎病毒(Infectious bronchitis virus,IBV)和监测IBV疫苗接种免疫水平.[方法]将重组IBV核衣壳(Nucleocapsid,N)蛋白包被在在试纸条检测线,羊抗兔IgG包被在试纸条控制线,用胶体金-兔抗鸡IgG偶联物作为示踪剂,检测血清中IBV抗体.[结果]制备所得的胶体金免疫层析试纸条与IBV抗体阳性血清反应呈阳性,与鸡新城疫病毒抗体阳性血清、鸡传染性法氏囊病毒抗体阳性血清、鸡毒霉形体抗体阳性血清和SPF标准鸡血清未发生交叉反应;当IBV抗体阳性血清以1:35比例稀释后,依然可以用该试纸条检出;该试纸条在60 d的保存期内重复性良好;以从不同养殖场采集到的鸡血清样本作为测试对象,比较该试纸条和进口 ELISA试剂盒的检测结果,两者符合率为97.44％.[结论]成功制备了检测耗时短、检测限低、特异性强、稳定性好的IBV抗体检测胶体金免疫层析试纸条,为鸡传染性支气管炎的早期诊断以及免疫监测提供了简便高效的技术方法.     

金黄色葡萄球菌肠毒素B的原核表达、纯化及鉴定

为高效获取金黄色葡萄球菌肠毒素B(SEB)用于SEB的快速检测,通过原核表达方法表达SEB,对其纯化方法进行研究,并通过Western-blot和制备的SEB纳米磁性免疫层析快速检测试纸条检测其抗原性。研究表明:SEB成功克隆入原核表达载体pET22b(+),并在E.coil BL21(DE3)中得到有效表达,表达量达65.8%;采用Ni柱亲和层析和DEAE阴离子层析两步纯化得到纯度为95.2%的SEB,Western-blot和SEB免疫层析快速检测试纸条检测显示所纯化的重组SEB具有SEB的抗原性,可用于SEB快速检测试纸的应用研究。     

辣椒、花椒中罗丹明B免疫层析分析

为建立以胶体金标记抗体为基础的罗丹明B(RB)快速检测技术,利用戊二醛法将RB的衍生物罗丹明123(R123)与牛血清白蛋白(BSA)、卵清白蛋白(OVA)分别合成完全抗原(R123-BSA)和包被抗原(R123-OVA),对雌性大耳白兔皮下注射R123-BSA获得多克隆抗体.胶体金作为示踪标记物标记到硝酸纤维素膜(NC膜)上,涂覆R123-OVA作为检测线,用羊抗兔IgG作为质控线,通过优化实验条件,开发了渗滤式胶体金免疫快速检测试纸条,研制出RB标准品浓度比色卡.以比色卡作为判定依据,以辣椒、花椒为研究对象,消除基质影响,进行试纸条检测.结果表明:RB线性为0.01~20 ng/mL,其最低检出限为0.5 ng/mL,试纸条显色结果明显.该试纸条实验成功,无需特殊设备即可实现现场快速检测,适用于快速定性检测香料中RB.     

睡眠类特膳食品中非法添加地西泮的快速检测

为快速检测特膳食品中非法添加的地西泮,文章基于经典的免疫层析侧流试纸(lateral flow strips, LFS)结合金纳米粒子(gold nanoparticles, GNPs)标记,建立一种快速、简单、方便的胶体金试纸条检测方法。结果表明：制备的试纸条能够在不到10 min的时间内完成检测,且无需复杂的预处理过程,特异性好,灵敏度高,视觉检测限可低至10.0μg/L。该方法操作简单、安全便捷,可以有效地实现睡眠类特殊膳食食品中非法添加地西泮的快速检测。     

脊髓灰质炎病毒Ⅰ型抗体胶体金检测试纸条的研制

目的:为研制快速检测脊髓灰质炎病毒Ⅰ型抗体的胶体金试纸条.方法:以辛酸-硫酸铵沉淀法纯化脊髓灰质炎病毒Ⅰ型单克隆抗体,用柠檬酸三钠还原法制备胶体金溶液,胶体金标记单抗,再结合脊灰病毒抗原形成复合物,固定于金标垫上,将鼠抗人IgG和羊抗鼠IgG分别包被于试纸条的T (检测线)处和C (质控线)处,应用该试纸条进行性能检测.结果:以已知的脊灰病毒Ⅰ型阳性血清作为检测样品,试纸条检测的灵敏度为1:80;特异性试验证明该试纸条与甲型肝炎病毒、乙型肝炎病毒、麻疹病毒抗体阳性血清没有交叉反应;检测结果与脊灰病毒Ⅰ型疫苗抗体检测试剂盒(ELISA)试验的符合率为86.7%.结论:该试纸条检测脊灰病毒Ⅰ型的抗体水平具有较好的特异性与灵敏性,适用于大规模临床血清抗体水平检测,具有良好的应用前景.     

烟草花叶病毒快速检测方法的建立及应用

应用单克隆抗体技术获得抗烟草花叶病毒(Tobacco mosaic virus,TMV)单克隆抗体,并通过胶体金标记技术,免疫层析快速检测技术和SQUID磁学定量测定技术建立起烟草花叶病毒的快速检测方法,并开发出用于定性检测的TMV胶体金快速检测试纸及用于定量检测的TMV纳米磁珠快速检测试纸,其检测灵敏度分别为1 ng/mL和0.1 ng/mL.所建立的方法操作简单,快速,灵敏度高,特异性好,是作为烟草育苗中各种TMV无症和有症烟株现场筛查的有效手段.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
