从实验室保存的菌株中分离筛选能够提高红景天苷含量的菌株,通过重氮盐比色法初筛和HPLC复筛得到一株黑曲霉Aspergillus niger(编号35),经发酵红景天原药材,提高其中红景天苷的含量.该菌株在接种量10%,料水比1∶15,30 ℃,200 r/min的最佳发酵和培养条件下发酵48 h,发酵后的原药材中红景天苷含量比发酵前同比提高52.83%,酪醇含量提高11.54%,证明微生物发酵能够提高红景天中红景天苷的含量.  相似文献

通过正交实验和对比实验,找出较优的红景天苷初步纯化的方法。以纯化后样品的红景天苷得率为考察指标,采用乙酸铅沉淀法正交实验和乙醇沉淀法正交实验,确定较优化的工艺参数。1.乙酸铅沉淀法的最佳方案即乙酸铅浓度为5%,沉淀时间为50min,温度为4℃,红景天苷含量为7.58%;2.乙醇沉淀法的最佳方案即乙醇浓度为70%,沉淀时间为24h,温度为4℃,红景天苷含量为20.69%。结论醇沉法和乙酸铅沉淀法均能提高红景天苷的纯度,醇沉法优于乙酸铅沉淀法。  相似文献

目的:降低红景天苷的制备成本并且提高红景天苷的产率,研究红景天苷合成的新工艺.方法:本实验以对羟基苯乙醇为原料,通过与糖苷化反应得到红景天苷化合物.结果:经高效液相色谱(HPLC)检测得到的红景天苷单体纯度达到98.5%以上,产率为46.2%.结论:本实验工艺可以制备高纯度红景天苷,主要降低了生产成本,并对于工业化生产高纯度红景天苷提供一定参考作用.  相似文献

为建立一个稳定可靠分离度好的方法测定红景天中含红景天苷的量.采用高效液相色谱法,色谱柱为BDS HYPERSIL-C18(4.6 mm×250 mm,5μm);以乙腈-0.03%磷酸为流动相进行梯度洗脱;流速1.0 m L/min;检测波长275 nm;柱温30℃.结果表明,红景天苷的进样量在1.25~10μg范围内有良好的线性关系,平均回收率为98.16%,RSD为0.89%.该方法准确、可靠、分离度好,可用于红景天的质量评价.  相似文献

本研究旨在建立高效液相色谱法测定红景天软胶囊中红景天苷含量的方法,为红景天软胶囊提供检测和质量控制参考。用甲醇对样品进行超声处理,色谱条件为色谱柱(C18,4.6 mm×250 mm,5.0μm),流动相为甲醇∶水=15∶85(V∶V),柱温为35℃,检测波长为215 nm,流速为1.0m L·min-1,外标法定量。结果表明,红景天苷在0.021~0.114 mg·m L-1范围内具有良好的线性关系(r2=0.99966),平均回收率为98.14%(n=9)。此法操作简单快速,准确性和重现性好,可作为红景天软胶囊中红景天苷含量的检测方法。  相似文献

用反相高效液相色谱法,流动相为甲醇-0.1mol/L磷酸二氢钠(15:85),紫外检测波长为280nm,流速1.0mL/min,测定新疆产红景天中红景天苷含量,结果表明,红景天苷对照品在10~320μg/mL内线性关系良好,r=0.999。平均加样回收率为96.5%;RSD为3.4%。稳定性良好。该法简便,分离完全,结果可靠,适用于红景天苷的含量测定。  相似文献

用超声波法水提取红景天,采用L16(4^5)正交试验法,以粗提物中红景天苷的含量为指标优选提取工艺.超声波法提取最佳提取方案为:溶剂为40℃高纯水,料液比为1:20,浸泡时间为1h,提取温度为60℃,提取时间为30min,提取次数为4次.大花红景天中红景天营的提取率为1.638%,狭叶红景天中红景天苷的提取率为2.401%.  相似文献

采用分光光度法,对天然药源红景天的汞含量及最佳浸提方法和测定条件进行了研究。结果表明,在120℃、硝酸与高氯酸体积比为4∶1条件下,湿法热浸提对汞离子的消解效果最好;显色剂甲基红用量为0.25 mL(质量浓度为0.5%)、增敏剂OP用量为2 mL(质量浓度为1%)、显色反应30 min、pH=2时,测定的灵敏度和精确度最高;红景天中的汞含量为0.11 ug∕kg;RSD为1.527%。此文方法可直接用于药材红景天中汞含量的测定。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
