Our study relates to the development of the membrane relief of the blastomeres of the trout egg from the stage before their division up to gastrulation. The surface of the blastoderm shows first a more or less dense system of crests which by the time of gastrulation are replaced by microvilli distributed over the whole surface of the enveloping layer. The deep blastomeres, loosely connected to each other by lamellipodes and filopodes produce only a few projections of their membranes and these are only semispherical bodies. Possible roles of the microvillosities in the respiratory exchange of the egg and in the motility of the enveloping layer, are considered.  相似文献

Summary 2-Mercaptopropionyl-glycine (Thiola) exerted a strong animalizing action on the development of the sea urchin,Paracentrotus lividus. The action of Thiola on the synthesis of superficial and cortical cellular structures newly synthesized during the segmentation is suggested. Summary The activity of the DPNH-diaphorase and the alkaline phosphatase were examined in golden-hamster eggs prior to fixation. The reactions for DPNH-diaphorase as well as for alkaline phosphatase were found to be positive already in the undivided egg and in early cleavage stages. The reaction products of both enzyme determinations showed a histophotometrically measurable polar distribution in the cytoplasm, in the one-cell stage as well as in the individual blastomeres of the examined early cleavage stages.  相似文献

Rotenone and high doses of chloramphenicol, both of which specifically inhibit electron transport between NADH and flavoprotein in the respiratory chain, caused fully separated Rana pipiens blastomeres to refuse, as shown by syncytium counts on embryos reconstructed from serial sections. With chloramphenicol, the effect was completely reversible: re-cleavage and normal development followed drug removal. The blastomere fusion effect was not produced by the succinic dehydrogenase-specific respiratory inhibitor, thenoyltrifluoroacetone, nor by a non-mitochondrial protein synthesis inhibitor, cycloheximide, both of which instead produced simple arrest of cleavage.  相似文献

One difficulty in the production of allophenic mice by aggregation of preimplantation embryos is that they frequently roll apart before the bonds between the blastomeres have had time to form. One solution to the problem, described here, is to pretreat one of the embryos with rabbit anti-mouse serum just prior to pushing them together. Blastocyst formation is unhampered by antibody treatment, and numerous allophenic mice have already been produced with this new procedure.  相似文献

The cosmological constant is back. Several lines of evidence point to the conclusion that either there is a positive cosmological constant or else the universe is filled with a strange form of matter (“quintessence”) that mimics some of the effects of a positive lambda. This paper investigates the implications of the former possibility. Two senses in which the cosmological constant can be a constant are distinguished: the capital Λ sense in which lambda is a universal constant on a par with the charge of the electron, and the lower case λ sense in which lambda is a humble constant of integration. The latter interpretation has been touted as the means to a solution to various problems in physics. These claims are critically examined with an eye to discerning the implications for philosophy of science and foundations of physics.  相似文献

A number of risk factors for cardiovascular disease including hyperinsulinemia, glucose intolerance, dyslipidemia, obesity, and elevated blood pressure are collectively known as metabolic syndrome (MS). Since mitochondrial activity is modulated by the availability of energy in cells, the disruption of key regulators of metabolism in MS not only affects the activity of mitochondria but also their dynamics and turnover. Therefore, a link of MS with mitochondrial dysfunction has been suspected since long. As a chronobiotic/cytoprotective agent, melatonin has a special place in prevention and treatment of MS. Melatonin levels are reduced in diseases associated with insulin resistance like MS. Melatonin improves sleep efficiency and has antioxidant and anti-inflammatory properties, partly for its role as a metabolic regulator and mitochondrial protector. We discuss in the present review the several cytoprotective melatonin actions that attenuate inflammatory responses in MS. The clinical data that support the potential therapeutical value of melatonin in human MS are reviewed.  相似文献

When N-methylformamide is administered to rats on the 11th day of pregnancy approximately 50% of the fetuses are resorbed and a reduced weight of the developed animals is found in comparison to the controls on the 21th day (delivery by Caesarian section). The toxic effect is increased by using nicotinamide and methionine. If a combination of these substances is employed practically all fetuses are resorbed. Tryptophan, however, has a considerably protective influence. N-Methylformamide has no influence on the NAD-synthesis induced by nicotinamide or tryptophan. It does, however, inhibits the activity of the ADPR transferase.  相似文献

Summary The octopamine, dopamine and noradrenaline content of the brain of the locust,Schistocerca gregaria has been determined using sensitive radiochemical-enzymatic assays. Octopamine and dopamine are present in high concentration but the noradrenaline content is only 1/25 that of octopamine. Both reserpine and fusaric acid (a dopamine--hydroxylase inhibitor) produce a significant depletion of the octopamine stores.The author is a M.R.C. of Canada Fellow. Thanks are due to Drs.A. A. During the last three decades, there has been a growing realization among physicists and cosmologists that the relation between particle physics and cosmology may constitute yet another successful example of the unity of science. However, there are important conceptual problems in the unification of the two disciplines, e.g. in connection with the cosmological constant and the conjecture of inflation. The present article will outline some of these problems, and argue that the victory for the unity of science in the context of cosmology and particle physics is still far from obvious.  相似文献

It is often held by philosophers of science that special, idealized situations are prior to complex cases in several senses: equations for complex cases are derived from those for special cases by “composing” special case equations; behavior in complex cases is explained in terms of behavior in special cases; one learns the true nature of a property in the special case where it is allowed to work in isolation. In this paper, I argue that a strand of non-equilibrium thermodynamics which attempts to go beyond the limitations of classical non-equilibrium thermodynamics adheres to something that is the reverse of this picture. Thus, the legitimacy (or lack thereof) of this picture lies very near to the heart of foundational issues in non-equilibrium thermodynamics.  相似文献

String theorists are certain that they are practicing physicists. Yet, some of their recent critics deny this. This paper argues that this conflict is really about who holds authority in making rational judgment in theoretical physics. At bottom, the conflict centers on the question: who is a proper physicist? To illustrate and understand the differing opinions about proper practice and identity, we discuss different appreciations of epistemic virtues and explanation among string theorists and their critics, and how these have been sourced in accounts of Einstein's biography. Just as Einstein is claimed by both sides, historiography offers examples of both successful and unsuccessful non-empirical science. History of science also teaches that times of conflict are often times of innovation, in which novel scholarly identities may come into being. At the same time, since the contributions of Thomas Kuhn historians have developed a critical attitude towards formal attempts and methodological recipes for epistemic demarcation and justification of scientific practice. These are now, however, being considered in the debate on non-empirical physics.  相似文献

The kinesins constitute a large family of motor proteins which are responsible for the distribution of numerous organelles, vesicles and macromolecular complexes throughout the cell. One class of these molecular motors, kinesin-II, is unique in that these proteins are typically found as heterotrimeric complexes containing two different, though related, kinesin-like motor subunits, and a single nonmotor subunit. The heteromeric nature of these kinesins appears to have resulted in a class of combinatorial kinesins which can 'mix and match' different motor subunits. Another novel feature of these motors is that the activities of several kinesin-II representatives are essential in the assembly of motile and nonmotile cilia, a role not attributed to any other kinesin. This review presents a brief overview of the structure and biological functions of kinesin-II, the heteromeric kinesin.  相似文献

Summary Cap formation in mouse spleen cells induced by antiimmunoglobulin was inhibited by the metals Be, Fe, Cu and Al. Be was especially strong as an inhibitor of cap formation. It is suggested that these metals might change the mobility of the membrane and have some biological effects on the cross association of antigen receptors when B lymphocytes are attached by them.  相似文献

Differential pulse voltammetry, performed with electrically treated carbon fiber electrodes, enables us to detect in vitro or in vivo in the striatum of anesthetized Rats, an oxidation peak 3 at a potential of +300 mV. Electrolytic, or 5,7-dihydroxytryptamine lesions of the medial forebrain bundle are followed by a decrease of respectively 59 and 62% of this peak. Biochemical measurements are significantly correlated to the measured peaks 3 and troughs. Thus, peak 3 increases obtained after injection of L-tryptophane and/or reserpine, as well as the troughs observed after injection of clorgyline and/or NSD 1015 confirm that the peak 3 is dependent upon 5-hydroxyindolacetic acid (5-HIAA) concentration.  相似文献

This review discusses the state-of-the-art in molecular research on the most prominent and widely applied lantibiotic, i.e., nisin. The developments in understanding its complex biosynthesis and mode of action are highlighted. Moreover, novel applications arising from engineering either nisin itself, or from the construction of totally novel dehydrated and/or lanthionine-containing peptides with desired bioactivities are described. Several challenges still exist in understanding the immunity system and the unique multiple reactions occurring on a single substrate molecule, carried out by the dehydratase NisB and the cyclization enzyme NisC. The recent elucidation of the 3-D structure of NisC forms the exciting beginning of further 3-D-structure determinations of the other biosynthetic enzymes, transporters and immunity proteins. Advances in achieving in vitro activities of lanthionine-forming enzymes will greatly enhance our understanding of the molecular characteristics of the biosynthesis process, opening up new avenues for developing unique and novel biocatalytic processes.
