The absolute configuration of the enantiomers of glutethimide and aminoglutethimide.

Aminoglutethimide (Elipten? CIBA) was resolved into the optical antipodes I and II. The endocrinological properties and the absolute configuration of both enantiomers I and II were determined. Most of the steroidal synthesis inhibition was found in the (+) enantiomer II. On the basis of circular dichroism, the R-configuration was assigned to the (+) enantiomer II.

A relationship between ring substituents and absolute configuration in the aporphine series. The structures of thalicmidine and argemonine

Résumé L'auteur démontre qu'il existe, d'une manière générale, un rapport entre les substituants du cycle D des aporphines et la configuration absolue de ces alkaloïdes. De nouvelles structures ont été proposées pour la thalicmidine et l'argémonine.

The relationship between the ring D-substituents and the absolute configuration for the aporphine alkaloids

Résumé Les alcaloïdes aporphiniques substitués en positions 1, 2, 9, 10 on en 1, 2, 10, 11 sont dextrorotatoires et du type I. Mais les aporphines monosubstituées ou non substituées dans le cycle D, et qui ne parviennent pas de la famille Papaveraceae, sont généralement lévorotatoires et du genre II.

Zusammenfassung 1,2,3,5-Tetrahydroimidazo(2,1-b)chinazolin wurde als neuer Typ eines bei Tieren gut wirksamen Hypotensivums erkannt. Bei -blockierender Komponente wurde keine orthostatische Hypotonie festgestellt.     

Relationship between the absolute configuration and the biological activity of juvenile hormone III

Summary The activity of the pure 10R (=natural) and 10S enantiomers of juvenile hormone III (JH III) was determined in 3 different bioassays, and the relative binding affinity of the 2 enantiomers to the haemolymph JH-binding protein of the cockroachNauphoeta cinerea was measured. In theGalleria wax test, a local morphogenetic assay, the 10R enantiomer was 5240 times more active than, the 10S enantiomer, 1Galleria unit corresponding to 0.42 pg of 10R-JH III as compared to 2.2 ng for 10S-JH III. In a systemic morphogenetic assay with the cockroachNauphoeta cinerea 380 times less 10R enantiomer was necessary in order to induce detectable juvenilisation (58 ng 10R and 22 g 10S) and in a systemic gonadotropic assay withNauphoeta cinerea 255 times less 10R was needed to induce vitellogenin synthesis in 50% of the insects (6.7 ng 10R and 1710 ng 10S). In the JH-binding protein assay 10R-JH III had an affinity for the JH-binding protein (lipophorin) which was approximately 46 times higher than that of 10S-JH III.     

The exotoxin ofBacillus thuringiensis: A new C-mitotic agent

Summary Thuringiensin A, an exotoxin fromBacillus thuringiensis, a constituent of the microbial insecticide thuricide has been found to inhibit mitotic spindle, condense and scatter chromosomes. It may therefore be a promizing tool in future cell biological studies.We thank Prof.H. Sharatchandra for facilities. S-adenosyl-L-homocysteine: A non-cytotoxic hypomethylating agent

The cytotoxic effect caused by the hypomethylating agent S-adenosyl-L-homocysteine (SAH) was compared with that of two drugs commonly used to induce DNA hypomethylation, 5-azacytidine and 5-aza-2-deocycytidine. Two in vitro cytotoxicity tests, the tetrazolium MTT assay and the intracellular lactate dehydrogenase (LDH) activity test, suggest that SAH induces hypomethylation without causing any cytotoxic effect. We propose the use of SAH as a non-cytotoxic agent which may be more suitable for inducing experimental DNA hypomethylation.

The specification of asymmetric configuration in organic chemistry

Zusammenfassung Ein früherer Vorschlag⁴ für die Spezifizierung der asymmetrischen Konfiguration organischer Verbindungen wurde modifiziert und erweitert. In der vorliegenden Abhandlung wird mehr Nachdruck auf Beispiele gelegt, welche das Verfahren erläutern, als auf eine formelle Darlegung der Prinzipien in allgemeiner Form. Die prinzipiellen und praktischen Gründe, welche für ein allgemeines Verfahren zur Spezifizierung der absoluten asymmetrischen Konfigurationen sprechen, werden diskutiert. Da ein solches Verfahren nur auf wenigen eindeutigen Regeln beruhen soll, muss es folgende Bedingungen erfüllen: 1. die Reste, welche die Asymmetric bedingen, müssen auf Grund einer permanenten, internen Eigenschaft geordnet werden; 2. das dreidimensionale Muster, welches so erhalten wird, muss direkt als Klassifikationsgrundlage dienen.Ebenso wie in der früheren Mitteilung wird auch in der vorliegenden Abhandlung als ordnende Eigenschaft von Gruppen, welche sich in ihrer chemischen Konstitution unterscheiden, die Atomnummer verwendet. Darüber hinaus wird gezeigt, wie die feineren Unterschiede, welche Asymmetrie verursachen, wie die Atommasse sowie Konfiguration und Position der Gruppen, berücksichtigt werden können.Das Verfahren wurde soweit ausgebaut, dass man es auf alle bekannten Typen der optischen Isomerie organischer Verbindungen anwenden kann. Zur Spezifizierung der Konfiguration werden neue Symbole vorgeschlagen, welche ausschliesslich die absolute Konfiguration darstellen sollen.     

A novel fluorination reaction: The interaction of perchloryl fluoride with active methylene compounds

Zusammenfassung Die Reaktion von Perchloryl Fluorid (PF) mit Anionen wurde untersucht. Eine leichte Difluorierung fand statt mit Carbanionen vom Typus CH (C=X)₂. Mechanismen für die verschiedenen Heterolysen von PF werden vorgeschlagen.

Summary A structure is assigned to saframycin A, a novel antitumor antibiotic fromStreptomyces lavendulae No. 314, on the basis of¹³C NMR-spectral data.     

A comparison of forecasts from least absolute value and least squares regression

A Monte Carlo simulation is used to compare forecasts from least absolute value and least squares estimated regression equations. When outliers are present, the least absolute value forecasts are shown to be superior to least squares forecasts. The results emphasize the importance of exercising caution when using forecasts from least squares estimated regressions. Use of least absolute value regression (or some other robust regression method) instead of, or as an adjunct to, least squares is recommended. The comparison of forecasts from the two methods provides one way of assessing whether the least squares forecasts have been adversely affected by outliers. If outliers are present, the least absolute value regression forecasts can be used.
