This paper reports quantitative examinations of some rat serum proteins which occur only, or in rising concentrations in pregnant rats before and after birth. A new serum protein is described which is only demonstrable in female rats. In male rats this protein is found rarely and then in very small quantities.  相似文献

Summary This paper reports evidence for a hormonal regulation of a new rat serum protein (female specific protein, FP) which is only demonstrable in female rats. Male and female rats were treated with testosterone and estrogen. The FP was assayed with immunological methods. The following results were obtained: 1. In testosterone-treated females the serum level of FP is reduced significantly. 2. In estrogenized males the FP was distinctly demonstrable but not in the control males.  相似文献

Summary Prolonged administration of glibenclamide decreased blood sugar, liver glycogen and protein and increased liver and serum lipids and organic phosphates of liver in normal rats. 24 h of a psychogenic stress (restraint) caused a strong increase of liver metallothionein (MT) levels. 3 h of stress were sufficient to induce an increase in liver MT, measured 21 h later, but the increase was much lower than in continuously restrained rats. Stress induction of liver MT was not due to food deprivation, since rats deprived for 24 h showed lower MT levels than stressed ones. Zn on MT presented the same qualitative but not quantitative pattern of response as MT protein. Liver cytosolic Zn was increased by restraint in spite of their being no decrease in serum Zn. Any treatment altered serum MT. Liver and serum MT were not correlated. The present results demonstrate that basically psychogenic stresses increased liver but not serum MT levels. No positive evidence for a relationship between corticosterone secretion and MT induction was found.  相似文献

目的观察解聚复肾宁(Jiejufushenning,JJFSN)对糖尿病(Diabetes mellitus,DM)大鼠肾组织肿瘤坏死因子-α(Tumorneerosisfactor-alpha,TNF-α)、单核细胞趋化蛋白-1(Monocyte chemoattractant protein-1,MCP-1)和血清TNF-α的影响,探讨其肾保护作用机制。方法腹腔注射链脲佐菌素(Streptozotocin,STZ)建立SD大鼠DM模型,将成模DM大鼠随机分为3组:DM模型组(B组)、JJFSN组(C组)、厄贝沙坦(Irbesartan)组(D组),另设正常对照组(A组)。采用相应干预措施处理12周。常规方法测定12周末各组大鼠空腹血糖(FBG)、血尿素氮(BUN)、血肌酐(Scr)、肾重/体重(KW/BW)、24h尿蛋白(24huFro);放免法测血清TNF-α含量:免疫组织化学方法测肾组织TNF—Ot、MCP-1的表达;PAS染色评估细胞外基质;电镜观察肾组织超微结构改变。结果与A组相比.B组大鼠FBG、BUN、Scr、KW/BW、24huPro及血清TNF-α升高(P〈0.05),肾组织TNF—Ot、MCP-1表达明显增高(P〈0.05);肾组织超微结构明显异常;C组和D组上述指标显著改善(P〈0.05),肾组织超微结构异常改善。结论竹FSN能延缓DM大鼠肾损害进程,其机制可能与抑制TNF-α、MCP-1上调有关。  相似文献

A marked increase of serum transaminase activities, histological changes of livers similar to those seen in viral hepatitis in man, and inhibition of hepatic protein synthesis were observed in rats following a single injection of D-galactosamine-HCl. These galactosamine-induced phenomena were prevented by the pretreatment of polyriboinosinic acid-polyribocytidylic acid 24 h before the galactosamine administration.  相似文献

A dose of soy bean fat emulsion which was injected i.v. in suckling rats accumulated in the cells of liver parenchyma, both in hepatocytes and in reticuloendothelial cells. Subsequent i.p. injection of E. coli endotoxin was followed by extensive liver tissue necrosis and increased activities of serum aspartic and alanine aminotransferase. These signs of liver damage were markedly more pronounced than those observed after the administration of E. coli endotoxin only.  相似文献

目的筛选胎儿宫内发育迟缓大鼠血清中差异表达的蛋白质,寻找与胎儿宫内发育迟缓相关的疾病标志物。方法收集大鼠疾病正常组、IUGR组血浆各3例;TCA法沉淀血清蛋白,建立疾病组和正常组蛋白质的双向凝胶电泳图谱;ImageMaster分析软件分析差异蛋白质;飞行时间质谱(MALDI—TOF—MS)获得差异蛋白肽指纹图谱,MASCOT引擎搜索蛋白质种类。结果鉴定出个差异蛋白点3个,分别为:IGF—I、IGF-Ⅱ、IGFBP-3。结论IGF—I、IGF-II、IGFBP-3,可能是胎儿宫内发育迟缓疾病诊断的生物标志物。  相似文献

目的通过观察链霉素诱导的2型糖尿病大鼠模型肾脏功能的变化,以探讨此种模型是否适用于糖尿病肾损害的研究。方法采用链脲佐菌素(streptozotocin,STZ)辅以高脂饮食建立2型糖尿病大鼠模型,筛选造模成功的大鼠为糖尿病组,并与正常大鼠比较,造模后8周观察两组大鼠血糖、血尿素氮(blood urea nitrogen,BUN)、血肌酐(creatinine,CR)、血糖基化终末产物(Advanced glycation end products,AGE)、尿微量白蛋白、肾重指数的变化以及AGE受体(Receptor of AGE,RAGE)、蛋白激酶C(protein kinase C,PKC)、蛋白激酶A(protein kinase A,PKA)在肾脏的表达变化。结果糖尿病组的血糖、血尿素氮、血肌酐、血AGE、尿微量白蛋白以及肾重指数均明显高于正常组(P0.05),RAGE、PKC的mRNA以及蛋白表达水平均较正常组高(P0.05),PKA的mRNA和蛋白表达水平则较正常组低(P0.05)。结论 STZ诱导的2型糖尿病大鼠模型在造模后第8周肾脏功能存在明显的损害,并且导致肾脏损害的上游重要介质RAGE、PKC、PKA在第8周都有着明显的变化。此种模型能够用于糖尿病肾损害的研究,并且是观察中早期糖尿病肾病的较好的模型。  相似文献

The effect of prolactin on estradiol receptors was determined in cytosol of estrogen-dependent mammary tumors of Sprague Dawley rats. Tumors were induced with 20 mg dimethylbenzanthracene, rats were ovariectomized, and the rats showing regression in tumor weight were given 1 mg sheep or rat prolactin for 5 days. Receptors were separated by dextran-charcoal and analyzed by sucrose gradient. Prolactin, milk proteins, serum proteins, and 1-alpha-feto-proteins were excluded immunologically. The estradiol receptors resembled uterine receptors in affinity, specificity, sedimentation, and nuclear transfer under estradiol influence. Castration diminished receptor content exponentially 170 f moles to 16 f moles/mg protein within 10 days; prolactin increased the number of receptors to 87 f moles compared with 15 in controls. Affinity, cellular location, and uterine receptor content were unaffected. Sheep and rat prolactin acted similarly. These results justify hypophysectomy as therapy for certain estrogen-dependent breast tumors.  相似文献

We compared the effects of 0.45% normal saline (NS), 5% Intralipid® (IL), and 16.7% glucose (Glu) infusions on total serum triglycerides and cholesterol, serum high-(HDL-c) and low-density lipoprotein cholesterol (LDL-c), and activity of serum lecithin: cholesterol acyltransferase (LCAT), and serum lipoprotein lipase (LPL) in rats implanted with a fibrosarcoma. In tumor-bearing rats given NS, a two-fold increase in total serum cholesterol, a four-fold increase in LDL-c, and a five-fold decrease in the HDL-c/LDL-c ratio were observed compared to tumor-free rats. In tumor-bearing rats administered IL, a two-fold increase in total serum triglyceride and cholesterol, a three-fold increase in HDL-c and HDL-c/LDL-c ratio, and a two-fold increase in LPL activity were observed compared to tumor-bearing rats administered NS. In tumor-bearing rats administered Glu, a two-fold decrease in total serum cholesterol, a two-fold decrease in HDL-c, and a three-fold decrease in LDL-c were observed compared to tumor-bearing rats administered NS. Tumor weights and LCAT activity did not differ significantly between treatment groups. Previous results have demonstrated that lipophilic compounds that interact with plasma lipoproteins have altered pharmacological effects when administered with IL. Therefore, this study suggests that IL infusions alter the HDL-c/LDL-c ratio and could affect the pharmacological behavior of anticancer compounds that predominantly distribute into the LDL fraction upon entrance into the bloodstream.  相似文献

Summary The amount of free fatty acid (FFA) liberated with intralipid in post-heparin guinea-pig serum, when serum from alloxan diabetic rat is incubated, is higher than that liberated when serum from healthy rats is used. The serum gastrin concentration and the gastric histidine decarboxylase activity are high in freely fed, unoperated rats but low in antrectomized rats. Following food deprivation the serum gastrin level and the enzyme activity are reduced simultaneously in the unoperated rats. After fasting for 36-48 h - but not before - the enzyme activity drops to the same low levels as in antrectomized rats.  相似文献

Administration of T4 on alternate weeks for 30 weeks at a dosage which does not alter body weight depresses basal serum corticosterone levels in older rats (575 days), but not in young animals (260 days). Summary We studied serum glycoproteins in rats and guinea pigs during the development of granulomas, provoked by carrageenin injection. In rats total serum sialic acid was raised as well as sialic acid soluble in sulfosalicylic acid and haptoglobin. The rise of these 2 fractions, seromucoid and haptoglobine accounts quantitatively for the rise of the total sialic acid. In guinea pigs total serum sialic acid was unchanged, but sulfosalicylosoluble sialic acid as well as haptoglobin were much higher than basal values. Quantitatively the serum glycoprotein reaction was much higher in rats than in guinea pigs.  相似文献

Summary Thyroidectomy decreased prolactin concentrations in the anterior pituitary (AP) and serum of the male rat. The amount of basal and thyrotropin-releasing hormone (TRH)-stimulated release of prolactin by AP in vitro was lower in thyroidectomized (Tx) rats than in sham Tx rats. These results suggest that the inhibitory effects of thyroidectomy on pituitary and serum prolactin in male rats are mediated in part by the reduction of the production and spontaneous release of prolactin and the responsiveness of prolactin to TRH.  相似文献

The chemical composition of liver plasma membrane was studied in Wistar rats aged between 3 and 24 months. Results obtained indicate a significant age-dependent positive correlation of both the protein: phospholipid and cholesterol: phospholipid ratios, whereas the protein: cholesterol ratio seems to remain unaffected. Phospholipid analysis of liver plasma membrane reveals that only the phosphatidylcholine content has a significant negative correlation with age; all other phospholipid species remain basically unchanged.  相似文献
