紫锥菊的组织培养及其实用快速繁殖   总被引:4,自引:0,他引:4  
利用植物组织培养技术快速育苗是一个新兴的生物技术应用领域.文中采用组织培养技术,以MS培养基为基本培养基,通过调节植物生长激素的配比对紫锥菊组织进行愈伤化、继代、分化、生根等系列培养,培育出完整植株,并采用声波刺激方式炼苗以提高移栽成活率.该方法避免了传统紫锥菊育苗时种子采集困难,发芽率低,难以大规模育苗的特点,从而为建立规模化培育紫锥菊苗以及为我国开发利用紫锥菊药用资源提供了理论依据.  相似文献

以自花受粉的笋玉米幼胚为外植体建立了胚性细胞悬浮系,酶解游离原生质体通过液体浅层培养得到了再生植株;通过PEG法和电激法对原生质体进行了遗传转化,转化用外源基因是潮霉素抗性基因和BT基因,原生质体再生愈伤组织经潮霉素筛选后,PEG法和电激法获得抗性愈伤组织的频率分别为9 3%和8.9%,Southem-blotting证明外源基因已整合到玉米抗性愈伤组织细胞基因组中,转基因工程植株的分化工作在正在进行中.  相似文献

以葡萄花药诱导的胚性愈伤组织为材料,在附加5%纤维素酶(OnozukaR-10)和0.5%蜗牛酶的混和酶液中分离,获得大量具有活力的原生质体.分离的原生质体在附加2mg/L 6—BA和0.5mg/L 2,4—D的改良B_5培养基中进行液体浅层培养,原生质体能再生细胞壁、生长和分裂,形成了较大的细胞团。  相似文献

酶解墨西哥黑甜玉米的愈伤组织产生的原生质体,经380μW/cm~2紫外线分别照射30 s,60 s,180 s,300 s,发现原生质体的活力及生长与紫外线的剂量有密切关系,随着紫外线剂量的增大,生长及活性趋于降低.辐照30 s的玉米原生质体与对照都可以恢复分裂并长成愈伤组织;辐照60 s的原生质体可以形成小细胞团,未见形成愈伤组织;而辐照180 s和300 s的原生质体都未见细胞分裂.  相似文献

小麦胚性愈伤组织酶解5小时后,原生质体产率可达6.3×106g.fw-1,酶种类和浓度对小麦原生质体的产率有影响小麦原生质体对不同浓度渗透剂的适应范围较广,在添加0.3mol·L-10.7mol·L-1甘露醇、山梨醇的培养基上,存活率超过50%所试培养基中,KM8P效果最好,实验表明可能与含丰富的附加成分有关除高浓度精胺外,小麦原生质体的存活率随腐胺、精胺浓度的增加而上升,但对原生质体的分裂频率影响不大在上述最佳条件下,小麦原生质体在培养35天后出现第一次分裂,20天后统计分裂频率为25.6%,3个月后植板率为28%,并形成较多的愈伤组织  相似文献

应用大孔吸附树脂吸附分离技术制备菊苣酸的研究   总被引:2,自引:1,他引:2  
考察了9种大孔吸附树脂对紫锥菊中菊苣酸的吸附分离性能,确定大孔吸附树脂吸附分离菊苣酸的工艺条件。结果表明AB-8树脂对菊苣酸有良好的吸附分离性能,其吸附分离菊苣酸的工艺条件为:质量浓度为3~4mg/mL,pH值为3的菊苣酸原料液以2mL/min的流速上柱吸附,再用6倍量树脂体积 (6BV)的30%乙醇以1mL/min的流速上柱进行解吸。AB-8树脂柱饱和吸附量可达18.0mg/mL,解吸率达90.2%。经AB-8树脂吸附分离,产品纯度达20.2%,纯度比紫锥菊初提物提高了近5倍。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
