一个拟南芥C2H2锌指蛋白的结构域分析

At3g23140是拟南芥中仅含有一个C2H2锌指结构的转录因子,主要含有N端的C2H2锌指结构和C端的类似EAR两个结构域.将〖WTBX〗〖STBX〗At3g23140〖WTBZ〗〖STB3〗中仅含C2H2锌指结构区域不同长度的两个基因片段〖WTBX〗〖STBX〗A1〖WTBZ〗〖STB3〗(240 bp),〖WTBX〗〖STBX〗A2〖STB3〗〖WTBZ〗(410 bp)克隆到植物表达载体pMON530 35S启动子的下游,并转化野生型拟南芥.经过筛选得到稳定遗传的T3代纯合转化子.对转基因植株研究表明, 35S::〖WTBX〗〖STBX〗A2〖STB3〗〖WTBZ〗转基因植株的内源乙烯释放量明显低于野生型,这与〖WTBX〗〖STBX〗At3g23140〖STB3〗〖WTBZ〗插入失活突变体〖WTBX〗〖STBX〗cs16966〖STB3〗〖WTBZ〗的表型是一致的,而35S::〖WTBX〗〖STBX〗A1〖WTBZ〗〖STB3〗转基因植株的内源乙烯释放量与野生型没有明显区别.表明A2片段的过量表达产生了显性抑制的作用,这种显性抑制效应很可能是由于 A2蛋白片段与〖WTBX〗〖STBX〗At3g23140〖STB3〗〖WTBZ〗表达产物所调控的核酸序列竞争结合所导致.而A2片段C端所含有的非锌指结构域是At3g23140蛋白与靶基因序列结合所必需的.     

人类神经生长抑制因子突变体E55/58/60Q和E58K的研究

人类神经生长抑制因子(GIF)又称MT3,其α 结构域存在一个特殊的插入六肽,其中含有多个谷氨酸残基,这些谷氨酸残基被认为与GIF具有保护神经元活性有关.为更好地研究这些Glu的作用,分别制备了E55/58/60Q和E58K2个突变体蛋白并对其稳定性进行了初步研究.结果显示,突变体蛋白的一系列光谱性质同野生型MT3非常一致,对酸的稳定性和对EDTA及DTNB的反应性上,也都表现出同野生型蛋白相似的性质.实验表明MT3α 结构域插入段中的Glu残基对蛋白的金属 硫簇结构及其稳定性没有显著的贡献.     

褪黑素单克隆抗体的制备及鉴定

为了获得抗褪黑素(MT)高效价的单克隆抗体,用甲醛将MT连结到BSA上,然后用连结物背部皮下多点免疫Balb／c小鼠,用PEG诱导免疫鼠脾细胞与Sp2／0细胞融合,经ELISA法筛选阳性克隆株及测定效价,用抗体与同类物之间的交叉反应进行特异性鉴定．最后获得5株(4C9D7、3E12C7H11E7、3E12A9D7、6A6A3A2C3、1E1E2H2H6)能稳定分泌高效价抗MT的单克隆抗体(McAb)的杂交瘤细胞。     

用分子动力学方法对Cystatin蛋白的研究

通过动力学模拟研究野生型和I66Q突变型cC的动力学特性,为设计和筛选治疗Cystatin C引起的脑淀粉样血管病药物提供重要的理论依据.通过分子动力学模拟的方法,选择可引起脑淀粉样血管病的Cystatin蛋白为模型,探讨了在形成致病状纤维过程中起到关键作用的蛋白质结构位点.研究分子动力学模拟显示,突变型cC较野生型更易形成淀粉样纤维,突变型蛋白的L1部位较野生型具有更高的柔性,更易通过结构域交换形成二聚体进而引发淀粉样纤维沉淀.该研究结果对于设计治疗相关疾病如疯牛病等的药物也有一定的借鉴意义.     

CYP2C9 基因多态性对苯妥因体内代谢的影响

研究了CYP2C9基因多态性对苯妥因代谢活性的影响.采用PCR扩增及直接测序的方法检测65名健康受试者CYP2C9基因型.并利用高效液相色谱考察了野生型CYP2C9*1/*1携带者和杂合突变型CYP2C9*1/*3携带者对苯妥英代谢的影响.结果检测到5名健康志愿者携带CYP2C9*3突变型等位基因(1075位碱基A-C),为CYP2C9*1/*3杂合突变型,未检测到CYP2C9*4(1076位碱基T-C)和CYP2C9*5(1080位碱基C-G)突变型等位基因.在受检人群中,未检测纯合突变型等位基因携带者.实验结果表明,CYP2C9基因型与p-HPPH/苯妥因代谢率之间具有显著的相关性.野生基因型携带者的平均代谢率为携带CYP2C9*1/*/*3杂合型等位基因志愿者的2倍以上,且两种基因型携带者间的代谢率具有统计学上的显著性差异.对本次健康受试者的CYP2C9基因型与苯妥因代谢的相关性研究表明,CYP2C9*3突变型等位基因引起酶活性降低,对苯妥因的代谢减慢,与CYP2C9野生型相比具有显著性差异.由于中国人群中CYP2C9*3(1075A-C)的发生频率较高,约为85～10%.因此,当CYP2C9在药物代谢中起主要作用时应检测其CYP2C9*3突变型.     

Periaxin蛋白中PDZ结构域的重组表达与纯化

Periaxin属于有髓施旺细胞非致密髓鞘中的重要蛋白之一,该蛋白的突变,会引起腓骨肌萎缩症4F亚型发生.文章以小鼠periaxin基因为模板,PCR扩增编码Periaxin-PDZ及其延长型的基因序列,插入pETM-3C表达载体中.转入E.coli BL21(DE3)中,经IPTG诱导表达重组蛋白.再经Ni2+-NTA亲和柱及Sephacryl S-200凝胶层析获得电泳纯的Periaxin-PDZ蛋白.荧光光谱法分析Periaxin-PDZ和脂膜的结合能力,结果显示Periaxin-PDZ(18-102aa)与脂质体的解离常数为Kd=4.415 μg/mL,延长型Periaxin-PDZ(18-129aa)与脂质体的解离常数为Kd=7.265 μg/mL,Periaxin-PDZ(18-102aa)与脂膜的结合能力略高于延长型Periaxin-PDZ(18-129aa).     

P53和MDM2蛋白表达对肝细胞肝癌预后判断的意义

目的 :研究肝细胞肝癌中突变型P5 3和MDM2蛋白的表达对临床预后判断的意义。方法 :应用免疫组织化学方法 ,检测 72例原发性肝细胞肝癌手术切除标本突变型P5 3、MDM2蛋白的表达 ;与临床病理学指标和术后生存期进行分析比较。结果 :突变型P5 3蛋白阳性 2 8例(38 89% ) ,MDM2蛋白阳性 2 3例 (31 94 % ) ,二者阳性表达有相关性 (r =0 2 4 8,P <0 0 5 )。突变型P5 3、MDM2蛋白阳性表达病例生存率明显低于突变型P5 3、MDM2蛋白阴性表达病例 (P <0 0 1)。突变型P5 3和MDM2蛋白表达均阳性病例 13例 (18 0 6 % ) ,中位生存期的生存率最低。单因素及多因素分析显示 ,突变型P5 3蛋白表达、MDM2蛋白表达、肿瘤大小与中位生存期的生存率有关 ,MDM2是统计学上最有意义的独立预后指标 (P <0 0 0 0 1)。结论 :应用免疫组织化学方法检测突变型P5 3和MDM2蛋白的表达可作为原发性肝细胞肝癌预后判断的指标。     

蛋白聚糖NG2中硫酸软骨素糖胺聚糖链调节稳定转染NG2的U251细胞迁移能力

应用PCR反应使编码蛋白聚糖NG2的核苷酸在3064-3065位置的AG突变成为GC, 构建突变型NG2/S999A. 用表达野生型NG2和突变型NG2/S999A及空白表达载体分别稳定转染人成胶质细胞瘤U251. 应用Western Blot方法鉴定转染后的U251细胞表达野生型及突变型NG2的状态, 证实突变型NG2A999稳定转染的U251细胞株表达的NG2缺失硫酸软骨素葡糖胺聚糖链（C S-GAG）. 比较了3种U251细胞株的迁移, 表明蛋白聚糖NG2中CS-GAG链影响细胞的迁移能 力.     

基因工程人表皮生长因子（rhEGF）单克隆抗体的制备与初步鉴定

以基因工程人表皮生长因子(rhEGF)为抗原,免疫BALB/c小鼠,将免疫的小鼠脾细胞与小鼠Sp2/0-Ag14骨髓瘤细胞进行融合,经多次筛选和再克隆化,获得3株能稳定分泌抗rhEGF单克隆抗体(McAbs)的杂交瘤细胞株.对该3株杂交瘤细胞小鼠腹水进行了抗体效价测定,结果显示在1×10-6～2×10-7之间.对上述3种杂交瘤单抗(分别命名为:AE-2A4,AE-2G5和AE-3E11)进行了特异性、抗原决定簇及相对亲和力测定.结果表明,它们都具有抗rhEGF高特异性;并分别抗两个不同抗原决定簇;相对亲和力大小次序为:AE-3E11>AE-2A4>AE-2G5.Ig亚类鉴定结果显示:AE-2A4,AE-3E11同为IgG1亚类,而AE-2G5为IgG2b亚类.确定AE-2A4单克隆抗体可用于制备高纯rhEGF.     

人GrnE单克隆抗体的制备和鉴定

为了制备获得针对人Grn E单克隆抗体,将原核表达GST-Grn E蛋白作为免疫原,免疫Balb/c小鼠,经过融合及初步筛选,获得5株针对Grn E的单克隆细胞株.选取其中3株(1D5、2E1和3F7)制备腹水,并通过western blot和免疫荧光染色对该3株单克隆抗体进行鉴定.在此基础上,进一步对1D5和3F7两株单克隆抗体是否可以用于免疫沉淀实验进行了检测.上述实验表明,本研究成功制备了针对Grn E结构域的单克隆抗体,而且所制备的单克隆抗体具有良好的特异性.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
