Tritium distribution on randomly labelled taurocholate (TC) was estimated at 28%, 4%, 1% and less than 0.5% on the hydrogens opposite the 3alpha-, 7alpha- and 12alpha-OH groups and taurine moiety respectively. Anomalously, C. perfringens 3alpha-hydroxysteroid dehydrogenase (3alpha-HSDH) catalyzed tritium loss of 36% on formation of 7alpha-, 12alpha-dihydroxy-3-keto-5beta-cholanoate, implying additional losses of tritium at other sites by this enzyme.  相似文献

Summary The common bile duct of male Sprague-Dawley rats was cannulated with either PE 10 or PE 50 tubing. Maximal secretory rate of taurocholate averaged 389±67 (SD) and 657±115 nmoles·min–1·g liver–1 in the PE 10 and PE 50 group, respectively (p<0.005). Maximal bile secretory pressure was significantly higher in the PE 10 group (240±28 vs 174±8 mm H20; p<0.005). When the maximal secretory rate was exceeded, bile flow decreased in both groups but this was accompanied with a decrease in maximal bile secretory pressure in the PE 10 group only. Maximal secretory rate of bile salts is markedly influenced by experimental technique. Use of small caliber common bile duct cannulae leads to partial obstruction and decreases the apparent maximal secretory rate for taurocholate.Acknowledgments. J. Summary Perfusion of a rat intestinal segment with a solution containing sodium deoxycholate (8 mM) increases the rate of firing in periarterial afferent nerves from the gut. Zusammenfassung Das Alkaloid Vincaleukoblastine (VLB) wurde als Base bei Anwendung der Wilzbach-Technik titriert. Die Verbindung, bis zur konstanten Aktivität gereinigt, enthielt kein Dihydro-VLB mehr. Das Alkaloid, als Sulfatsalz dem Tritium unterworfen, inkorporiert die Substanz nur geringfügig, wobei das Tritium enhaltende Dihydro-VLB ensteht.

Zusammenfassung Zur Lokalisierung der potentiell synthetisierenden mitotischen Epidermiszellen wurden Knochenfische mit H3-Thymidin injiziert, bzw. ihre Epidermis mit H3-Thymidin inkubiert. Radioaktiv markierte Zellen wurden in allen Epidermisschichten gefunden. Diese wahllose Verteilung der DNS-synthetisierenden Zellen in der Knochenfischepidermis steht im Gegensatz zu Befunden bei anderen Wirbeltieren, wo die radioaktive Markierung nur auf die Basalschicht beschränkt bleibt.

Summary Uptake of both taurocholate and indocyanine green (ICG) by isolated rat hepatocytes was saturable and obeyed Michaelis-Menten kinetics. Only uptake of taurocholate exhibited Na+-dependence, a phenomenon compatible with active membrane transport. These findings suggest that the mechanisms responsible for hepatocellular uptake of bile acids and anionic dyes differ fundamentally.This work was supported by the Swiss National Science Foundation.The authors thank Mr Th. The injection of 3-dehydroecdysone (3dhE, 5 /g), the major ecdysteroid secreted by the Y-organ of crayfishProcambarus clarkii, resulted in apolysis within about 5 days. The hormonal response at the molecular level was investigated by injection of the radio-labeled compound; within 3 h of injection of [3H]3dhE, most radio-isotope was found in the extracted epidermal tissues and identified as ecdysone, 20-hydroxyecdysone (20E), and their 3-hydroxy epimers. The biotransformation was undoubtedly performed in the peripheral area of the Y-organ. Cleavage of the polar conjugates, using an enzyme fromHelix pomatia, gave all of the above ecdysteroids including 3dhE. It was also found that the biosynthetic site of 3dhE was different from that of ecdysone at the subcellular level of the Y-organ.  相似文献

The 3 day old Chick notochord capacity to fix tritiated serotonin is maximal in its axis and in cephalic region. Our observations permitting us to find, the intracellular serotonin binding sites, contribute to an explanation of the capture mechanism and suggest a special direct role of the notochord on the monoaminergic neuron cytodifferentiation.  相似文献
