Exchange of tritium from randomly tritiated taurocholate by microbial bile salt oxidoreductases.

Tritium distribution on randomly labelled taurocholate (TC) was estimated at 28%, 4%, 1% and less than 0.5% on the hydrogens opposite the 3alpha-, 7alpha- and 12alpha-OH groups and taurine moiety respectively. Anomalously, C. perfringens 3alpha-hydroxysteroid dehydrogenase (3alpha-HSDH) catalyzed tritium loss of 36% on formation of 7alpha-, 12alpha-dihydroxy-3-keto-5beta-cholanoate, implying additional losses of tritium at other sites by this enzyme.     

Influence of common bile duct cannula size on maximal secretory rate of taurocholate in the rat

Summary The common bile duct of male Sprague-Dawley rats was cannulated with either PE 10 or PE 50 tubing. Maximal secretory rate of taurocholate averaged 389±67 (SD) and 657±115 nmoles·min^–1·g liver^–1 in the PE 10 and PE 50 group, respectively (p<0.005). Maximal bile secretory pressure was significantly higher in the PE 10 group (240±28 vs 174±8 mm H20; p<0.005). When the maximal secretory rate was exceeded, bile flow decreased in both groups but this was accompanied with a decrease in maximal bile secretory pressure in the PE 10 group only. Maximal secretory rate of bile salts is markedly influenced by experimental technique. Use of small caliber common bile duct cannulae leads to partial obstruction and decreases the apparent maximal secretory rate for taurocholate.Acknowledgments. J. Differences in utilization of tritiated thymidine and tritiated deoxycytidine by rat lymph node cells

Summary Large lymphoid cells and plasma cells in antigenically stimulated rat lymph nodes retain less label after injection of³H-CdR than after injection of³H-TdR. There is no difference amongst small lymphocytes. The data are consistent with a defect in the utilization of³H-CdR in the late stages of B cell maturation in the rat.     

Reduced microbial transformation of bile acids in cystic fibrosis

Summary The microbial transformation of bile acids by incubates of stool homogenates from children with cystic fibrosis is decreased.This work was supported by grant MT 4433 from the Medical Council of Canada and from the Canadian Cystic Fibrosis Foundation.

Intraluminal bile salt increases rate of firing in afferent fibers from the small intestine of the rat

Summary Perfusion of a rat intestinal segment with a solution containing sodium deoxycholate (8 mM) increases the rate of firing in periarterial afferent nerves from the gut. This observation indirectly supports our earlier proposal that bile salt evokes a net fluid secretion in the small intestine via an activation of the enteric nervous system.This study was supported by grants from the Swedish Medical Research Council (2855), The Medical Council of Swedish Life Insurance Companies and the Faculty of Medicine, University of Göteborg.

Zusammenfassung Das Alkaloid Vincaleukoblastine (VLB) wurde als Base bei Anwendung der Wilzbach-Technik titriert. Die Verbindung, bis zur konstanten Aktivität gereinigt, enthielt kein Dihydro-VLB mehr. Das Alkaloid, als Sulfatsalz dem Tritium unterworfen, inkorporiert die Substanz nur geringfügig, wobei das Tritium enhaltende Dihydro-VLB ensteht.

Incorporation of tritiated thymidine by teleost epidermal cells

Zusammenfassung Zur Lokalisierung der potentiell synthetisierenden mitotischen Epidermiszellen wurden Knochenfische mit H³-Thymidin injiziert, bzw. ihre Epidermis mit H³-Thymidin inkubiert. Radioaktiv markierte Zellen wurden in allen Epidermisschichten gefunden. Diese wahllose Verteilung der DNS-synthetisierenden Zellen in der Knochenfischepidermis steht im Gegensatz zu Befunden bei anderen Wirbeltieren, wo die radioaktive Markierung nur auf die Basalschicht beschränkt bleibt.

Summary In a survey of the intracellular bile salt oxidoreductase activity in fecal bacteria, 16 strains of nuclear dehydrogenating clostridia and 2 strains of non-nuclear dehydrogenatingC. paraputrificum were demonstrated unable to oxidize cholate at any of the 3 OH groups. Since nuclear dehydrogenation at the -1 and -4 position requires a 3-oxo precursor steroid, it appears that these organisms require the presence of a 3-hydroxysteroid dehydrogenating organism for nuclear dehydrogenation.This work is supported by the National Cancer Institute of Canada (IAM) and by the Cancer Research Campaign (MJH).

Disparate Na+-requirement of taurocholate and indocyanine green uptake by isolated hepatocytes

Summary Uptake of both taurocholate and indocyanine green (ICG) by isolated rat hepatocytes was saturable and obeyed Michaelis-Menten kinetics. Only uptake of taurocholate exhibited Na⁺-dependence, a phenomenon compatible with active membrane transport. These findings suggest that the mechanisms responsible for hepatocellular uptake of bile acids and anionic dyes differ fundamentally.This work was supported by the Swiss National Science Foundation.The authors thank Mr Th. The biotransformation of tritiated 3-dehydroecdysone by crayfish,Procambarus clarkii

The injection of 3-dehydroecdysone (3dhE, 5 /g), the major ecdysteroid secreted by the Y-organ of crayfishProcambarus clarkii, resulted in apolysis within about 5 days. The hormonal response at the molecular level was investigated by injection of the radio-labeled compound; within 3 h of injection of [³H]3dhE, most radio-isotope was found in the extracted epidermal tissues and identified as ecdysone, 20-hydroxyecdysone (20E), and their 3-hydroxy epimers. The biotransformation was undoubtedly performed in the peripheral area of the Y-organ. Cleavage of the polar conjugates, using an enzyme fromHelix pomatia, gave all of the above ecdysteroids including 3dhE. It was also found that the biosynthetic site of 3dhE was different from that of ecdysone at the subcellular level of the Y-organ.

Capture and fixation of tritiated serotonin by chick embryo notochord]

The 3 day old Chick notochord capacity to fix tritiated serotonin is maximal in its axis and in cephalic region. Our observations permitting us to find, the intracellular serotonin binding sites, contribute to an explanation of the capture mechanism and suggest a special direct role of the notochord on the monoaminergic neuron cytodifferentiation.     

The uptake by skeletal muscle of tritium labeled decamethonium and dimethyltubocurarine

Résumé Le décaméthonium, une substance dépolarisante, est absorbé par le muscle squelettal tandis que la substance non-dépolarisante, le diméthyltubocurarine, ne l'est pas.
