目的:探讨miR-503通过调控Bcl-2的表达介导肝癌耐药细胞株BEL-7402/5-FU对5-氟尿嘧啶敏感性的影响.方法:microRNA(miRNA)芯片筛选BEL-7402与BEL-7402/5-FU细胞中表达差异的miR-503为候选的miRNA;脂质体转染法将miR-503模拟物瞬时转染至BEL-7402/5-FU细胞;实时定量聚合酶链式反应(qRT-PCR)检测BEL-7402/5-FU及其亲本细胞中miR-503、Bcl-2 mRNA的表达水平;Western Blot观察肝癌细胞中Bcl-2蛋白的表达水平;CCK-8法检测细胞药物敏感性的改变.结果:相比于BEL-7402细胞,miR-503在BEL-7402/5-FU细胞中表达水平下调,而Bcl-2蛋白的表达水平上调;miR-503过表达后BEL-7402/5-FU细胞中的Bcl-2在mRNA和蛋白水平上表达降低;miR-503转染组的BEL-7402/5-FU细胞活力较miRNA阴性对照组显著降低.结论:miR-503可能通过下调Bcl-2的表达,进而增强BEL-7402/5-FU细胞对5-氟尿嘧啶的药物敏感性,抑制细胞增殖.  相似文献

三氧化二砷抑制肝癌细胞端粒酶活性的实验研究   总被引:6,自引:0,他引:6  
目的:观察三氧化二砷对肝癌细胞株BEL-7402和SMMC-7721端粒酶活性的影响。方法:应用端粒酶多聚酶联反应一酶联免疫测定(PCR-ELISA)方法,检测三氧化二砷作用后,肝癌细胞系BEL-7402和SMMC-7721细胞端粒酶活性的变化并比较两种细胞端粒酶对三氧化二砷敏感性的差异。结果:0.25~2.00μmol/L三氧化二砷(24~96h)可抑制肝癌细胞系BFL-7402的端粒酶活性,抑制作用与时间、剂量呈依赖性关系;0.25~0.50μmol/L三氧化二砷对SMMC-7721细胞端粒酶活性没有影响(96h以内),1.00~2.00μmol/L三氧化二砷可抑制SMMC-7721细胞端粒酶活性(4H8~96h),有时间、剂量依赖关系。结论:三氧化二砷可以抑制肝癌细胞系BEL-7402和SMMC-7721细胞端粒酶的活性;BEL-7402细胞端粒酶对三氧化二砷的敏感性较SMMC-7721细胞端粒酶高。  相似文献

目的:比较肝癌BEL-7402和SMMC-7721细胞对三氧化二砷(As2O3)诱导凋亡的敏感性差异,探讨细胞内谷胱甘肽(GSH)含量与其作用的关系。方法:不同浓度As2O3作用BEL-7402和SMMC-7721细胞24~96 h,流式细胞仪检测细胞DNA含量,计算细胞凋亡百分率;GSH检测试剂盒检测细胞内GSH含量。结果:0.25μmol/L As2O3作用24 h,有效地诱导BEL-7402细胞凋亡;对SMMC-7721细胞来说,需要用1.0μmol/L As2O3作用24 h才能达到相同的抑制效果。1.0μmol/LAs2O3作用72 h时,BEL-7402细胞的凋亡率为(43.8±0.2)%,SMMC-7721细胞的凋亡率为(12.8±0.2)%,检测BEL-7402细胞内GSH为(18.7±1.4)nmol/mg;SMMC-7721细胞内GSH为(50.8±5.2)nmol/mg,两者比较均有显著差异。结论:肝癌BEL-7402和SMMC-7721细胞对As2O3诱导细胞凋亡的敏感性不同,可能与细胞内GSH含量有关。  相似文献

目的测定中药柴胡(Bupleurun Chinese DC, BCDC)提取物对人肝癌细胞BEL-7402细胞内游离钙离子浓度([Ca2 ]i)的影响,以探索柴胡逆转BEL-7402细胞多药耐药的机制.方法以Fura-2/AM为细胞内钙离子的荧光指示剂,用双波长荧光分光光度计分别测定不同条件下的BEL-7402细胞内[Ca2 ]i.结果柴胡可使人肝癌细胞BEL-7402细胞内游离钙离子浓度下降(P<0.001).结论柴胡可使人肝癌细胞BEL-7402细胞内游离钙离子浓度下降,为一种钙离子通道阻滞剂(Calcium channel blocker, CCB),提示钙离子通道阻滞作用为柴胡逆转BEL-7402细胞多药耐药的机制之一.  相似文献

目的测定中药柴胡(Bupleurun Chinese DC,BCDC)提取物对人肝癌细胞BEL-7402细胞内VCR蓄积的影响,以探索柴胡逆转BEL-7402细胞多药耐药的机制.方法高效液相色谱法测定BCDC作用于BEL-7402后细胞内VCR蓄积情况的变化.结果柴胡可使人肝癌细胞BEL-7402细胞内VCR浓度升高(P<0.01).结论柴胡可以增加VCR在BEL-7402细胞内的积聚浓度,部分逆转BEL-7402细胞的MDR.  相似文献

为研究牛膝多糖对肝癌BEL-7402细胞凋亡及相关蛋白Caspase-3表达的作用,采用不同浓度ABPS作用于肝癌BEL-7402细胞48h,MTT法检测细胞生长抑制率,荧光显微镜观察细胞形态变化,Western Blot检测Caspase-3蛋白表达.实验结果表明,ABPS对BEL-7402细胞具有生长抑制作用,促进细胞凋亡,作用机制可能与上调Caspase-3蛋白表达有关.  相似文献

目的:探讨水飞蓟宾诱导人肝癌HepG2细胞凋亡并提高5-FU、顺铂的敏感性.方法:采用MTT法和克隆形成抑制实验观察不同浓度的水飞蓟宾对人肝癌HepG2细胞增殖抑制作用;Hoechst 33258染色法检测水飞蓟宾作用于人肝癌HepG2细胞后其细胞核形态的改变;Western blotting检测凋亡相关蛋白(Caspase-3,Caspase-9)的表达;采用水飞蓟宾联合5-氟尿嘧啶(5-FU)、顺铂(DDP),观察对人肝癌HepG2细胞增殖抑制作用.结果:MTT法检测显示,水飞蓟宾对肝癌HepG2细胞有明显的增殖抑制作用,并随着水飞蓟宾浓度的增大而增强,作用于肝癌HepG2细胞48h的IC50为195.38 μmol/L;克隆形成抑制实验表明随着水飞蓟宾药物浓度的增加,细胞克隆形成逐渐减少,与对照组相比有显著差异;其细胞核固缩、边聚、裂解等细胞凋亡形态学变化;存在Caspase-3和Caspase-9蛋白的活化和降解;150 μmol/L的水飞蓟宾与不同质量浓度的化疗药物(5-FU,DDP)联合作用于HepG2细胞48h,可提高HepG2细胞对这些化疗药物的敏感性,增敏倍数分别为39.63和21.54倍.结论:水飞蓟宾通过诱导人肝癌HepG2细胞凋亡抑制细胞增殖,并提高HepG2细胞对5-FU,DDP的敏感性.  相似文献

探讨维药异常黑胆质成熟剂(ASM)抗肿瘤作用及其对肿瘤细胞迁移的影响.体外培养人肝癌Bel-7402细胞株、人乳腺癌BCAP 细胞株、人宫颈癌 Hela 细胞株、人胃癌BGC-823细胞株,采用四甲基偶氮唑蓝(tetrzolium-based colorimetric assay,MTT)法检测不同浓度ASM(1、2.5、5、10、15、20、50mg/mL)对人肝癌Bel-7402细胞株、人乳腺癌BCAP细胞株、人宫颈癌Hela细胞株、人胃癌BGC-823细胞株4种肿瘤细胞增殖的影响;选择人肝癌Bel-7402细胞株为研究对象,采用细胞划痕运动实验和扫描电镜技术,观察不同浓度ASM对肝癌细胞迁移的影响.MTT检测结果显示,ASM对人肝癌Bel-7402细胞株、人乳腺癌BCAP细胞株、人宫颈癌Hela细胞株、人胃癌BGC-823细胞株生长均有一定抑制作用,且在15~20mg/mL作用下最为明显,对不同肿瘤细胞株抑制作用不一;体外细胞划痕运动实验和扫描电镜结果显示,ASM对人肝癌Bel-7402细胞运动有一定抑制作用.由此推论,ASM对肿瘤细胞增殖和肿瘤细胞迁移有一定抑制作用.  相似文献

应用增殖抑制方法发现,无生长抑制作用的0.5mg ·L~(-1)的蝙蝠葛碱(Dau)使博莱霉素A_5单独对小鼠肉瘤S-180V细胞作用的I_(C50)值从2.1mg·L~(-1)降低到1.3mg·L~(-1),无抑制作用的1 mg·L~(-1) Dau使博莱霉素A_5单独对人喉癌HEP2细胞作用的I_(C50) 值从0.33 mg·L~(-1)降低到0.1 mg·L~(-1).克隆形成法证明:无细胞毒性的10 mg·L~(-1)的Dau明显增强博莱霉素A_5对HEP2细胞毒性.TFP法证实,有一定生长抑制作用的Dau对HEP2细胞内活化钙调素有影响.结果表明:蝙蝠葛碱有一定的增强博莱霉素A_5对肿瘤细胞的毒性.  相似文献

为研究超级黑糯玉米芯色素(super black glutinous corncob pigment,SBCP)对肿瘤细胞的增殖抑制效应和体外抗氧化活性.采用MTT法检测了SBCP对人肝癌细胞系BEL-7402以及胃癌细胞系SGC-7901增殖的影响;在体外化学模拟的条件下,通过建立DPPH自由基、羟基自由基、超氧阴离子自由基、还原力试验,采用比色法测定对比了SBCP和抗坏血酸(Vc)对DPPH自由基、羟基自由基和超氧阴离子的清除作用以及还原能力.结果表明SBCP对BEL-7402细胞和SGC--7901细胞增殖有明显的抑制作用,且随药物浓度的增大而增强;SBCP对两种肿瘤细胞的半数抑制浓度(IC50)分别为4.13 mg/mL和3.83 mg/mL.SBCP还具有较好的抗氧化作用,对羟基自由基和超氧阴离子的清除作用分别是Vc的1.4倍和1.23倍,还原能力是Vc的1.77倍,但对DPPH自由基的清除作用略低于Vc.SBCP在癌症预防和治疗以及食品保健方面具有潜在的应用价值;SBCP具有很好的体外抗氧化活性.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
