天花粉蛋白抗人免疫缺陷病毒1型研究

目的检测用基因工程方法制备的天花粉蛋白抗人免疫缺陷病毒1型的活性。方法将人免疫缺陷病毒1型在细胞系中培养,加入天花粉蛋白后,观察细胞病变情况,检测P24抗原及逆转录酶的活性。结果加入天花粉蛋白的细胞始终未见细胞病变,P24抗原及逆转录酶检测均为阴性;阴性对照组细胞在第7 d以后均出现人免疫缺陷病毒1型感染典型细胞病变,P24抗原及逆转录酶检测均为阳性;阳性对照组始终未见HIV-1感染典型细胞病变,逆转录酶的活性检测和P24抗原检测结果均为阴性。结论天花粉蛋白在培养细胞系中可以有效地抑制人免疫缺陷病毒1型。     

天花粉蛋白基因的克隆、表达和活性鉴定

用PCR（Polymerase Chain Reaction)扩增出天芬粉蛋白（Trichosanthin,TCS)基因全长序列，插入表达载体，经双酶切鉴定及测序分析，表明阅读框正确，表达His-TCS活性融合蛋白并纯化天花粉蛋白，鉴定出其具有RNA N－糖苷酶活性和抗原抗体结合活性。     

天花粉蛋白抗单纯疱疹病毒1型的实验研究

检测用基因工程方法制备的天花粉蛋白抗单纯疱疹病毒1型的活性。将单纯疱疹病毒1型在细胞系中培养．加入天花粉蛋白后,观察细胞病变情况,用酶联免疫吸附（ELISA）的方法检测病毒生长情况。结果显示,加入天花粉蛋白的细胞始终未见细胞病变,ELISA检测为阴性;阴性对照组细胞在第3d以后均出现单纯疱疹病毒1型感染典型细胞病变．ELISA检测结果为阳性;阳性对照组始终未见单纯疱疹病毒1型感染典型细胞病变,ELISA检测结果为阴性。天花粉蛋白在体外培养细胞系中可以抑制单纯疱疹病毒1型。     

天花粉蛋白基因克隆、序列分析及在E.Coli中表达

天花粉蛋白为单链核糖体失活蛋白,具糖苷酶活性,对 HIV 和多种动植物病毒有明显的抑制作用,有广泛的理论价值和应用前景.本文在国内首次采用聚合酶链式反应(PCR)方法,以栝楼染色体 DNA 为模板、合成的寡聚核苷酸为引物,扩增出了编码成熟天花粉蛋白的 DNA 片段(称 gene 1)和编码含23个氨基酸组成的分泌型信号肽、成熟天花粉蛋白以及羧基端延伸因子在内的 DNA 片段(称 gene 2).两 DNA 片段已被组装于载体     

天花粉蛋白与腺嘌呤、单磷酸腺苷、三磷酸腺苷之间的相互作用

利用荧光光谱和透析平衡法研究了腺嘌呤、单磷酸腺苷（ＡＭＰ）、三磷酸腺苷（ＡＴＰ）与天花粉蛋白之间的相互作用。荧光光谱揭示腺嘌呤与天花粉蛋白结合后对其荧光强度影响较小，但使蛋白的荧光峰位红移，而ＡＭＰ和ＡＴＰ只引起蛋白荧光强度下降而不改变峰位。ＡＭＰ，ＡＴＰ引起的荧光强度下降包含着快慢两个过程。透析平衡法测得ＡＭＰ，ＡＴＰ与天花粉蛋白相互作用的结合常数分别为１．２×１０￣（－１）ｍｏｌ／Ｌ和４．５×１０￣（－５）ｍｏｌ／Ｌ，二者自由能之差为－２．２６ｋＪ／ｍｏｌ。     

天花粉与甲氨喋呤保守治疗异位妊娠40例临床疗效比较

目的:比较天花粉蛋白肌肉注射与甲氨喋呤(MTX)肌肉注射保守治疗异位妊娠的有效性和安全性.方法:将40例异位妊娠患者随机分为两组[甲氨喋呤(A)组20例,天花粉(B)组20例].分别采用肌肉注射天花粉和甲氨喋呤,对两组患者治疗后血β-HCG下降情况,药物不良反应以及药物耐受性等指标进行量化分析.结果:两组患者治疗前组间差异无统计学意义,治疗有效率B组为86.31%,A组为78.25%,组间差异有统计学意义(t=2.543,P=0.018);治疗不良反应B组明显小于A组.结论:肌肉注射天花粉蛋白治疗异位妊娠是安全、有效的,在治愈率和患者药物耐受力方面明显优于甲氨喋呤,有一定推广价值.     

天花粉蛋白结构改造对引产活性的影响

目的：研究结构改造对天花粉蛋白（ＴＣＳ）引产活性的影响。方法：利用定点突变将ＴＣＳ第２１９位的天冬酰胺残基改变为半胱氨酸残基（Ｑ２１９Ｃ）以供定点聚乙二醇（ＰＥＧ）修饰,测定修饰产物的致本失活活性及小鼠中期引产活性。结果：ＰＥＧ修饰使的致核糖体失活活性约降低９０％;相对分子质量为５０００的ＰＥＧ修饰对引产活性无明显影响,而相对分子质量为２００００的ＰＥＧ修饰却明显增强引产活性;此外,ＰＥＧ修饰使Ｔ     

王瓜根蛋白的柱层析分离及其抗生育作用

应用Ambelie.IRC.50柱层析分离纯化王瓜根抗早孕蛋白,图谱显示抗早孕蛋白达到基线分离,用SDS.PAGE不连续电泳证实是电泳纯。应用本工艺生产的王瓜根抗早孕蛋白作小白鼠抗着床试验,其ED_(100)(100％抑制率)为1.5mg/kg,是结晶天花粉ED_(100)(10mg/kg)的6倍以上,提示这种蛋白有可能作妇女房事后避孕的新药。对应用王瓜抗早孕蛋白注射后的小白鼠子宫作组织学鉴定,由显微组织切片显示王瓜根抗早孕蛋白较天花粉蛋白作用强,子宫组织发生了变化。用王瓜根做抗早孕蛋白口服试验,也证实其效果好。这提示王瓜根蛋白可制成简便口服避孕片。     

巯基丙酸在金电极上的自组装及天花粉蛋白在它上面的电化学

巯基丙酸在金盘电极表面采用不同的时间进行自组装，可以观察到循环伏安曲线中峰电流(ip)的改变。通过石英晶体金电极在组装巯基丙酸前后频率改变趋势的不同来讨论自组装的结果并观察巯基丙酸在一定电位下的脱落。利用巯基丙酸与天花粉蛋白之间正负电荷的静电吸引作用，在巯基丙酸达到饱和吸附后的金电极表面组装上天花粉蛋白，利用循环伏安法表征其在自组装电极上的电化学性质。     

天花粉蛋白诱发人体免疫抑制中DQA1和DQB1等位基因的顺式互补受邻近DQ基因的调变

人类白细胞抗原HLA－DQ等位基因DQA1*0501和DQB1*0201以顺式和反式两种互补格局决定中药提取物天花粉蛋白诱发人体免疫抑制中CDS介导型性状的表达．研究工作进一步揭示其中的顺式互补受另一单元型上DQB1等位基因的调变．其中起正向调变作用的等位基因是DQB1*0302，0303，0401，0601；发挥负向调变作用的等位基因是DQB1*0201，0501，0602．以上现象表明天花粉蛋白诱发人体免疫应答的遗传控制呈现十分复杂的格局．     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
