不同培养基对土壤细菌可培养性的影响

采用不同培养基培养土壤微生物,以提高土壤细菌可培养性。以番茄大田和温室土壤为材料,采用4种培养基进行细菌的分离培养,经16SrDNA分子生物学鉴定,明确不同培养基获得细菌的种群差异。结果表明：在细菌数量方面,2种土样分别在各种培养基上生长的细菌数量未达到显著性差异,但培养基间细菌数量差异较大,LB培养基分离的细菌数量分别是YEM Medium,TWYE Medium和Flour Medium培养基的2.04倍、1.88倍和3.5倍;在细菌多样性方面,细菌在温室土分离的种类多于在大田土中分离的种类,分离细菌的多样性从大到小为LB>TWYE Medium>Flour Medium>YEM Medium培养基。此外,在LB和TWYE Medium培养基中各获得一株潜在新种。综上,LB培养基分离的细菌数量和多样性最佳,其他培养基在细菌多样性方面具有补充作用。     

杭州西溪湿地沉积物中不同培养基分离的细菌种群多样性

以西溪湿地沉积物的细菌群落为对象,对4种不同培养基(M1、R2A、NB和DNB)分离细菌的能力及分离物的种群多样性进行了比较研究.共分离得到191株菌株,经16SrRNA基因测序和系统发育分析,可归于13个不同的属,其中R2A培养基分离的细菌种类最多,共有11个不同的属,其次是NB培养基,分离得到8个属,M1和DNB培养基均分离得到7个属.优势种属为根瘤菌属(Rhizobium),占总数的39.27%.研究结果表明,R2A培养基分离的培养物种群多样性更为丰富.     

酸奶中嗜热链球菌的分离及鉴定

对黑龙江省牡丹江市"三道"牌酸奶的乳酸菌进行了分离,分离培养基BCG牛乳培养基.分离得纯菌,革兰氏阳性,链球状排列,能发酵葡萄糖产乳酸,确定其为嗜热乳酸链球菌.     

刺五加内生放线菌的分离与多样性研究

从采集自5个不同地点的刺五加全株植物,采用8种分离培养基,从植株不同部位分离,经BOX-PCR排重后,共得到内生放线菌265株.从不同培养基分离出的放线菌数目有较大差异.LNMS培养基分离菌株最多,60株.海藻糖-无机盐培养基分离最少,17株.从刺五加植株根及根茎分离到的放线菌数量最多,占菌株总数的77%,叶中分离的最少,为6%.从吉林柳河和吉林梅河口植株内分离的菌株较多.利用16SrRNA基因序列分析鉴定,菌株分属于7个亚目,9个科,9个属.     

普通培养基与优化培养基对防城港海域真菌分离效果的影响

以Pilidium concavum为例探讨真菌培养条件的优化

在基物上筛选出一株Pilidium concavum真菌,以Pilidium concavum菌株为例研究了适合其生长和产子实体的培养基配方。在培养基含酵母膏3g/L、蔗糖20 g/L的情况下,Pilidium concavum生长和产子实体效果最佳。优化培养基中添加孟加拉红3mg/L可作为选择性培养基使用。本次试验优化的培养基具有清澈透明,便于观察分离,配置方便等优势,促进了菌物分离工作的开展。     

几种乳酸菌的分离与筛选

本文通过选用较为简单的培养基和较为简单的实验流程,从自然发酵泡菜杂居混生的微生物中,分离筛选出具有优良发酵特性的乳酸菌菌种。根据各类微生物的形态结构和新陈代谢类型,利用合适的分离培养基和鉴别性培养基,结合显微镜镜检、菌落特征以及相关生化特性进行鉴定筛选。为发掘新的乳酸菌种或新菌种的增殖培养基的研制与改良提供些许参考。     

平菇(P123)母种培养基的筛选试验

通过对平菇(P123)进行组织块分离,用六种不同配方培养基进行对比培养,选择出适合于平菇(P123)母种分离和保存的培养基配方.实验结果表明,酵母粉蔗糖培养基适合于平菇(P123)的母种分离,马铃薯葡萄糖培养基适合于平菇(P123)的菌种保藏.     

高效脱酚菌的筛选及微生物多样性分析

为了揭示不同培养基分离的细菌种群的多样性差异及获得高效脱酚菌,采用单链构象多态性技术,以16S rRNA基因的V3区为靶对象,对3种不同培养基SJ(自制培养基)、LB(牛肉膏蛋白胨培养基)、YEB(酵母牛肉膏培养基)分离细菌的种群多样性进行了比较研究.结果表明:SJ培养基比LB和YEB培养基有更高的种群多样性,更适于细菌的分离筛选.利用SJ培养基筛选出的3株高效脱酚菌降解废水中总酚的能力均在99%以上,经过生理生化鉴定和16S rDNA测序,建立了系统发育树,鉴定出这3株菌分别属于气单胞菌属(Aeromonas)、芽孢杆菌属(Bacillus)和不动细菌属(Acinetobacter).     

六种培养基对金针菇(F19)母种分离对比试验

对金针菇（E19）进行组织块分离实验中,参照有关材料,选出6种不同配方的培养基进行对比实验,意在选择适合于金针菇母种分离和保存的培养基配方.实验证明,玉米粉综合培养基比较适合于金针菇的母种分离,CPDA比较适合于金针菇的菌种保藏.     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. 低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
