Summary A thin film gold electrode for biological oxygen measurement was constructed using gold paste. This electrode is easy to construct, has high sensitivity and is disturbed only slightly by bubbling of the surrounding fluid. A method of measuring and studying metabolic bacterial activity is proposed, by following the kinetic evolution of the ratio of the oxidized and reduced forms of an electron transporter as a consequence of decreasing oxidizing power--due to oxygen consumption in the culture,--and increasing. Reduction power of bacterial activity. Namely, with minimum composition using salts and glucose the oxido-reduction of lipoic acid is well indicated by a gold electrode without any major bio-or electrochemical interference. A kinetic diffusion reaction theory takes into account the passive or active transmembrane transport of lipoic acid in good agreement with the experimentally observed shapes of the electrical signal. The various types of antibiotic activities are well reflected by the modifications of the signal.  相似文献

Summary Similar diurnal periodicity in oxygen consumption of liver slices in Wistar rats was observed as that previously found in Sprague-Dawley rats. The rate of oxygen consumption was low in the morning and high in the evening. After inversion of lighting regimes, the phase shifted and reached the reversal curve in about 30 days. On the basis of these findings, it is estimated that diurnal periodicity in oxygen consumption of liver slices is influenced by alteration of the periods of light and darkness. Liver glycogen rhythm showed a reversed correlation to that of oxygen consumption in both lighting regimes.  相似文献

The thermogenic action of beta-adrenoceptor agonists may be due, in part, to increased metabolism in skeletal muscle. Previous results suggest that vasoconstriction is also necessary, and that the effect can be blocked by vasodilators. Both noradrenaline and the beta-3 agonist, ICI D7114, were studied using two dog hindlimb protocols. During constant perfusion conditions, ICI D7114 caused a significant increase in hindlimb oxygen consumption although it is a vasodilator. Noradrenaline resulted in a smaller rise in oxygen consumption, and produced a marked vasoconstriction. Both noradrenaline and ICI D7114 resulted in decreased oxygen consumption when the blood flow was allowed to vary in response to the drug treatment. The results suggest that changes in tissue oxygen consumption caused by beta-agonists are not related to vasomotion.  相似文献

Summary A method is described for simultaneous measurements of pO2 and action potentials with microelectrodes using prefabricated gold wires. The construction and measuring with such gold electrodes is easier than with traditionally used platinum electrodes. Single electrode recording is done with a specially adapted electronic circuit, allowing the separation of both signals (pO2 and action potentials. The effect of treatment with acetyl-L-carnitine on hepatic mitochondrial respiration and biosynthetic function in perfused liver from young (90 days) and old (22-24 months) rats was studied. Rats were given a 1.5% (w/v) solution of acetyl-L-carnitine in their drinking water for 1 month and oxygen consumption together with the rate of gluconeogenesis, urea synthesis, and ketogenesis with and without added substrates were measured in perfused liver. Mitochondrial oxygen consumption was also assessed in liver homogenate and isolated mitochondria to determine the maximal capacity for oxidative phosphorylation. Acetyl-L-carnitine treatment almost completely restored the age-dependent decline in oxygen consumption, gluconeogenesis, urea synthesis, and ketogenesis found in perfused liver of old rats to the levels found in young rats. In addition, acetyl-L-carnitine treatment increased oxygen consumption and biosynthetic function in perfused liver from young rats. After acetyl-L-carnitine treatment, we found detectable 3-oxoacyl-CoA-transferase activity associated with a consumption of ketone bodies in young and old rats. Finally, oxygen consumption measured in homogenate and isolated mitochondria did not change with age and acetyl-L-carnitine treatment. Our results show that in perfused liver, acetyl-L-carnitine treatment slows the age-associated decline in mitochondrial respiration and biosynthetic function. In addition, treatment of young rats with acetyl-L-carnitine has a stimulating effect on liver metabolism, probably through an increase in ATP production. Summary The study of the relationship between the activity of the thyroid of Cichlid fishes and oxygen consumption has given the following results: The stimulation by thyroxine causes a significant increase of oxygen consumption (+35%) after a long treatment. This increase is not immediate; a latent period is observed till the gland is in repose. The antithyroid rapidly decreases the oxygen consumption (–24%). Metasystox exposure decreased the oxygen consumption of all tissues. Maximum reduction is noted in gill followed by brain and muscle. Some physicians use the electrical conductance of the skin, particularly at the acupuncture points, for diagnostic purposes. This paper deals with the quantification of the skin conductance at some acupuncture points under well defined conditions using the electrode materials gold, graphite, silver and brass. The average oxygen utilization coefficient of 26% is calculated from PO2 determinations during the perfusion of the fetal circulation of isolated human term placental lobules. The oxygen consumption approximates 0.4 ml/min/kg placenta when Earle's salt solution is used as a perfusion medium and is equilibrated with a gas mixture containing 5% O2. The oxygen consumption reaches 2 ml/min/kg when the same Earle's solution is equilibrated with a gas mixture containing 95% O2.  相似文献

Summary The quantitative study of whole liver DNA and liver nuclei DNA at various ages, and the simultaneous study of oxygen uptake shows the following facts:(1) The rate of oxygen consumption to the fresh weight or to the DNA or to diploid chromosomes group is the same for the adult or the young aged 8 days. The values of oxygen consumption calculated for one cell or for one nucleus (taking notice of the fact that the mean level of liver nuclei DNA varies with the age) at 8 days is only the half of the adult value. Even at two months the value is 20% lower than in the adult.(2) In older animals there is a decrease of oxygen uptake which is more noticeable on a cellular scale than with the classical rate to fresh or dry weight. We affirm the fact that the diminution of the oxygen uptake of each cell is compensated by the increase of the number of cells.  相似文献

Summary Camphor has been found to decrease the rate of oxygen consumption by rat kidney mitochondria. The rate of oxygen consumption is nearly halved by the addition of 8×10–3 M camphor. It is suggested that camphor may be of use in oxygenating tumours prior to radiotherapy.Acknowledgments. Summary Male rats were housed in continous illumination or blinded when 21 day-old and killed 69 days later. The continuous illumination exposure increased the weights of testes and sex accessory organs and reduced the pineal gland weight. Blindness decreased weights of testes, sex accessory organs and anterior pituitary. The oxygen consumption rate of the hypothalamus was higher in the blinded animals than in the controls and lower in the continuously illuminated rats. No one of such groups showed significant changes in the oxygen consumption by either the amygdata or the hippocampus.  相似文献

Summary Camphor at <8 moles/mg protein reduced the rate of oxygen consumption by rat liver mitochondria. The effect occurs only with NAD+-linked substrates. Succinate linked respiration was inhibited but this appears to be caused by some conversion of succinate to malate. At higher levels, camphor increases oxygen consumption with succinate substrate, by uncoupling at site II.Acknowledgment. The steady state oxygen consumption of two echidnas was measured during locomotion on a treadmill. The change in power input with change in velocity is similar to that found in other mammals, but the total energy requirement for locomotion is less. Neonatal mice were injected with monosodium glutamate (MSG) followed 10 days later by treatment with gold thioglucose (GTG). Weight gain, food consumption and body lipid determinations were made. Results indicated that pretreatment with MSG does not block subsequent response to GTG and the resulting changes are similar to those induced by GTG alone.  相似文献
