An uptake of fluorescein isothiocyanate labeled neocarzinostatin into the cancer and normal cells.

An uptake of fluorescein isothiocyanate labeled neocarzinostatin into normal and cancerous epithelial cells from bladder was investigated. Results showed that neocarzinostatin traversed the cell membrane into cytosol and nuclei, and it appeared to have a preferential cytotoxicity for the cancer cell.     

Morphogenesis of human colon cancer cells with fetal rat mesenchymes in organ culture

Summary The morphogenesis and cytodifferentiation of human colon cancer cells (LS174T and HT29) were examined by combining cancer cells with fetal rat digestive-tract mesenchyme in organ culture. LS174T cells migrated into the mesenchyme to form glandular structures composed of single columnar cells with their nuclei oriented basally, while HT29 cells formed cell masses with little lumen formation. Immunohistochemical studies with antibodies against carcinoembryonic antigen and secretory components showed that the composition of cell surface glycoproteins was not necessarily reversed to the normal type, even when neoplastic cells exhibited normal glandular structures.This work was supported by grants-in-aid for Cancer Research from the Ministry of Education, Science, and Culture, Japan, and by the Veterans Administration Medical Research Service, USA. Immigration of lymphoid precursor cells into the thymic rudiment inXenopus

Summary The origin of thymic lymphocytes was investigated, using a new reliable method to mark cells inXenopus. It was easily observed that extraneous cells immigrated into the thymic rudiment 4 days after fertilization and differentiated into a cell population identified as thymic lymphocytes in a fully developed thymus. Vitamin A and tumor immunity

Summary Intraperitoneal administration of vitamin A into the BALB/c mice inoculated with a syngeneic fibrosarcoma, Meth A, caused a remarkable augmentation of tumor rejection. A cell-depletion technique revealed that the primary effector cells responsible for the augmented rejection were Thy-1 positive, Lyt-1 negative, Lyt-2 positive lymphocytes, suggesting the involvement of cytotoxic lymphocytes.This work was supported in part by a Grant-in-Aid for scientific research from the Ministry of Education, Science and Culture, Japan, and in part by a Grant-in-Aid for cancer research from the Fukuoka Cancer Society, Japan.We thank Mr M. Lipid peroxidation caused by chinoform-ferric chelate in cultured neural retinal cells

Summary Incorporation of chinoform-ferric chelate was demonstrable in cultured neural retinal cells of chick embryos after 1 h of incubation, and the lipid peroxide level in the cells was increased strikingly 1 h thereafter. On the other hand, free ferric ions were scarcely incorporated into the cells, and a significant increase in the lipid peroxide level in the cells was not observed. These data indicate that chinoform is carrier of iron for its passage through cell membranes and that the incorporated iron induces lipid peroxidation which in turn leads to neural cell degeneration.This work was supported in part by a grant from the Ministry of Health and Welfare of Japan     

Ocular involvement in hamsters transplanted with a human leukemic T-cell line

Summary A leukemic T-cell line (TALL-1) was serially transplanted for 5 passages into newborn hamsters treated with antilymphocyte serum. This cell line was derived from a leukemic patient with clinical evidence of ocular involvement. I.p. implantation of 1–3×10⁷ cells resulted in disseminated growth of tumors in all 15 recipients after 23–41 days and 8 of them showed leukemic infiltration of the uveal tract of one or both eyes.Supported by a Cancer Research Grant from the Ministry of Health and Welfare of Japan.     

Quantitation of proinsulin mRNA sequences in hamster insulinoma cells in culture by molecular hybridization

Summary The content of proinsulin mRNA sequences was measured in a cultured cell line established from a transplantable hamster islet cell tumor, by hybridization with proinsulin cDNA. The cells cultured in vitro were found to contain a significant amount of proinsulin mRNA sequences, compared with non-insulin-producing hamster tissues, and seem to be a useful system for the study of insulin gene expression.This work was supported in part by Grants-in-Aid Scientific Research (248129, 387065, 337013, 378052) and a Grant-in-Aid for Cancer Research (301538) from the Ministry of Education, Science and Culture, Japan.     

Inhibition of testicular androgenesis by urinary gonadotropin-inhibiting substances in rats

Summary The effect of urinary gonadotropin-inhibiting substances (GIS) on the androgen synthesis in rat testes was studied in vitro and in vivo. GIS, which was added to the incubation medium containing teased testicular tissues and injected into rats for 2 days, showed a suppressive effect on the formation of androstenedione from pregnenolone in the testis.Supported by a Scientific Research Grant from the Ministry of Education of Japan.     

2/8 translocation in a Japanese Burkitt's lymphoma

Summary A new translocation between chromsomes 2 and 8 t(2p^–; 8q⁺), was found in fresh lymphoma cells from a Japanese patient with Epstein-Barr virus-carrying Burkitt's lymphoma, and in a lymphoma cell line derived from this patient. There was no 14q⁺ translocation, as has been previously described in African and North American Burkitt's lymphomas.Supported by a grant-in-aid for Cancer Research (52-11) from the Ministry of Health and Welfare of Japan.     

Cardiac hypertrophy in surgically denervated dogs with aortic stenosis

Summary Left ventricular cell hypertrophy in dogs with aortic stenosis was accelerated by surgical denervation of the left ventricle. We conclude that there are neural mechanisms which, when present, inhibit cardiac cell hypertrophy.This work was supported by a grant of the Ministry of Education in Japan for 1981.     

Quantitative studies on in vitro transformation of hamster chondrocytes

Summary Hamster chondrocytes could be transformed in a quantitative assay system which used X-irradiated feeder layer cells. Morphological transformation occurred on addition of, 4NQO, but not in control cultures. Differentiation was classified into 3 types (good, poor and none); normal and transformed colonies contained similar proportions of the 3 types.This work was supported by a grant from the Ministry of Education, Science and Culture, Japan.We thank Miss M. Improved separation at low temperature of glycoproteins by Con A-Sepharose affinity chromatography in the presence of sodium dodecyl sulfate (SDS)

Summary The Con A-Sepharose affinity chromatography of glycoproteins was even more effective at 4°C than that at room temperature (26–28°C) in the presence of sodium dodecyl sulfate (SDS). Application of this methodology to the seperation of several glycoproteins from SDS-solubilized membrane proteins in rat cerebellum, including a glycoprotein characteristics of the Purkinje cells, was sucessful.Acknowledgments. Immuno-electron microscopic localization of fibronectin on rat mast cells

Summary Rabbit anti-rat plasma fibronectin (pFN) causes histamine release from rat mast cells in the presence of complement. Fibronectin (FN) on rat mast cells, as shown by immuno-electron microscopy, is principally localized on cell folds, so they may play a role of attachment in the matrix of connective tissue.Acknowledgment. The growth responses of hamster chondrocytes,dermal fibroblasts and embryo cells to whole blood serum and plasma-derived serum

Summary Autoradiographic studies with³H-thymidine demonstrated that the growth responses of hamster chondrocytes, dermal fibroblasts and embryo cells, respectively, differed in media containing whole blood serum (WBS) and plasmaderived serum (PDS). Dermal fibroblasts seemed to require a growth factor from platelets for growth, but chondrocytes did not. A human calcitonin-like molecule in the ultimobranchial body of the amphibia (Rana pipiens)

Summary As assessed by radioimmunoassay, and high performance liquid chromatography (HPLC), frog ultimobranchial calcitonin was found to be similar to synthetic human calcitonin but completely different from synthetic salmon calcitonin.Acknowledgments. The dopaminergic innervation as observed by immunohistochemistry using anti-dopamine serum in the rat cerebral cortex

Summary By using an antiserum raised against dopamine bound to bovine serum albumin, thinner dopamine-labeled nerve terminals were visualized immunohistochemically within neocortical areas, in addition to well-documented dopaminergic innervation into the prefrontal and limbic cortices.This study was supported in part by a Grant-in-Aid for Scientific Research on Priority Areas, Japanese Ministry of Education, Science, Culture (No. 62623002).     

Ultrastructural changes in uterine myometrium of mice with experimentally-induced adenomyosis

Summary Ectopic pituitary transplantation induced a high incidence of adenomyosis in SHN mice. Early signs of the development of adenomyosis were the penetration of stromal connective tissue into myometrium followed by uterine gland invasion. Involvement of prolyl endopeptidase in ascidian fertilization

Summary Inhibitory effects on fertilization of the ascidian of three benzyloxycarbonyl(Z)-aminoacyl prolinals and Z-Gly-Pro-chloromethyl ketone added before and after insemination were examined. The results suggest that the prolyl endopeptidase is involved in the process of fertilization, especially in a process taking place between chorion elevation and cell cleavage.Acknowledgment. 岷江源头区水土流失模拟计算与动态分析

通过对卫星遥感图片的解译以及ArcGIS软件的应用,借助已有水文数据和通用水土流失方程(USLE)模拟计算了岷江源头区1974年、1994年以及2002年的年水土流失总量。同时,按照国家水利部公布的水土流失强度标准,将该区域分为无流失区、轻度流失区、中度流失区和强度流失区四个水土流失等级。结果表明:(1)该区域年水土流失总量从1974年到2002年呈下降趋势,1994年~2002年减少的速度远远大于前20年。(2)研究区域的水土流失强度有从流失严重的等级向流失轻微的等级转化的趋势,从转化面积来看,主要是由轻度流失区向无流失区转化以及中度流失区向轻度流失区转化。从转化比例来看,强度流失区减少得最多,大多转化为中度流失区。     

人结直肠癌细胞系的成球培养方法及转移侵袭能力研究

目的探索人结直肠癌细胞系形成肿瘤细胞球的培养方法及球体内间质化标志物的表达检测。方法结直肠癌细胞系SW480、SW620、LOVO、HT29及HCT116在添加了不同细胞生长因子的无血清培养基（SFM）中悬浮培养,传代更新,诱导分化。免疫荧光技术和RT-PCR分别检测细胞球中间质化标志物N—cadherin及Vimentin的表迭情况。结果5种细胞系均能在特制的SFM中形成细胞球,并稳定的传代增殖。在用血清诱导分化后,又重新贴壁生长,且与亲代细胞保持一致的细胞形态。细胞球中间质化标志物N—cadherin、Vimentin表达均上调。结论结直肠癌细胞系在低黏附及添加了细胞生长因子R—Spondin 1、Noggin的无血清环境中更容易形成悬浮生长的肿瘤细胞球,细胞球细胞比亲代细胞更具有转移侵袭能力。
