Summary The3H-oestradiol receptor complex obtained from the cytosol fraction of foetal guinea-pig uterus was purified by the following steps: column chromatography in Sephadex G-15 and Ultrogel, and by electrofocusing on polyacrylamide plates. In the final step a concentration of 15–17% of the foetal uterine oestradiol receptor protein was obtained. The isoelectric point (pl) of this receptor was determined to be 6.1–6.2.The expenses of the investigation were partially defrayed by a grant from the Centre National de la Recherche Scientifique (CNRS), France (Equipe de Recherche CNRS No. 187).  相似文献

Isolated islets of Langerhans from 21.5 day-old foetal Rats were studied in a perifusion system in vitro. The overall dynamics of insulin release by the foetal islets in response to glucose 13.9 mM is biphasic and qualitatively similar to that obtained with islets of adult Rats. The magnitude of the initial phase of insulin secretion is similar for the foetal and adult islets. The second phase is fourfold higher in the adult than in the foetus. The response of foetal islets occurs, 45 to 60 sec. after the increase of glucose concentration in the medium and a maximum insulin release for the first phase is obtained with a lag period of 2 min. The difference between foetal and adult islets is essentially quantitative.  相似文献

Summary In rats, placental gamma glutamyl transpeptidase (GGTP) specific activity declined linearly during the last third of gestation. In contrast, the kidney enzyme activity progressively increased 15-fold. In the gut, peak activity was reached at mid-lactation (age 12 days). Hepatic GGTP levels were maximal during the foetal stage. Brain GGTP was negligible at birth and attained its highest specific activity at weaning.This work was supported in part by USPHS NIH SO8 RR-09128-03.  相似文献

Glycolysis was assessed in the isolated foetal Rat in hypoxia. Measurements were made of glucose uptake, lactate output, C14 glucose incorporation into glycogen and tissular levels of ATP, PCr, glycogen and lactate. Glycolysis was stimulated by hypoxia to a greater extent in the young foetal heart of 16.5 days post coitum than in the foetal heart at term. Thus high energy phosphates were maintained at a higher level in the younger heart. The results are discussed in relation to the high resistance to hypoxia of the foetal heart.  相似文献

The type 1 insulin-like growth factor receptor (IGF-1R), a transmembrane tyrosine kinase, is widely expressed across many cell types in foetal and postnatal tissues. Activation of the receptor following binding of the secreted growth factor ligands IGF-1 and IGF-2 elicits a repertoire of cellular responses including proliferation, and the protection of cells from programmed cell death or apoptosis. As a result, signalling through the IGF-1R is the principal pathway responsible for somatic growth in foetal mammals, whereas somatic growth in postnatal animals is achieved through the synergistic interaction of growth hormone and the IGFs. Forced overexpression of the IGF-1R results in the malignant transformation of cultured cells: conversely, downregulation of IGF-1R levels can reverse the transformed phenotype of tumour cells, and may render them sensitive to apoptosis in vivo. Elevated levels of IGF-IR are observed in a variety of human tumour types, whereas epidemiological studies implicate the IGF-1 axis as a predisposing factor in the pathogenesis of human breast and prostate cancer. The IGF-1R has thus emerged as a therapeutic target for the development of antitumour agents. Recent progress towards the elucidation of the three-dimensional structure of the extracellular domain of the IGF-1R represents an opportunity for the rational assembly of small molecule antagonists of receptor function for clinical use.  相似文献

Summary The development of NA-uptake mechanisms in the human foetal heart start at the same time as the adrenergic terminals were visible. Ethanol may modulate endogenous opioid systems by disrupting opioid receptor signalling. Low concentrations of ethanol slightly potentiate mu-opioid receptor binding by increasing receptor Bmax, and, in some cases, chronic ethanol exposure decreases the density or affinity of the mu-opioid receptors. By contrast, high concentrations of ethanol acutely decrease delta-opioid receptor binding by decreasing receptor affinity, whereas chronic exposure of animals and neuronal cell lines to lower concentrations of ethanol leads to possibly adaptive increases in the density or affinity of the delta-opioid receptors. In the neuronal cell line NG108-15, ethanol does not up-regulate the delta-opioid receptor by blocking receptor degradation or endocytosis, but protein synthesis is required for this response. Up-regulation of the delta-opioid receptor renders ethanol-treated NG108-15 cells 3.5-fold more sensitive to opioid inhibition of adenylyl cyclase. Long-term treatment with ethanol also increases maximal opioid inhibition in NG108-15 cells, possibly by decreasing levels of G alpha s and its mRNA. Ethanol differentially modulates signal transduction proteins in three additional neuronal cell lines, N18TG2, N4TG1, and N1E-115. Ethanol-treated N18TG2 cells show the least up-regulation of the delta-opioid receptor, little heterologous desensitization of adenylyl cyclase, and no changes in G alpha s or G alpha i. By contrast, ethanol-treated N1E-115 cells show the largest up-regulation of the delta-opioid receptor, the most heterologous desensitization of adenylyl cyclase, and concentration-dependent decreases in G alpha s and increases in G alpha i. Further analysis of these related neuronal cell lines may help to identify the molecular elements that endow some, but not all, neuronal cells with the capacity to adapt to ethanol.  相似文献

The antibodies against estrogen receptor were obtained after injecting Rabbits with a cytoplasmic receptor fraction isolated from Calf uterus. The estrogen receptor was partially proteolysed by the action of trypsin and subsequently purified by affinity chromatography (purification 4,000 to 10,000 fold, to a purity of 5-20%). The affinity of the antibody for the proteolysed receptor is KD approximately 1 nM and serum titres have reached values of approximately 50 nM. The values remained constant after the third injection. Preliminary results indicate that the antibody has approximately the same affinity for "native" cytoplasmic estrogen receptor from Calf uterus, as well as for the "trypsinized" forms of estrogen receptor isolated from Calf uterine cytosol and Hen oviduct nuclei.  相似文献

The binding of asialoglycoproteins by hepatic binding protein was studied in freshly isolated hepatocytes from genetically diabetic BB Wistar rats. The number of cell surface asialoglycoprotein receptors was dramatically decreased (58,000 +/- 38,000 for diabetic rats compared to 267,000 +/- 70,000 for normal rats), while the association equilibrium constant was not changed. These results parallel those obtained with streptozotocin-diabetic rats and support the hypothesis that insulin deprivation is responsible for the decrease in the receptor number.  相似文献

The effect of dynorphin A-(1-13) (Dyn A-(1-13] and other opioids on the cytosolic free calcium concentration [(Ca2+]i) in rat anterior pituitary cells was examined using the fluorescent indicator fura-2. A commercial synthetic Dyn A-(1-13) preparation elevated [Ca2+]i. Results, which were obtained with receptor antagonists, and in LHRH receptor radioligand binding studies as well as by HPLC combined with LHRH radioimmunoassay, strongly suggest that this effect of the dynorphin preparation was due to contamination with a LHRH-like compound. Dyn A-(1-13), purified by HPLC, as well as Dyn A-(2-13), [Leu5]enkephalin, beta-endorphin, morphine, or U50,488H had no effect on [Ca2+]i. LHRH caused a rapid increase in [Ca2+]i by about 50 nM which was blocked by the LHRH antagonist, [D-pGlu1,D-Phe2,D-Trp3,6] LHRH.  相似文献

In vitro ecdysteroid production by dissociated Y-organ cells of the shore crabCarcinus maenas was characterized during short-term incubations. Under optimized conditions (M199 adjusted to crab osmolality and with the addition of 10% foetal calf serum), ecdysteroid production by dispersed cells increased linearly during 4-hour incubations, with little intra-assay variation. 25-deoxyecdysone was mainly produced. PurifiedCarcinus molt inhbiting hormone (CamMIH) produced a dose-dependent inhibition of ecdysteroid production by dispersed cells. The cells were about 50 times more sensitive than whole glands to MIH. Other structurally-related peptides were tested.  相似文献
