为检测黄瓜花叶病毒的存在,应用Northernblot,将提取的黄瓜花叶病毒RNA样品,经甲醛变性凝胶电泳分离,使RNA解离成单链,然后用上行毛细管转移法,将凝胶上的单链RNA片段转移到尼龙膜上.经适当洗涤、干燥后,在烤箱中80℃保温2h,用于杂交.以带有黄瓜花叶病毒目的片段的细菌质粒制备DNA探针,用放射性同位素32P标记探针DNA,100℃变性处理用于杂交的探针,然后进行杂交.结果表明,在杂交膜上显示与特异性探针结合的阳性RNA条带,检测到了黄瓜花叶病毒的存在.  相似文献

从内蒙古巴盟地区分离的蓝舌病病毒株(BTV-NM)提取总RNA,经反转录PCR扩增VP2基因5′端片段,并构建至pGEM-T载体中.序列测定后,将这一序列与蓝舌病毒澳大利亚株VP2基因5′端进行比较分析:同源性为40%.通过PCR法标记克隆的cDNA片段,制备地高辛标记探针,与粗提的蓝舌病发病羊病毒RNA进行Northern blot杂交,并作敏感性试验,结果表明此探针对蓝舌病毒内蒙古分离株具有特异性,可检测出50 pg的病毒RNA.  相似文献

本文利用RT-PCR和PCR技术制备地高辛标记的谷胱甘肽过氧化物酶(ATGPX3)cDNA探针,分别进行DNA和RNA斑点和印迹杂交分析.通过调整杂交液组分的浓度和增加10%硫酸葡聚糖的方法,改进杂交反应.实验结果表明,改良的方法不仅提高了杂交效率,而且明显检测到RNA杂交印迹反应.另外,利用地高辛标记cDNA探针技术也检测到了植物激素ABA诱导的ATGPX3基因的表达,同时证明了该方法可以用来检测植物基因的表达.  相似文献

在水溶液中合成了表面具有自由羧基的硫化铅(PbS)纳米颗粒,以乙基-(3-二甲基丙基)碳二亚胺盐酸盐(EDAC)为偶联活化剂,将其标记于人工合成的5’端氨基修饰的寡聚核苷酸(ODN)片段上,制备成PbS纳米颗粒标记DNA探针.在一定的条件下,使其与固定在玻碳电极表面的待测DNA序列进行杂交反应,利用阳极溶出示差脉冲伏安法间接测定Pb(Ⅱ)的量,从而实现对互补、非互补DNA片段的识别和电化学检测.同时对该探针的稳定性、选择性进行了讨论。  相似文献

用PCR法成功制备了DIG标记探针,探针长度为547bp,探针的产量为21.6mg/μL。此探针与随机引物合成探针检测样品灵敏度相近。用此探针核酸斑点杂交法检测了54尾中国对虾。结果表明:此探针在对白斑综合症病毒的检测、对虾暴发性流行病的诊断等方面具有很高的应用价值。  相似文献

从前列腺癌细胞系LNCaP中提取总RNA,经RT-PCR扩增前列腺特异性膜抗原(PSM)靶序列,将其定向插入带T7RNA聚合酶启动子的pSPORT1质粒,转入大肠杆菌DH5α,经蓝白斑筛选获得重组质粒,以此重组质粒线性化DNA为模板,以地高辛标记的rUTP及其他三种rNTP为底物,经T7RNA聚合酶体外转录,成功地制备了地高辛标记的PSM特异性RNA探针,为研究以PSM为分子生物学指标的前列腺癌分期诊断奠定了基础。  相似文献

结合恒温滚环扩增(rolling circle amlification,RCA)、单链特异性核酸外切酶Ⅰ(exonucleaseⅠ,ExoⅠ)和阳离子共轭聚合物(Cationic conjugated polymer,CCP)荧光共振能量转移(fluorescence reso-nance energy transfer,FRET)技术,建立了一种特异、灵敏的均相检测microRNA(miRNA)的新方法.该方法应用荧光标记探针与RCA扩增的长链DNA产物杂交,当加入CCP时,其与杂交的标记探针通过静电力结合,发生高效的FRET.未杂交的标记探针利用ExoⅠ水解成单核苷酸,其与CCP相互作用力弱,不能发生有效的FRET.基于此,无需分离和洗涤步骤,实现了RCA扩增miRNA的均相检测.方法特异性好,灵敏度高,线性为0.5~20pmol/L,检出限为0.2pmol/L.方法为miRNA均相检测和原位成像分析以及临床诊断提供了新策略.  相似文献

以1-乙基-3-(3-二甲基氮丙基)-碳化二亚胺(EDC)为偶联剂,将氨基标记的寡核苷酸探针(包含捕获探针、检测偶联效率的Tag序列以及间隔臂三部分)固定在羧基末端荧光微球表面.以生物素标记的cTag与微球表面探针杂交,再加入荧光物质PE标记的亲和素(SA-PE),通过Luminex-100TM荧光微球检测仪对荧光微球表面探针的偶联效果进行了检测.结果表明:不同间隔臂的氨基标记寡核苷酸探针具有不同的偶联效率,C12偶联效率优于C6-5T,C6-5T优于C6;且平均荧光强度(MFI)值最高可达3000以上,为后续核酸杂交检测建立了基础.  相似文献

Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
