Forecasts have little value to decision makers unless it is known how much confidence to place in them. Those expressions of confidence have, in turn, little value unless forecasters are able to assess the limits of their own knowledge accurately Previous research has shown very robust patterns in the judgements of individuals who have not received special training in confidence assessment: Knowledge generally increases as confidence increases. However, it increases too swiftly, with a doubling of confidence being associated with perhaps a 50 per cent increase in knowledge. With all but the easiest of tasks, people tend to be overconfident regarding how much they know These results have typically been derived from studies of judgements of general knowledge. The present study found that they also pertained to confidence in forecasts. Indeed, the confidence-knowledge curves observed here were strikingly similar to those observed previously. The only deviation was the discovery that a substantial minority of judges never expressed complete confidence in any of their forecasts. These individuals also proved to be better assessors of the extent of their own knowledge Apparently confidence in forecasts is determined by processes similar to those that determine confidence in general knowledge. Decision makers can use forecasters assessments in a relative sense, in order to predict when they are more and less likely to be correct. However, they should be hesitant to take confidence assessments literally. Someone is more likely to be right when he or she is ‘certain’than when he or she is ‘fairly confident’; but there is no guarantee that the certain forecast will come true.  相似文献

Summary Halothane anesthesia was found to be hepatotoxic in the rat, as demonstrated by a significant elevation of serum xanthine oxidase (SXO) level. For a long time, the accepted explanatory model of language acquisition was the so-called Principles and Parameters framework (P&P). P&P seemingly provides an elegant answer to the central puzzle of generative linguistics: How do children acquire their native language given the limited time and input resources available to them? Yet P&P tells a story that is evolutionarily implausible, and for this reason it has since been abandoned. I argue that this is an unwarranted move, and that it could and should be avoided by reassessing the epistemic status of P&P. In particular, I argue that contrary to extant accounts, P&P ought to be retrospectively construed as a highly idealized (toy) model of language acquisition. The proposed reinterpretation is vindicated, I argue, insofar as it paves the way for a reconciliation of the two central explanatory challenges of modern generative linguistics.  相似文献

Summary Morphogenetic fields must be generated by mechanisms based on known physical forces which include gravitational forces, mechanical forces, electrical forces, or some combination of these. While it is unrealistic to expect a single force, such as a voltage gradient, to be the sole cause of a morphogenetic event, spatial and temporal information about the electrical fields and ion concentration gradients in and around a cell or embryo undergoing morphogenesis can take us one step further toward understanding the entire morphogenetic mechanism. This is especially true because one of the handful of identified morphogens is Ca2+, an ion that will not only generate a current as it moves, but which is known to directly influence the plasma membrane's permeability to other ions, leading to other transcellular currents. It would be expected that movements of this morphogen across the plasma membrane might generate ionic currents and gradients of both electrical potential and intracellular concentration. Such ionic currents have been found to be integral components of the morphogenetic mechanism in some cases and only secondary components in other cases. My goal in this review is to discuss examples of both of these levels of involvement that have resulted from investigations conducted during the past several years, and to point to areas that are ripe for future investigation. This will include the history and theory of ionic current measurements, and a discussion of examples in both plant and animal systems in which ionic currents and intracellular concentration gradients are integral components of morphogenesis as well as cases in which they play only a secondary role. By far the strongest cases for a direct role of ionic currents in morphogenesis is the polarizing fucoid egg where the current is carried in part by Ca2+ and generates an intracellular concentration gradient of this ion that orients the outgrowth, and the insect follicle in which an intracellular voltage gradient is responsible for the polarized transport from nurse cell to oocyte. However, in most of the systems studied, the experiments to determine if the observed ionic currents are directly involved in the morphogenetic mechanism are yet to be done. Our experience with the fucoid egg and the fungal hypha ofAchlya suggest that it is the change in the intracellular ion concentration resulting from the ionic current that is critical for morphogenesis.  相似文献

Surprisingly little attention has been given hitherto to the definition of the laboratory. A space has to be specially adapted to deserve that title. It would be easy to assume that the two leading experimental sciences, physics and chemistry, have historically depended in a similar way on access to a laboratory. But while chemistry, through its alchemical ancestry with batteries of stills, had many fully fledged laboratories by the seventeenth century, physics was discovering the value of mathematics. Even experimental physics was content to make use of almost any indoor space, if not outdoors, ignoring the possible value of a laboratory. Summary remarks It is evident that the bulbar EEG is a complex response which can be influenced by several methodological and stimulant variables. What EEG patterns tell us about actual behavior remains obscure. The bulbar EEG which is evoked by homestream water is not necessarily a reflection of olfactory memory. The evoked bulbar EEG also does not necessarily demonstrate the salmon has distinguished the home-water from another water in terms of migrational orientation. Despite lack of absolute specificity, a correlation between bulbar EEG and actual behavioral performance has been observed even though some non-home waters evoke responses similar to that of the home water. In general it has been found that the home water response can be distinguished from the response to other natural waters. Failure to obtain complete specificity may be due to a variety of variables which have been alluded to earlier in this section. Thus considering that the evoked EEG is a reflection of the integration of a diverse afferent input, further electrical and computer analyses may eventually permit the decoding of the EEG in terms of behavior. However, if the EEG proves to be more a quantitative rather than qualitative reflection of brain activity, relating EEG to behavior will prove to be of limited value. Since afferent responses in the olfactory bulb are influenced by impulses from higher brain centers, studies on centrifugal aspects during olfactory stimulation may be useful in gaining some qualitative understanding of the home water evoked EEG of the olfactory bulb.  相似文献

In this paper we examine how causality inference and forecasting within a bivariate VAR, consisting of y(t) and x(t), are affected by the omission of a third variable, w(t), which causes (a) none, (b) one, and (c) both variables in the bivariate system. We also derive conditions under which causality inference and forecasting are invariant to the selection of a bivariate or a trivariate model. The most general condition for the invariance of both causality and forecasting to model selection is shown to require the omitted variable not to cause any of the variables in the bivariate system, although it allows the omitted variable to be caused by the other two. We also show that the conditions for one-way causality inference to be invariant to model selection are not sufficient to ensure that forecasting will also be invariant to the model selected. Finally, we present a numerical illustration of the potential losses, in terms of the variance of the forecast, as a function of the forecast horizon and for alternative parameter values—they can be rather large, as the omission of a variable can make the incomplete model unstable. © 1997 John Wiley & Sons, Ltd.  相似文献

Early investigations into the nature of the coupling between energy transduction and metabolism in smooth muscle, particularly from the laboratories of Bülbring and Lundholm, suggested that specific metabolic pathways could independently supply energy for ion transport and actin-myosin interactions. Subsequent work has solidified the concept that oxidative phosphorylation is specifically coupled to tension generation and maintenance, whereas, aerobic glycolysis is not only a vital characteristic of smooth muscle metabolism, but also is likely to be independently coupled to Na-K transport at the plasmalemma. The independence of oxidative and glycolytic metabolism is reflected as a compartmentation of carbohydrate metabolism in the porcine carotid artery. The coupling of these independent metabolic pathways with specific energy utilizing processes, indicates a means by which energy production and transduction can be closely and efficiently regulated. The coupling of glycogenolysis to mitochondrial respiration may have evolved as a direct response to the energetic needs of VSM. That is, the large glycogenolytic response in the initial minutes of stimulation may be necessary to maximize the cellular production of ATP during the presteady state. Likewise, the coupling between aerobic glycolysis and Na-K transport indicates a sensitive and efficient means of coordinating energy metabolism with ion transport at the membrane level. Additionally, the regulation of substrate supply, i.e. glucose transport, also may be closely coordinated with changes in ion transport. One may speculate that alterations in the microenvironment of each compartment can independently regulate intermediary metabolism and therefore allow the cell to quickly and efficiently respond to localized stimuli. Thus, stimulation of Na-K transport could effectively regulate energy production at the membrane level without mobilizing or competing with the energy transduction of other cellular processes. This compartmentation of energy utilization may be highly advantageous, since oxidative metabolism is closely coordinated with mechanical activity and therefore regulation of blood flow. Future investigations will attempt to elucidate which intracellular signals which are responsible for the regulation of these functionally independent compartments of energy metabolism and transduction in VSM. In more general terms, our findings provide a basis from which future questions concerning the regulation of cellular metabolism must be directed. The cellular cytoplasm can no longer be envisioned as a homogeneous compartment, but rather a complex array of functional subcompartments which may be individual  相似文献

Mucosa-associated lymphoid tissue in the respiratory and digestive tracts are covered by a specialized epithelium, the follicle-associated epithelium, which includes M cells, which are specialized for the uptake and transcytosis of macromolecules and microorganisms. Following transcytosis, antigens are released to cells of the immune system in lymphoid aggregates beneath the epithelium where antigen processing and presentation and stimulation of specific B and T lymphocytes are achieved. Circulation of the lymphoid cells enables their homing to their original, and other, mucosal sites where they exert the effector function. Such a response may be dominated by secretory immunoglobulin A release and may include cytotoxic T lymphocyte action. Binding of particles to the apical M cell membrane may be nonspecific or due to specific interaction between molecules such as integrins and lectins. Exploiting the specific binding to M cells is an aim for mucosal vaccination, for example to increase the efficiency of uptake of an oral vaccine by its conjugation to an M-cell-specific molecule. Alternatively, an M-cell-specific live vector, such as attenuated Salmonella bacteria, may be used to deliver epitopes of other organisms. Mucosal vaccination efficiency may also be enhanced by a temporary increase in the number of M cells. Therefore, investigation of the properties and ontogeny of M cells must be pursued to allow the development of better mucosal vaccines for the future.  相似文献

Malaria presents a challenge to world health that to date has been beyond the abilities of researchers to conquer. This critique presents some of the strategies employed by the parasite to overcome immunity and the immunological challenges that we face to develop vaccines. A conclusion is that a vaccine must identify novel antigens or epitopes that are not normally immunogenic and which are therefore not under immune pressure and most likely to be conserved between different strains. Such antigens are most likely to be targets of cellular immunity. The case for a whole parasite blood stage vaccine is presented based on these premises.  相似文献
