It has often been claimed that Priestley was a skilful experimenter who lacked the capacities to analyze his own experiments and bring them to a theoretical closure. In attempts to revise this view some scholars have alluded to Priestley’s ‘synoptic’ powers while others stressed the contextual role of British Enlightenment in understanding his chemical research. A careful analysis of his pneumatic reports, privileging the dynamics of his experimental practice, uncovers significant yet neglected aspects of Priestley’s science. By focusing on his early experimental conduct and writing on nitrous air, I demonstrate how his methodological and rhetorical devices, far from being consequences of compulsive writing or theoretical naïveté, were deeply entwined with his chemical research. I employ the notion of ‘style of experimental reasoning’ (SER)—derived from A. C. Crombie and I. Hacking—to shed light on the intersection at which Priestley’s unique method, literary style, and epistemology converged to generate scientific knowledge. Establishing Priestley’s SER advances a finer understanding of the interactive character of his pneumatic experimentalism, peculiar dimensions of which have evaded both traditional as well as revisionist scholarship, thus infusing the longstanding historiographic debate over his scientific merits.  相似文献

Summary The metastasis of malignant tumors from a primary site to near and distant secondary sites is probably the most important event in the pathogenesis of cancer and it accounts for most cancer deaths1. Whereas advances in the treatment of primary cancer have led to increased patient survival, metastatic cancers are still the most difficult group of diseases to treat successfully2. As organ-characteristic lectins play an important role in the organ manifestation of metastatic islets3,4, it might be possible (e.g. during surgical operations on malignant tumors) to block those organ-characteristic lectins with the appropriate receptor-bearing glycoconjugates in order to inhibit the metastatic spread. Recent experiments have demonstrated that neuraminidase treatment of tumor cells (mouse sarcoma-1) alters in vivo (Balb/c-mice) the organotropic distribution of metastases; instead of being found exclusively in the lung, they are found both in lung and liver. However, pre-injection and regular application of D-galactose — the same holds for arabinogalactan5,6,13 — prevents the settling of metastases in the liver but does not influence the metastatic process to the lung, whereas mannan — as a galactose-free control substance — does not alter the initial pattern of metastasis to lung and liver.  相似文献

Summary Efficient excited state formation — much higher than that hitherto expected — may occur in organelles and in intact cells. Excited triplet states can be enzymatically generated in high yields by different routes. An example is the oxidation of isobutanal to acetone and formic acid, catalyzed by horseradish peroxidase. Other enzymatic systems that generate triplet carbonyls are linear aliphatic aldehydes when oxidized by peroxidase/O2, or the indole-3-acetic acid/peroxidase/O2-reaction. The latter is widespread in plants.This new field — photobiochemistry without light — has led to a growing awareness of the idea that cells may utilize excited states to trigger photochemical processes even in the dark. Such phenomena are of considerable importance, also for the understanding of weak photon emission from biological systems.  相似文献

Thirty years after the publication of Thomas Kuhn's The Structure of Scientific Revolutions, sharp disagreement persists concerning the implications of Kuhn's ‘historicist’ challenge to empiricism. I discuss the historicist movement over the past thirty years, and the extent to which the discourse between two branches of the historical school has been influenced by tacit assumptions shared with Rudolf Carnap's empiricism. I begin with an examination of Carnap's logicism — his logic of science — and his 1960 correspondence with Kuhn. I focus on problems in the analysis applied to the unit of meta-scientific study or appraisal, arguing for a re-assessment of historicist treatment of the internal/external distinction and historiographic meta-methodology. The critique of objectivism and relativism that eventuates from this re-assessment is a double-edged blade, undercutting both objectivist and relativist treatments of cognitive evaluation and scientific change. I use it to cut across an otherwise diverse group of historicist-influenced writers, including Imre Lakatos, Larry Laudan, H.M. Collins and Stephen Stich.  相似文献

Melatonin from the retina and the pineal gland functions in neuroendocrine hierarchies. Photoreceptors — eyes and extraretinal — detect light. Oscillators — pineal and suprachiasmatic nuclei — act as pacemakers. Driven neuroendocrine rhythms carry temporal hormone signals throughout the body. Light controls melatonin: light sets the phase of the melatonin rhythm and determines the duration of melatonin synthesis. Efferent motor signals to skeletal muscles concern not only the space/time pattern of motion, but also the setting of muscular performance and through this the control of the current metabolic rate. For an optimal adjustment of metabolic rate during heavy exercise — e.g. in athletic competitions — a feedback control system must exist, including a programmer that takes into consideration a finishing point (teleoanticipation). The presented experiments, using Borg's scale, indicate the existence and functioning of a system for optimal adjustment of performance during heavy exercise and the relevance of teleoanticipatory effects. Thus motor learning includes not only somatosensory control, but also metabolic control. With regard to migratory birds, such metabolic control would have to operate in the individual as well as in the migrating flock as a whole.  相似文献

This paper investigates Hermann Weyl’s reception of philosophical concepts stemming from the German Idealist Johann Gottlieb Fichte. In particular, Weyl’s ‘agens theory’ of matter, which he held around 1925, will be looked at. In the extant literature, the—admittedly also important—influence of Husserl on Weyl has mainly been addressed. Thus, apart from investigating some detailed Fichtean inheritances in Weyl’s concepts of causality, chance and continuity, the general difference which Weyl saw between the philosophies of Fichte and Husserl will also be discussed. For Weyl this is above all a difference between an active constructivism and a rather passive phenomenological seeing (Schau). Further, the paper shows in some detail the way Weyl was drawn into a certain reading of Fichte by his Zurich colleague, the philosopher Fritz Medicus. The methodological frame of the paper is that of Konstellationsforschung, a historical and systematic approach which proves to be particularly fruitful when investigating a (broadly speaking) German Idealist context and which allows special attention to be given to the acting subjects within the constellation under investigation. Conversely, Weyl’s agens theory suggests amendments to this methodology.  相似文献

In this paper, I consider Kitcher’s (1993) account of reference for the expressions of past science. Kitcher’s case study is of Joseph Priestley and his expression ‘dephlogisticated air’. There is a strong intuitive case that ‘dephlogisticated air’ referred to oxygen, but it was underpinned by very mistaken phlogiston theory, so concluding either that dephlogisticated air referred straightforwardly or that it failed to refer both have unpalatable consequences. Kitcher argues that the reference of such terms is best considered relative to each token—some tokens refer, and others do not. His account thus relies crucially on how this distinction between tokens can be made good—a puzzle I call the discrimination problem. I argue that the discrimination problem cannot be solved. On any reading of Kitcher’s defence of the distinction, the grounds provided are either insufficient or illegitimate. On the first reading, Kitcher violates the principle of humanity by making Priestley’s referential success a matter of the mental contents of modern speakers. The second reading sidesteps the problem of beliefs by appealing to mind-independent facts, but I argue that these are insufficient to achieve reference because of the indeterminacy introduced by the qua problem. On the third and final reading, Priestley’s success is given by what he would say in counterfactual circumstances. I argue that even if there are facts about what Priestley would say, and there is reason for doubt, there is no motivation to think that such facts determine how Priestley referred in the actual world.  相似文献

This paper discusses the historiography of the ‘two cultures’ controversy. C. P. Snow’s lament about the ‘two cultures’, literary and scientific, has inspired a wide range of comment—much of which begins by citing Snow and his thesis, before going on to discuss very different things. This paper focuses upon one strand of this commentary, the historical analysis of the controversy itself. A ‘historical’ analysis is defined here as one that resists the impulse to enter the argument on behalf of Snow or Leavis, to conceive of their argument in the terms that Snow defined, or to invoke their argument as a precursor to some contemporary issue. Instead, a historical interpretation registers distance between that day and this, takes the controversy itself as its object of study, and explores the tensions and associations that came to be packed into those now familiar terms. As the fiftieth anniversary of Snow’s Rede Lecture nears, this approach—rather than the repetition of clichés about the bridging of cultures—offers both analytical perspective on the controversy and interpretive possibilities for its examination.  相似文献

Conclusion If the hundred years of study on theHensen's node — i.e. on gastrulation and early determination of the embryos of amniote vertebrates — teach anything, they teach in the first place how limited and fragmentary our knowledge is about one of the most central problems of the whole developmental biology. We know that the events in early amniote development — or early avian development, on which our data and ideas are nearly all based — in many ways resemble those in early Amphibian development, which is only slightly better understood, but we also know that direct extrapolations from anamniotes to amniotes cannot be made without proper reservations and without studying the amniote embryos themselves. And we have practically no idea of what is really going on in the cells of the blastoderm when they move, invaginate, induce or are induced, interact, become determined and begin their differentiation. We know that at the stages of gastrulation, the node, and indeed the whole blastoderm, is in a very labile state and can be regulated in many ways to produce a harmonious whole — or a monster — although we only understand very poorly the modes of this regulation. The progress made during the decades, and particularly in recent years, shows, however, that useful information is accumulating to produce a coherent picture, and there is no reason to be pessimistic.Dedicated to ProfessorEtienne Wolff on the occasion of his retirement.  相似文献
