采用HPLC法同时测定罗布麻浸膏粉中金丝桃苷、异槲皮苷、槲皮素的含量.色谱柱为YMC-Pack C18(250×4.6 mm),流动相为乙腈-甲醇(10∶1)和0.4%磷酸,梯度洗脱;二极管阵列检测器:检测波长360 nm;分别以金丝桃苷、异槲皮苷、槲皮素对照品为外标,用标准曲线法进行定量.在0.25~10μg/mL的范围内,金丝桃苷、异槲皮苷、槲皮素的浓度与吸收峰面积之间线性关系良好,相关系数分别为r金丝桃苷=0.999 4、r异槲皮苷=0.999 5、r槲皮素=0.999 6,回收率在85.57%~99.96%之间(RSD小于4.45%).  相似文献

采用超高效液相色谱法对五子衍宗丸中金丝桃苷的含量进行了测定,并比较了70℃蒸馏水、常温蒸馏水、体积分数70%乙醇、无水乙醇和DMSO等5种溶剂对金丝桃苷的提取效果;在此基础上,优选出细胞干预液制备方法,观察五子衍宗丸对NTDs细胞凋亡的防控作用。结果显示,金丝桃苷在0.004~0.1μg范围内具有良好的线性关系(r=0.999 9);平均回收率为98.85%,RSD为3.86%;5种提取溶剂中体积分数为70%乙醇对金丝桃苷的提取效果最好;以五子衍宗丸70%乙醇提取物干预NTDs细胞,能够明显提高细胞存活率,有效抑制细胞凋亡。  相似文献

建立了高效液相色谱-串联质谱(HPLC-ESI/MS)法对芦丁、金丝桃苷、槲皮素和槲皮苷的抗氧化活性进行定量研究。样品用甲醇溶解,采用Hewlett Packard ODS Hypersil C_(18)色谱柱(100 mm×4.6 mm,5μm)、0.1%甲酸水-甲醇为流动相梯度洗脱,流速0.6 m L·min~(-1),柱温25℃;电喷雾源,负离子模式多反应选择离子检测(MRM),用于定性、定量的母离子和子离子分别为m/z 609→300(芦丁)、m/z 463→300(金丝桃苷)、m/z 301→151(槲皮素)、m/z 447→300(槲皮苷),采用外标法进行定量分析。结果表明:芦丁、金丝桃苷、槲皮素和槲皮苷在0.5~1 600 ng·mL~(-1)范围内线性关系良好(R0.998),定量限为0.5 ng·m L~(-1),仪器精密度良好(RSD3.0%),样品反应液在24 h内稳定性良好。本方法具有灵敏度高,分析时间短等特点,相对于其他检测方法,提高了检测的准确性,适用于对贯叶连翘中黄酮醇类化合物抗氧化活性的定量研究。经计算,芦丁、金丝桃苷、槲皮素和槲皮苷对ABTS~(.+)的清除率分别为18.35%、36.28%、41.50%和37.18%。  相似文献

建立HPLC测定短梗五加果实中异嗪皮啶和金丝桃苷含量的方法.用Waiers e2695-C18色谱柱(4.6nm×150nm,5μm)以乙腈-水溶液比例为(20∶80)为流动相,流速为1.0 mL/min,柱温:30℃,检测波长355nm.异嗪皮啶进样量在10μL内具有良好的线性关系,y=151 334 510 x+5 939 724,R2=0.939 3.异嗪皮啶的平均回收率为101.02%,RSD为2.09%.金丝桃苷进样量在10μL内呈良好的线性关系,y=27 639 620 x+76 443,R2=0.999 9.平均回收率为99.7%,RSD为2.02%.HPLC灵敏快速,结果准确,重现性好,可以用于短梗五加果实中金丝桃苷和异嗪皮啶的含量测定.  相似文献

目的:通过测定大理苍山两种不同鹿蹄草中金丝桃苷的含量,为控制鹿蹄草的质量和临床应用提供理论依据。方法:HPLC测定金丝桃苷含量的色谱条件:ZORBAX SB-C18色谱柱(4.6mm×150mm,5μm),流动相:甲醇∶水(35∶65),检测波长:358nm,进样量:8.00μl,流速:1.0ml/min,柱温:25℃。结果:金丝桃苷的线性范围为0.18~0.90μg(r=0.9991),回收率为100.60%(RSD=1.78%)。结论:两种鹿蹄草中金丝桃苷的含量存在差异。  相似文献

建立了HPLC法同时测定小花鬼针草中芦丁、金丝桃苷和槲皮苷含量的方法。采用Diamonsil C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.2%冰醋酸梯度洗脱,流速为1 mL/min;检测波长为360 nm。结果表明,芦丁、金丝桃苷和槲皮苷在0.50~12.50μg/mL(r=0.999 9)、0.49~12.35μg/mL(r=0.999 8)、0.42~10.55μg/mL(r=0.999 5)范围内线性良好,平均回收率为99.37%(RSD=0.56%)、99.35%(RSD=0.57%)、99.68%(RSD=0.56%)。该方法简便、准确、重现性好,可用于同时测定鬼针草中3种黄酮成分的含量。  相似文献

建立刺玫果提取物胶囊的定性鉴别和含量测定方法.采用薄层色谱法对刺玫果提取物胶囊进行了定性分析;采用紫外-可见分光光度法,建立了刺玫果提取物胶囊总黄酮的含量测定方法;采用高效液相色谱法,建立了刺玫果提取物胶囊中金丝桃苷的含量测定方法.结果表明建立的薄层色谱鉴别方法,荧光斑点清晰,阴性无干扰;芦丁在4.0~48.0μg/mL(r=0.9996),金丝桃苷在0.192~1.152μg(r=0.9999)线性关系良好;芦丁平均回收率100.05%,RSD为1.40%(n=6);金丝桃苷平均回收率99.39%,RSD为0.66%(n=6).建立的薄层鉴别方法专属性高,含量测定方法具有良好的精密度,结果准确可靠,可用于刺玫果提取物胶囊的质量控制.  相似文献

采用高速逆流色谱结合半制备液相色谱技术快速分离山楂叶中低含量的牡荆素、金丝桃苷、异槲皮苷单体。以氯仿-甲醇-水-正丁醇(4∶3∶2∶1.5,V/V)为溶剂系统,流速为5.0 m L/min,转速为850 r/min,检测波长为254 nm,从山楂叶黄酮提取物中富集得到含牡荆素、金丝桃苷、异槲皮苷3种化合物的混合组分,经半制备液相进行二次分离纯化,得到纯度大于98%的牡荆素、金丝桃苷、异槲皮苷单体。研究结果表明,该技术对山楂叶中低含量黄酮类化合物的分离快速、高效,且纯度高。  相似文献

目的采用HPLC建立金银花中芦丁、金丝桃苷、木犀草苷、木犀草素4种黄酮成分含量测定的方法,分析不同产地金银花中这4种成分的含量差异,为评价金银花药材质量提供参考。方法色谱柱为Agilent Eclipse XDB-C18(250 mm×4.6 mm, 5μm),流动相为乙腈-0.5%冰醋酸,梯度洗脱,流速1 mL·min~(-1),检测波长350 nm,柱温30℃。结果 4种成分均达到基线分离,线性关系良好(r≥0.9996),平均加样回收率为99.51%~100.35%,RSD2%,建立的方法简便快速,准确度高,重现性好。结论不同产地金银花中芦丁和木犀草苷的含量差异较大,而金丝桃苷和木犀草素的含量均较低,差异较小。本实验为金银花药材的质量评价提供了一定的科学依据。  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
