Cloning and sequence analysis of cDNA for bovine adrenal preproenkephalin   总被引:60,自引:0,他引:60  
The nucleotide sequence of cloned cDNA for preproenkephalin from bovine adrenal medulla indicates that the precursor protein contains four copies of Met-enkephalin and one copy each of Leu-enkephalin, Met-enkephalin-Arg6-Phe7 and Met-enkephalin-Arg6-Gly7-Leu8, a previously undetected opioid peptide. The enkephalin and extended enkephalin sequences are each bounded by paired basic amino acid residues. Preproenkephalin may represent a multi-hormone precursor, like the corticotropin-beta-lipotropin precursor.  相似文献

以无花瓣与正常甘蓝型油菜幼嫩花蕾(长度小于0.2 mm)建立的差异表达文库中发现的异胡豆苷合成酶基因(STR)的一个cDNA片段为基础,利用RACE方法获得了甘蓝型油菜的STR基因cDNA全序列BnSTR.序列分析表明BnSTR全长1408 bp,开放阅读框从第20个碱基到第1291个碱基,推导的蛋白序列与Arabidopsis thaliana, Lycopersicon esculentum, Oryza sativa, Rauvolfia manni, Catharanthus roseus和Ophiorrhiza pumila中STR的同源性分别是91, 41.8, 36.2, 31.2, 29.2和28.4%.表达结果显示,与野生型相比,油菜BnSTR在无花瓣突变型幼嫩花蕾中表达量明显下降, BnSTR可能与花瓣的发育有关.  相似文献

根据GenBank中绵羊催乳素受体(PRLR)基因序列(AF041257.1)设计1对引物,以乐至黑山羊脑垂体总RNA为模板,通过RT-PCR技术对乐至黑山羊PRLR基因cDNA克隆测序和序列分析.结果表明:扩增出的乐至黑山羊PRLR基因cDNA序列长为1743bp,编码581个氨基酸.乐至黑山羊与绵羊、牦牛、欧洲牛、野猪和人的核苷酸同源性分别为:98.34%、94.67%、94.27%、77.48%、74.33%.以核苷酸序列构建分子进化树,乐至黑山羊先与绵羊聚为一类,再与欧洲牛和牦牛聚为一类,而后与野猪聚为一类,最后与人聚为一类.  相似文献

Atrial extracts contain factors which induce potent natriuresis changes in renal haemodynamics, and relax pre-contracted vascular smooth muscle. Low-molecular-weight peptides which mimic these actions have now been purified by several groups, including ours (see accompanying paper), and higher-molecular-weight proteins with similar but less potent biological activities have also been identified and are presumed to be precursors. If released into the circulation, these peptides, collectively called atrial natriuretic factor (ANF), may play a significant part in blood-pressure homeostasis, regulation of extracellular fluid volume and as antagonists to the hypertensive effects of the renin-angiotensin system and other hormonal and neurotransmitter systems. We describe here the isolation and characterization of rat atrial cDNA clones which encode ANF. Nucleotide sequence analysis shows that auriculin corresponds to the 25 amino acids located close to the C-terminus of a 152-amino acid ANF precursor. Analysis of the in vitro translation products of precursor ANF mRNA suggests that multiple forms of the precursor may exist.  相似文献

Recent identification of natriuretic-diuretic activity in peptides isolated from human and rat atrial tissue implicates them in the control of extracellular fluid volume and electrolytic homeostasis. The presence of multiple forms of the peptides ranging from 3,000 to 13,000 molecular weight (MW) suggests they may all derive from the same precursor. The established amino acid sequence of alpha-human atrial natriuretic polypeptide (alpha- hANP ), a 28-residue peptide with potent natriuretic activity, provided the means to elucidate the structure of the precursor for alpha- hANP and the gene encoding it. Here we report the cloning and sequence analysis of the cDNA of human atrial mRNA encoding a precursor of alpha- hANP . The cDNA encodes gamma-human atrial natriuretic polypeptide (gamma- hANP ) of 13,000 MW, whose C-terminal 28 amino acid residues may be processed as alpha- hANP .  相似文献

The nucleotide sequence of a 1,091-base pair cloned cDNA insert encoding bovine corticotropin-beta-lipotropin precursor mRNA is reported. The corresponding amino acid sequence indicates that the precursor protein consists of repetitive units and includes a third melanotropin sequence in its cryptic portion. The 2595 bp cDNA, designatedMBLL (muscleblind-like), contains an open reading frame which encodes 255 amino acids and has 4 Cys3His motifs (GenBank Acc. AF061261). The amino acids sequence shares high homology toDrosophila’s mbl. The sequence of a cDNA encoding the nonapeptide arginine vasopressin (AVP) and its carrier protein, neurophysin II (NpII) from bovine hypothalamus, proves that the 166-amino acid precursor molecule contains a signal peptide of 19 amino acids followed directly by AVP connected to NpII by a Gly-Lys-Arg sequence. The carboxy-terminal region of the precursor contains a naturally occurring glycopolypeptide of 39 amino acids which is separated from NpII by a single arginine residue.  相似文献

The nonapeptide hormone oxytocin-like arginine-vasopressin (AVP) is synthesized as part of a larger precursor polypeptide. The precursor also includes the neurophysin molecule with which the hormone is associated in the neurosecretory granules of the hypothalamo-pituitary tract. A protein of molecular weight (Mr) approximately 20,000 has been isolated from supraoptic nuclei of rat hypothalami which, after tryptic cleavage, released a neurophysin-like molecule of Mr approximately 10,000 and an oligopeptide related to oxytocin. This result was complemented by in vitro translation of bovine hypothalamic mRNA. Among the primary translation products a single polypeptide of Mr approximately 16,500 was shown to contain antigenic determinants recognized by specific antisera against bovine neurophysin I and oxytocin. Here we report the amino acid sequence of the bovine oxytocin-neurophysin I (OT-NpI) precursor which was derived from sequence analysis of the cloned cDNA. As is the case for the bovine arginine-vasopressin-neurophysin II (AVP-NpII) precursor, the signal sequence of the OT-NpI precursor is immediately followed by the nonapeptide hormone which is connected to neurophysin I by a Gly-Lys-Arg sequence. A striking feature of the nucleic acid sequence is the 197-nucleotide long perfect homology with the AVP-NpII precursor mRNA sequence encoding the conserved middle part of neurophysins I and II.  相似文献

目的:克隆人转铁蛋白基因并对其编码序列进行分析.方法:以人胎肝cDNA为模板,利用PCR方法克隆人转铁蛋白基因;通过与基因组序列对比分析基因组结构;通过TargetP 1.1和SignalP 3.0预测信号肽;通过Clustal X(1.81)进行蛋白序列联配.结果:PCR扩增了一个长2 160 bp的基因片断,序列分析表明其覆盖了完整编码框,编码由698个氨基酸组成的人转铁蛋白.进一步分析发现人转铁蛋白基因有19个外显子和18个内含子,编码人转铁蛋白N端具有19个氨基酸组成的信号肽序列.人转铁蛋白与猩猩、猴子、兔子和老鼠的转铁蛋白氨基酸相似率分别为94%、91%、78%、73%.生物信息分析表明,人转铁蛋白含有高度保守的参与蛋白二硫键形成的半胱氨酸以及铁离子结合位点,有两个序列较同源的结构域.结论:成功克隆人转铁蛋白基因,人转铁蛋白与其它物种转铁蛋白同源.  相似文献

参考GenBank中多个BVDV毒株基因组序列,利用生物软件primer5.0设计两对引物,扩增出BVDV牦牛株 P20基因和P14基因.结果表明,克隆得到的P20基因序列为504 bp,与GenBank上发表的BVDV P20大小一致,P14基因序列为312 bp,与GenBank上发表的BVDV毒株比较有6个插入碱基.核苷酸序列的同源性及系统发生分析的结果表明,牦牛(Yak)株属于BVDV-1.  相似文献

根据牛钙蛋白酶抑制蛋白(Calpastatin, CAST) 设计并合成一对引物,从九龙牦牛肌肉组织提取总RNA,利用RT-PCR扩增获得九龙牦牛CAST基因部分片段,利用SQRT-PCR技术检测九龙牦牛各组织中CAST mRNA的表达差异.结果,成功克隆九龙牦牛CAST部分cDNA序列485bp,获得GenBank登录号为FJ483833;用DNAman软件分析发现九龙牦牛CAST基因与普通牛的核苷酸同源性依次为99%;SQRT-PCR组织表达检测表明,CAST基因在心脏、肝脏、脾脏、肺脏、肾脏、肌肉、脂肪中均表达,在心脏中表达最高,在脂肪组织中表达最低.  相似文献

根据猪MyoG基因的部分序列和绵羊MyoG mRNA设计引物,以成都麻羊、金堂黑山羊、白玉黑山羊和高原型藏山羊基因组为模板,应用PCR技术克隆测定MyoG基因序列.序列分析结果表明,成都麻羊MyoG基因长1957bp,由3个外显子和2个内含子组成,其中外显子Ⅰ、Ⅱ、Ⅲ的大小分别为398bp、82bp、122bp.内含子Ⅰ和内含子Ⅱ的大小分别为765bp和589bp.成都麻羊MyoG基因序列与金堂黑山羊、白玉黑山羊和高原型藏山羊的MyoG基因序列进行比对,同源性分别为99.9%、99.7%和99.3%,而它们氨基酸序列的同源性都为100%.这为进一步研究成都麻羊MyoG基因的表达、生物学活性和应用奠定了一定基础.  相似文献
